It has been reported that Aloe vera contains vitamin, enzyme, mineral, sugar, phenolic compound, fatty acid and amino acid. Aloe vera changes its color from green to red under the extreme thermal and arid climate to protect itself. These morphological changes induce variation of composition such as increasing of aloe-emodin content. Aloe-emodin is one of the major anthraquinone in aloe family plants. Since aloe-emodin contains a polyphenolic structure, this compound may be responsible for the reported antioxidant and anti-inflammatory effects of aloe. However, there is no research on the process of increasing the compounds of Aloe vera. Therefore, the purpose of this study is to develop a pink aloe manufacturing process that increases the aloe-emodin content and enhances the antioxidant and anti-inflammatory activities of aloe. As a result of heating aloe under appropriate conditions, pink aloe increased aloe-emodin content compared to general aloe, and exhibited effects such as increasing antioxidant activity, inhibiting NO production, and promoting cell regeneration. Lee","doi":"10.15230/SCSK.2020.46.3.207","DOIUrl":"https://doi.org/10.15230/SCSK.2020.46.3.207","url":null,"abstract":"In this study, the anti-inflammatory and anti-bacterial activities of the extracts from the leaves of the Hydrangea petiolaris were identified, and the chemical structure was identified by separating the active ingredient. As the result of the anti-inflammatory activity experiment using RAW 264.7 cells, it was confirmed that the n-hexane (Hex) and ethyl acetate (EtOAc) fractions inhibited the production of nitric oxide (NO) and the expression of iNOS protein in a concentration-dependent manner without cytotoxicity. In addition, the n-Hex and EtOAc fractions reduced the production of pro-inflammatory cytokines (TNF-α, IL-1β and IL-6). Upon the anti-bacterial tests using Staphylococcus epidermidis and Cutibacterium acnes, the extract, n-Hex, EtOAc and n-butanol (BuOH) fractions showed potent activities. In order to isolate the active constituents, the n-Hex and EtOAc fractions were further purified to afford four phytochemicals; phytol (1), corosolic acid (2), asiatic acid (3) and 1-O-p-coumaroyl-β-D-glucopyranoside (4). All of the compounds 1 – 4 were isolated for the first time from this plant. In addition, the contents of isolated compounds were determined by HPLC and the quantity of phytol (1) was 27.8 mg/g for the 70% EtOH extract. As a result of confirming the cytotoxicity and collagen synthesis ability according to the concentration of the polar low molecular weight keratin peptide in human fibroblasts, it was confirmed that the cytotoxicity did not appear and the collagen synthesis in human fibroblasts was increased. A mask pack containing a polar low-molecular weight keratin peptide was used, and a test product was used for 4 weeks in 22 healthy women subjects. As a result, it showed statistically significant effects on skin elasticity, skin torsion elasticity, skin color and moisture improvement. After extraction by stirring, ultrasound and microwave method using 70% ethanol and distilled water as solvents, the results of considering the content of ginsenoside Re and Rb1, total polyphenol content, antioxidant activity, and whether it is an environmentally friendly manufacturing method, it was confirmed that the microwave method using distilled water is good method of extraction. The optimization of extraction conditions for microwave method were made by response surface methodology (RSM). Microwave power (50 ∼ 200 W, X1), solvent and ginseng berry ratio (5 ∼ 20 times, X2) and the extraction time (30 ∼ 120 s, X3) were used as independent variables. The model showed a good fit having a determination coefficient of the regression equation of 0.9 or more and a p-value less than 0.05. Estimated conditions for the maximized extraction of ginsenoside contents and total polyphenols were 200 w in microwave power, 20 times in solvent and ginseng berry ratio, and 90 s in extraction time. Predicted values at the optimum conditions were total polyphenols of 6.23 mg GAE/g, ginsenoside Re of 17.69 mg/g, and ginsenoside Rb1 of 16.01 mg/g.
