As the result of the anti-inflammatory activity experiment using RAW 264.7 cells, it was confirmed that the n-hexane (Hex) and ethyl acetate (EtOAc) fractions inhibited the production of nitric oxide (NO) and the expression of iNOS protein in a concentration-dependent manner without cytotoxicity. In addition, the n-Hex and EtOAc fractions reduced the production of pro-inflammatory cytokines (TNF-α, IL-1β and IL-6). Upon the anti-bacterial tests using Staphylococcus epidermidis and Cutibacterium acnes, the extract, n-Hex, EtOAc and n-butanol (BuOH) fractions showed potent activities. In order to isolate the active constituents, the n-Hex and EtOAc fractions were further purified to afford four phytochemicals; phytol (1), corosolic acid (2), asiatic acid (3) and 1-O-p-coumaroyl-β-D-glucopyranoside (4). All of the compounds 1 – 4 were isolated for the first time from this plant. In addition, the contents of isolated compounds were determined by HPLC and the quantity of phytol (1) was 27.8 mg/g for the 70% EtOH extract. 本研究鉴定了小绣球叶提取物的抗炎和抗菌活性,并通过分离活性成分对其化学结构进行了鉴定。通过对RAW 264.7细胞的抗炎活性实验,证实了正己烷(Hex)和乙酸乙酯(EtOAc)组分抑制一氧化氮(NO)的产生和iNOS蛋白的表达呈浓度依赖性,无细胞毒性。此外,n-Hex和EtOAc组分减少了促炎细胞因子(TNF-α, IL-1β和IL-6)的产生。经表皮葡萄球菌和痤疮角质杆菌的抑菌试验,提取物、正己烯、乙酸乙酯和正丁醇(BuOH)组分显示出较强的抑菌活性。为了分离活性成分,进一步纯化了n-Hex和EtOAc组分,得到了4种植物化学物质;其中叶绿醇(1)、花冠果酸(2)、亚洲果酸(3)和1- o -对香豆醇-β- d -葡萄糖吡喃苷(4)。化合物1 ~ 4均为首次从该植物中分离得到。HPLC法测定分离化合物的含量,70% EtOH提取物中叶绿醇(1)含量为27.8 mg/g。基于以上研究结果,认为利用小叶红叶提取物开发具有抗炎抗菌作用的天然化妆品材料是可能的。
