The various colors are determined by the accumulation of molecules such as chlorophyll, carotenoids (including lycopene) and flavonoids. Since the DR5 promoter is induced by auxin, we can infer that the auxin responses differ at different developmental stages in tomato. Fruit-specific promoters can effectively drive gene expression in fruits, and the E8 promoter is one of the most widely used fruit-specific promoters. In this study, DR5:: Ruby transgenic tomato plants were used to further clarify the auxin content at different developmental stages in tomato. The results showed that the nonuniformity of Ruby activity was consistent with the natural distribution characteristics of native auxin, and the results were also consistent with the qRT ‒ PCR results. Furthermore, the accumulation of endogenous auxin at the immature green stage (IMG) was higher than that at the other stages. In addition, E8:: Ruby was transformed into different tomato varieties (red, white and yellow) to generate new tomato varieties with fruit of different colors. The results showed that the E8:: Ruby-transformed plants exhibited asymmetrical pink coloration in different tomato tissues of plants from different tomato cultivars. In conclusion, all the results showed that DR5 and E8 are suitable systems for studying the application of the Ruby reporter gene in tomato. It is cultivated for its fresh fruits and processed paste, with over 153 million metric tons of global production. However, modern tomato cultivars have limited sugars, acids, and volatiles allelic diversity as flavor has generally been less prioritized in breeding programs. Invertase is an essential regulator of flavor and sugar metabolism in tomato. Genetic control of tomato flavor is still incomplete without a clear understanding of the roles of invertase and sucrose metabolism. This review provides an overview of our current understanding of the invertase mode of action in sucrose metabolism, their evolutionary and functional divergence in the tomato genome, role in stress response, genetic and hormonal control of fruit flavor and quality. We summarized the primary roles of invertase in sugar metabolism and fruit flavor. Citation: Ahiakpa JK, Karikari B, Magdy M, Munir S, Mumtaz MA, et al. 2021. In this study distribution and characteristics of SSR loci in the genome of Chinese cabbage 'Chiifu' was analyzed. Finally, a total of 173,892 SSR markers that occurred in 10 chromosomes were identified, and Perfect Repeat Motifs (P type) which is a main form of SSR loci with a mean distance of 1.89 kb/SSR was identified. Among them, mono-, di-, and tri-nucleotide repeat motifs were equally distributed across chromosomes compared with tetra-, penta-, and hexa-nucleotide repeat motifs. The largest proportion of SSR loci consisted of mono- and dinucleotide repeat motifs, which accounted for 93.04% of the total SSR loci. A/T and AT/TA were the most abundant motifs, accounting for 97.61% and 64.05% of the mono- and dinucleotide repeats, respectively. Furthermore, 200 SSR markers were used for the amplification of 12 Chinese cabbage germplasm, in which 30 were highly polymorphic. These SSR markers were further used for the association analyses of flowering time of 190 Chinese cabbage accessions. The results revealed that 9 and 10 alleles were associated with bolting and flowering time, respectivley. The results show that development of SSR markers is feasible and useful in marker assisted selection of Chinese cabbage.","PeriodicalId":160756,"journal":{"name":"Vegetable Research","volume":"1 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"127480505","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Although the unicellular trichomes in Arabidopsis have been well studied, little is known of the development of multicellular trichomes in tomato. With seven morphologically and functionally different types, tomato trichomes are more complex than the unicellular trichomes in Arabidopsis. Here, we comprehensively analyzed the spatiotemporal pattern of tomato trichome development. Our observations show that the gradual reduction of type I/IV trichomes during leaf development is accompanied by an increase of other trichome types. We further examined the gland development of type I/IV trichomes by constructing a marker reporter, pSlAT2: GUS-YFP which is specifically expressed in the top cell of the trichomes. Interestingly, we found the multicellular base of trichomes is initiated as early as the two-cell stage. Through tracking a nuclear marker, H2B-GFP in tomato trichomes, we found the correlation between stalk cell length and nuclear size, suggesting that cell ploidy level could affect trichome cell size. Finally, we found type VII glandular trichomes exhibit two morphologically distinct forms, with the majority covered by a wax-like substance, which could help to form extra storage space for secretory metabolites. Our results provide the basis for further mechanistic study of trichome development in tomato.","PeriodicalId":160756,"journal":{"name":"Vegetable Research","volume":"16 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"1900-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"130219646","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
