Bai, Li Jiale","doi":"10.3724/SP.J.1231.2013.38454","DOIUrl":"https://doi.org/10.3724/SP.J.1231.2013.38454","url":null,"abstract":"In order to study genes related to pearl formation in freshwater pearl mussel and their regulation mechanism,a 1 437 bp cDNA sequence of calreticulin gene from Hyriopsis cumingii(HcCRT)was obtained by rapid amplification of cDNA ends(RACE).It consisted of a 231 bp 5′-untranslation region(UTR),a 615 bp 3′UTR and a 591 bp open reading frame(ORF).The inferred amino acids sequence was composed of 196 amino acids,including a signal peptide of 21 amino acids and a mature peptide of 175 amino acids.The molecular weight of the peptide was predicted to be 22.4 ku,with a theoretical isoelectric point of 5.01.Amino acid sequence analysis showed that there was no obvious amino acid sequence of membrane domain.Results of the ProtScale online analysis showed that the protein was hydrophilic protein.Homology analysis indicated that HcCRT amino acid had a conservative sequence of calreticulin family and was highly conserved with Danio rerio(77%),Crassostrea gigas(70%),Pinctada fucata(70%).The sequence analysis showed that HcCRT shared two potential calreticulin family signature motifs with CRT from other species,KHEQNIDCGGGYLKVF and IMFGPDICG.The prediction results of H.cumingii HcCRT protein secondary structure and tertiary structure indicated that the protein contains the alpha helix and beta folding.Real-time quantitative PCR(RT-PCR)showed that HcCRT is expressed in a wide range of tissues including the mantle,blood,gill,foot,liver,kidney,intestine and muscle,with the highest level of transcripts in mantle,followed by blood,while there is rare expression in other tissues.These data suggested that HcCRT might be involved in pearl formation of H.cumingii.This study which was first time to obtain HcCRT gene from H.cumingii played an important role in exploring pearl formation of H.cumingii.","PeriodicalId":15710,"journal":{"name":"Journal of Fisheries of China","volume":"37 1","pages":"679"},"PeriodicalIF":0.0,"publicationDate":"2013-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"69967461","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Wang, Xin-cang Li, Shuai Zhou","doi":"10.3724/SP.J.1231.2013.38500","DOIUrl":"https://doi.org/10.3724/SP.J.1231.2013.38500","url":null,"abstract":"In order to evaluate the hazard of chlorpyrifos to the cultured shrimp Litopenaeus vannamei,the lethal effect of the shrimp infected with white spot syndrome virus(WSSV)under the chlorpyrifos stress was tested,while the quantity of WSSV in the gill tissue and acetylcholinesterase acticity in the muscle tissue were analyzed.The 96 h median lethal concentration(LC50)of chlorpyrifos to the shrimp was determined.The results showed that the 96 h LC50 is 0.758 μg/L(0.521-0.987 μg/L),and as time prolonged,the values of LC50 significantly declined,which showed a positive concentration-response relationship.On this basis,the stress concentration of chlorpyrifos of 0.2 μg/L was determined.After the shrimp were exposed to 0.2 μg/L chlorpyrifos for 4 days,they were infected with WSSV by injection.It showed that the cumulate mortality of chlorpyrifos-WSSV treatment(83.33%±4.7%)was significantly higher than that of ethanol-WSSV group(40.00%±0.9%).Meanwhile,the amount of WSSV was also determined by Real-time Relative PCR at 48,72 and 96 h post-injection.WSSV quantity of chlorpyrifos-WSSV treatment was about 4 times that of ethanol-WSSV control at 72 h.WSSV quantity of chlorpyrifos-WSSV treatment was significantly increased at 96 h,which was 4.9 times that of chlorpyrifos-WSSV treatment at 72 h,and 5.9 times that of ethanol-WSSV treatment at 96 h.The acetylcholinesterase(AchE)activity in shrimp muscle tissue under chlorpyrifos stress was 20% lower than that in control group.This showed that the proliferation rate of WSSV in shrimp increased rapidly under chlorpyrifos stress,which raised the mortality rate of the shrimp.","PeriodicalId":15710,"journal":{"name":"Journal of Fisheries of China","volume":"37 1","pages":"1106"},"PeriodicalIF":0.0,"publicationDate":"2013-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"69968046","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Zhigang, Li Qingyong, Li Yujiao","doi":"10.3724/SP.J.1231.2013.38611","DOIUrl":"https://doi.org/10.3724/SP.J.1231.2013.38611","url":null,"abstract":"In this study,using the methylation sensitive amplified polymorphism(MSAP) method,we globally assessed the effect of low temperature adaptation on the level and variations of cytosine methylation alterations at CCGG sites of genomic DNA in Nile tilapia(Oreochromis niloticus) strains with good coldresistance characters developed by the successive and directional selection breeding of multi-generations. Total of 849 fragments were identified by 18 pairs of se1ective primer combinations of which 411 and 438 were detected in cold-resistance tilapia strain and the control,respectively. Among these fragments,the methylation sites were 72 and 104,and the corresponding total-methylation levels w ere at 17.52% and 23.74%,respectively. The full methylation sites at the internal Cs were 37 and 65,and the corresponding full methylation levels were at 9. 00% and 14. 84%,respectively,and the hemi-methylation sites at the external Cs were 35 and 39,and the corresponding hemi-methylation levels were at 8.52% and 8.90% in coldresistance tilapia strain and the control,respectively. Further analysis of DNA methylation indicated that a statistically decrease in the overall level of total-methylation levels and both methylation types was detected in cold-resistance tilapia strain compared with the control(decrease at 6.22%,5.84% and 0.38%, respectively),and the changes of DNA methylation pattern were recognized mainly in the full methylation sites. The level of decrease in DNA methylation suggested that the levels of genomic DNA methylation were changed during the successive cold stress of multi-generations in tilapia,and it also indicated that DNA methylation alteration in tilapia treated with cold stress was mainly through de-methylation that occurred in some CCGG sites. All these results implied that the change of DNA methylation was closely associated with cold tolerance of tilapia. The results showed that the fatty acid composition of these ten P. yezonsis samples was not affected; however,the content of each fatty acid was affected and the difference among ten samples was significant. The saturated fatty acids(SFAs) w ere mainly composed of 13:0,16∶0,18∶0 with 16∶0 as the highest component(about 20%). The unsaturated fatty acids(UFAs) included 18∶1n-9,18∶2n-6,18∶3n-3,20∶1n-9,20∶2n-7,20∶3n-7,20∶4n-6( AA),20∶5n-3(EPA) and the major componet w as EPA(about 50%). Samples harvested during the low est w ater temperature period(February) contained the highest total fatty acids and EPA concentration; the result indicates that the low water temperature has advantages in accumulating fatty acids especially UFAs of P. yezoenis. P. yezoensis is rich in UFAs(especially EPA). Using RT-PCR and Quantitative real-time PCR analyzed dsx expression in different reproductive modes' daphnias. In addition,w e synthesized DIG labeled RNA probe of dsx by in vitro transcription and used w hole mount in situ hybridization researching dsx location expression in different reproductive modes' daphnia. Results show ed that dsx in male and female had no gender specificity of alternative splicing,only a same transcription. Realtime PCR show ed dsx highest expression level w as in male,w hich w as parthenogenetic female's 2. 6 times and 4. 9 times that of the female. They had significant difference( P 0. 05) and gender differences in expression each other. Whole mount in situ hybridization found that dsx gene w as expressed in the somatic tissure in different from reproductive modes ' daphnia,but the difference in quantity and parts. The expression also show ed the gender differences in male and female. In male the signals w ere revealed in the first antenna,the first thoracic limb and the compound eye. The parthenogenetic female and sexual female corresponding sites' expressions w ere relatively w eak. Jian","doi":"10.3724/SP.J.1231.2013.38138","DOIUrl":"https://doi.org/10.3724/SP.J.1231.2013.38138","url":null,"abstract":"To confirm whether there is anadromous estuarine tapertail anchovy Coilia nasus now in Poyang Lake,an electron probe microanalyzer(EPMA)was applied to analyze the element Sr and Ca microchemistry patterns in otoliths of two individuals collected from Xingzi area of the lake.Results of line transect analysis showed that the ratios of Sr∶ Ca×1 000 in the otolith of the individual with a shorter supramaxilla(PYCB02)were consistently low(1.66±0.54),indicating that it experienced in freshwater habitat during its whole life history.Another individual with a longer supramaxilla(PYCE05)had a complex Sr/Ca ratio pattern which can be divided into three phases(1.82±0.50,5.79±0.91,and 4.14±0.88),suggesting it experienced not only the freshwater,but also the seawater and brackish water during different stages in its life history.All these differences could be well confirmed by the results of X-ray mapping analysis of EPMA.The whole otolith of PYCB02 presented a uniform bluish pattern(low Sr level)from the core to the outermost regions.In contrast,the otolith of PYCE05 showed a multi-color pattern.There were greenish(moderate Sr level),yellowish(moderate Sr level),or even reddish(high Sr level)in outer regions adjacent to the bluish central regions.Results of the two different microchemistry patterns obtained by both the line transect and mapping analyses confirmed objectively,for the first time,the coexistence of resident and anadromous forms of Coilia nasus in Poyang Lake and suggested that Coilia nasus is still possible to migrate anadromously for spawning into Poyang Lake,which is about 800 km away from the river mouth of Yangtze River.These aforementioned microchemistry patterns will provide important information not only to more effectively evaluate and protect the resources of Coilia nasus,but also to rationally plan and guide future ecological economic development in Poyang Lake.","PeriodicalId":15710,"journal":{"name":"Journal of Fisheries of China","volume":"37 1","pages":"239-244"},"PeriodicalIF":0.0,"publicationDate":"2013-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"69963009","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
