The conception per say happens through the contributions by both the male and the female counter parts and is equally influenced by them. It is important to evaluate both the partners for a healthy conception. Ayurveda explains four important factors which are required for the manifestation of the garbha or for the conception to take place; they are rutu, kshetra, ambu and beeja. Alteration in any of the four factors might cause a hindrance for conception. Hence, evaluating these factors in both male and female counter parts as applicable is of utmost importance prior to conception. A male patient aged 34 years consulted us for pre conception care with a trying period of 6 months and marital life of 6 months. He was advised to undergo semen analysis which revealed presence of a large number of leukocytes in the semen sample and hence was diagnosed as pyospermic/leukocytospermic, which can be understood as pooti puya shukra dushti in Ayurveda, which can further be classified as a pittaja shukra dushti in a broader sense. He was treated with shodhana procedures of virechana and nasya to correct the beeja dushti followed by vajikarana chikitsa to enhance the health of beeja. Post treatment pus cells in semen reduced completely. Currently, it poses a major risk to the global health. There are numerous risk factors associated with cancer. They include tobacco, obesity, excessive consumption of packed foods, lack of physical activity, alcohol consumption, etc. Continued prevention, research & early detection are required to improve outcomes for cancer patients. Anticancer medications have been associated with cardiotoxicity, compromising individuals' heart function. Early detection of cardiotoxicity is essential for avoiding irreparable heart damage. Cardio-oncology has arisen as a distinct area of study and therapeutic practice in recent years. Several aspects of drug-induced cardiotoxicity remain unknown. Such toxicities are considered as garavisha in Ayurveda. They can be treated by using Agada principles. Insect bites can lead to cardiac symptoms, according to both ancient and modern science. Acharya Vagbhata has mentioned hrudpeeda as one of the lakshana of Vataja Vrischika damsha. Dashanga Agada (DA) is the formulation mentioned in context of Keeta visha. As it is the formulation of choice in insect bites exhibiting cardiac symptoms, it is studied in present research to reduce the cardiotoxicity induced by doxorubicin (Dox). The study was carried out on H9C2 mice cardiomyoblasts. Materials and Methods: Hydroalcoholic extract of Ayurvedic formulation Dashanga agada was prepared. H9C2 mice cardiomyoblasts were used to study trial drug Dashanga Agada on cardiotoxicity induced by a chemotherapy drug named Doxorubicin. MTT assay, DAPI nuclear staining, DCFH-DA assay for intracellular ROS generation & SOD assay were the four parameters studied on H9C2 cell line. 4 groups were taken for the study. Dashanga agada was studied at different concentrations & different time intervals on the above mentioned parameters. Results: IC50 values of DA & Dox for 24hrs were calculated to be 357.15 & 5.48 µg/ml respectively. Dashanga agada was effective in increasing cell viability, improving the nuclear morphology & antioxidant activity when compared to Doxorubicin treated group. Conclusion: Dashanga Agada has cardioprotective effect. HPLC method was developed by C-18 column with the mobile phase comprising of 0.4% orthophosphoric acid [OPA]: Acetonitrile [ACN] (45:55, v/v), isocratically pumped with a rate of flow 1 mL/min and 255 nm detection wavelength. The method validation of HPLC parameters and degradation tests was analysed by International Conference on Harmonization (ICH) guidelines. The developed HPLC method was found to be specific, linear (r2>0.999) over the selected range of concentration 2-14μg/mL, precise, with the limit of detection (LOD) and limit of quantification (LOQ) as 0.021 and 0.064μg/mL respectively. The entrapment efficiency of quercetin for prepared Nanostructure Liquid Crystalline Dispersion (NLCD) was observed to be 94.55 %. The developed HPLC method was applied for the quantification of quercetin in NLCD. The quercetin peak was completely resolved without any disturbances from degrading products and additives. Based on theoretical reference, FGD and IDI conducted among experts 35 items are generated in two domains (Physical and Psychological). From this 29 items are selected for final administration on selected sample for validity and reliability assessment. Each item is scored using 4 point Likert scale -no, rarely, often, every time with scoring 0, 1, 2, 3 respectively. Subjective question assessed by interrogation and objective parameter dryness of body parts are also assessed by this scoring method. Total 100 samples are selected for final administration of sample. The tool with 29 items gave a Cronbach’s Alpha 0.745 (n=29) showing high construct validity and internal consistency. After the reliability and validity assessment 1 items  deleted from the  administered tool in the selected sample. In Ayurveda, a similar condition called Madhumeha is attributed to Dhatu kshaya (Emaciation) and Dosha avarana (occlusion of humoral factors). Rasayana therapy, involving herbal formulations like Nishamalaki, is recommended to strengthen the body's major structural components (dhatus), boost immunity, and promote longevity. Nishamalaki, a combination of Haridra (Curcuma longa Linn.) and Amalaki (Phyllantus emblica Linn.), exhibits antioxidant, anti-inflammatory, and lipid-reducing properties. It holds potential as a preventive measure to enhance the Quality of Life. The aim of this study was to validate the rejuvenation property of Nishamalaki in Diabetes mellitus using an insilico approach. Through tools such as SwissADME, SwissTargetPrediction, and Therapeutic Target Database (TTD), bioavailable components of Nishamalaki and Diabetes were analyzed, and a network was created using CytoScape. The results of the study identified two key compounds, Ellagic acid and Corilagin, which significantly modulate IGF1R and PTPN1, respectively, in Diabetes mellitus. Diabetes is increasing day by day because of lifestyle changes. As per the WHO statistics around 2 million deaths occur each year because of diabetes. Hence there is a serious concern to find an effective and safe medicine for the treatment of diabetes. Many herbal drugs have been researched and found to be effective in controlling the diabetes. One of such herbs is Gymnema sylvestre (Retz.) R.Br. ex Sm. (GS). It is equated with Meshashrungi, mentioned in the Ayurvedic texts. Ajashrungi (Pergularia daemia (Forssk.) Chiov.) (PD) is also taken as a substitute for Meshashrungi. So, to find out the efficacy of both the drugs in the management of diabetes, the present work is taken up. It was a Prospective, Randomised, Single blind, Comparative Clinical Study. A total of 40 subjects were randomised into two groups with 20 subjects each. Group A were administered Meshashrungi patra vati and Group B were administered Ajashrungi patra vati for a period of 60 days with a follow up for every 15 days. Assessment was done based on changes seen in subjective and objective Parameters. Significant changes were observed in both the groups before and after the treatment, but when compared between the groups there was no significant difference. This indicates that both the drugs are equally efficacious in the management of diabetes mellitus. On August 5, 2022, a 48-year-old female patient attended the OPD of Santhigiri Siddha Medical College and Hospital, Trivandrum, with the following complaints: dilatation of veins, present in the right lower limb for 6 years; hyperpigmented lesion with itching and swelling above the medial malleolus of the right lower limb for 6 months; and an ulcer present above the medial malleolus for the last 3 months. She had a known case of varicose veins and had taken sclerotherapy three times but did not have satisfaction with the allopathic medication. The majority of the symptoms were significantly reduced in 60 days under the Siddha medications, such as a combination of Siddha preparation (Parangipattai Chooranam, Sangu Parpam, and Silasathu Parpam), Rasa gandhi mezhugu for internal, Padikara neer washing, wound dressing with Mathan thailam for external for 2 months, and attai vidal (leech application) done in 3 sittings in the gap of 10 days. No recurrence of any chronic varicose ulcer was observed during the 1 year of follow-up, and relevant preventive Siddha medication for Naalavibatha punn was given. The doctor advised the patient to do exercises that strengthen the legs and compression therapy. This study aims to identify potential inhibitors against the TGF-β-type-I receptor (PDB Id-1VJY) by developing ligands through molecular docking and ADMET-based virtual screening. The plant-based nature product database of Morus alba Linn  is utilized for this purpose. The resultants hits, identified as actives were evaluated by molecular docking studies to get insight into their potential binding interaction with the target protein. 1-deoxynojirimycin (A1), Catechin (A2), Cyanidine-3-rutinoside (A3), Cyclomulberrin (A4), Kaempferol (A5), Kuwanon-G (A6), Morusin (A7), Mulberrin (A8), Mulberrofuran-G (A9), Quercetin-3-(6-malonylglucoside) (A10), Quercitrin (A11), Rutin (A12), were selected for the molecular docking.ADMET based virtual screening ligand/Compound (A1, A2, A4, A5, A7, A8, A9) passes the lipinski’s rule. Compound A4 (-10.7 kcal/mol), A5 (-10.4 kcal/mol) and A8 (-9.9 kcal/mol) had the highest binding affinity to the active site in TGF-β-type-I receptor. Body mass index (BMI) values within 25 and 29.9 kg/m2 and below 30 kg/m2 are two measures of the positive energy imbalance that leads to obesity. In 2000, World Health Organization identified Obesity as the most clearly evident yet most ignored public health problem worldwide. The World Health Organization ranks obesity as the second most dangerous disease in the world. The worldwide impact of Obesity is recorded to be 4 million obesity-related deaths and 40 million disability-adjusted life years among adults globally in 2015. Family history, frequent overeating patterns, and lack of physical activity are all significant risk factors. When treating Sthula (obese patients), Ruksha Udvartana is mentioned, which involves using a dry powder of herbs without oil is used. Therefore, for the current study, it was decided to choose the Rodhradigana Udvartana followed by Bhaspaswedana and Gomaya Mashi Udvartana followed by Bhaspaswedana as a comparative study. The purpose of the current study is to document and examine this process for statistical interpretation. Aim: Evaluation of Comparative efficacy of Rodhradi Gana Udvartana with Bhaspaswedana and Gomaya Mashi Udvartana with Bhaspawedana in the management of Sthoulya (obesity). Material and Methods: Data related to the disease as well as the drug was collected from Samhitas, other classical Ayurvedic literature, and various modern science books. Subjects visited to outdoor and indoor Panchakarma department and from the specialty camps were registered for the present study. Result: Significant improvement was observed in both groups and during comparison, more results were found in the Gomaya Mashi group (Group B). For the global reorganisation of Ayurvedic drugs advanced standardisation techniques may be involved to maintain quality assurance. Madhumalini Vasant Ras (MVR) is a well-known Ayurvedic herbo-mineral formulation used in the treatment of IUGR (Intra Uterine Growth Retardation). This formulation is not mentioned in the Ayurveda Formulary of India and there is no data on the quality standards of MVR available. So in this study, preliminary data on physicochemical analysis along with HPTLC evaluation of Madhumalini vasant Ras is generated. Aim and objective: To evaluate and generate data on the physicochemical parameters of the MVR tablet. Materials and Method: The raw drugs were authenticated by experts of Dravya Guna and Rassastra Bhaishajya Kalpana departments and tablets (125 mg) were prepared at the pharmacy, Parul institute of Ayurveda, Vadodara. Then the physico-chemical parameters and microbial limits were checked and HPTLC analysis was done. Results: While evaluating the physicochemical parameters, the pH value was 4.19, LOD 6.01%, Total ash value 11.54%, Acid insoluble ash 8.70%, water-soluble extractive value was 22.20% w/w, alcohol soluble extractive value was 13.40% w/w, HPTLC at 254 nm and at 366 nm showed five common spots. The Microbiological analysis shows that the formulation is free from microbes. Conclusion: Preliminary data on organoleptic characters, antimicrobial evaluation, various physicochemical parameters and HPTLC of MVR tablet is generated.
