This study investigates naringenin, a flavonoid with potential antioxidant and anti-inflammatory properties, as a novel therapeutic candidate targeting HO-1 upregulation to mitigate retinal damage. In this regard, a rat model of streptozocin-induced diabetic retinopathy was established and naringenin was administrated (40 mg/Kg/day and 80 mg/Kg/day). Two other untreated groups of diabetic and non-diabetic rats were used to compare with naringenin-treated groups. The findings revealed that naringenin decreased the overexpression of the pro-inflammatory factors IL-1β, IL-6, and TNF-α in the retinal tissue. Moreover, naringenin inhibited the overexpression of TLR4, NF-κB, and CASP-3, caused heme oxygenase-1 (HO-1) overexpression, upregulated BCL-2, reduced the levels of malondialdehyde, and elevated the levels of superoxide dismutase, catalase, and glutathione peroxidase (P-value<0.05). Intraperitoneal injection of the HO-1 inhibitor zinc protoporphyrin (ZnPP) blocked the protective effect of naringenin. The emerging role of pyroptosis, a novel form of regulated cell death, in the pathogenesis of podocyte damage in MN underscores the need to identify additional regulators and inhibitors of pyroptosis to optimize MN treatment. This study examined osteomodulin (OMD) as a potential regulator of pyroptosis in MN using a passive Heymann nephritis (PHN) rat model. OMD silencing in PHN rats resulted in an alleviation of nephrotic syndrome, as evidenced by reductions in 24-h urine protein and low-density lipoprotein levels, and increased albumin levels. Additionally, OMD silencing mitigated podocyte injury, as indicated by the restoration of podocyte foot process architecture, enhanced expression and normalization of Nephrin and Podocin, and reduced Desmin expression in glomeruli. Mechanistically, OMD silencing led to the inhibition of renal p38 MAPK activation and pyroptosis, demonstrated by decreased expression of intrarenal p-p38 MAPK (Thr180/Tyr182) and key pyroptosis markers, including NLRP3, ASC, Caspase-1, Caspase-1 p20, IL-1β, IL-18, GSDMD, and GSDMD-N, as well as lowered serum levels of IL-1β and IL-18. To investigate the possible role of calcineurin in taurine upregulating CYP7A1 expression in HepG2 cells under high cholesterol conditions. High cellular cholesterol conditions were achieved using 0.2 mM cholesterol in HepG2 cells. Calcineurin was decreased by FK506 and was depleted in CnAβ-/- cells. HepG2 cells were cultured in a taurine-containing medium for 24 or 48 h. The levels of total intracellular cholesterol were determined by an enzymatic method, and the expression levels of calcineurin, CYP7A1, and key transcriptional regulatory molecules were detected by western blotting. High-cholesterol resulted in increased CYP7A1 and calcineurin expressions. Taurine exhibited cholesterol-lowering effects regardless of low/high cholesterol conditions or calcineurin status. Taurine induced the expression of CYP7A1, which was abolished by inhibiting or deleting calcineurin. Taurine suppressed MEK1/2, p-c-Jun, and SHP-1, key molecules in one inhibitory pathway of CYP7A1 transcription. In contrast, suppression of MEK1/2 but not p-c-Jun or SHP-1 was reversed after completely knocking down calcineurin. Vascular endothelial growth factor (VEGF) and Notch ligand Delta-like 4 (Dll4) play a critical role in vessel formation and angiogenesis. This study aimed to determine anti-angiogenesis effects of Sini Decoction serum on human umbilical vein endothelial cells (HUVECs). We prepared Sini Decoction serum in doses of 0.925, 1.85, 3.7 g/mL, and administered to HUVECs. HUVECs migration was evaluated using a wound healing assay. Formation of tube-like structures in HUVECs was observed using a tube-formation assay. Gene transcriptions of Dll4, Notch1, VEGF receptor 2 (VEGFR-2) and VEGF were examined using a PCR assay. Protein expressions of Janus kinase-2/signal transducer and activator of transcription 3 (JAK2/STAT3) molecules and signal molecules downstream of VEGF/VEGFR related to angiogenesis (including mitogen-activated protein kinase (p38MAPK), extracellular signal-regulated kinase-1/2 (ERK1/2), focal adhesion kinase (FAK) and phosphorylated protein kinase B (p-Akt)) were detected by western blot. Sini Decoction serum significantly inhibited cell migration and the formation of tube-shaped structures in HUVECs compared to Control group (P<0.05). Sini Decoction serum markedly decreased Dll4, Notch1, VEGF, and increased VEGFR-2 gene transcriptions, compared to those of Control group (P<0.05). Sini Decoction serum significantly reduced expression of p-Akt, p38MAPK and ERK1/2, compared to Control group (P<0.05). Although a reduction in FAK and STAT3 expression was discovered in HUVECs administered by Sini Decoction serum, without statistical significance compared to Control group (P>0.05). Its combination with Shennao fuyuan decoction (SFD), a traditional Chinese medicine formulation for ischemic stroke treatment, additionally augments neural deficit repair in rats post-cerebral ischemia. Nonetheless, there is a notable need for more comprehensive research into the exact mechanisms by which this combined treatment ameliorates brain injuries. Consequently, we conducted thorough research on the mechanisms of action of these entities, utilizing network pharmacology approaches and a rat model of middle cerebral artery occlusion (MCAO). Our findings identified the hsa05417: Lipid and atherosclerosis pathway as the primary channel through which SFD impacts the cardiovascular system. The integrated treatment of SFD and hUCMSC-exo attenuated TNF and HSP90AA1 expression and elevated IL6 and GSK3B levels in the brains of rats subjected to MCAO. Moreover, the discrete utilization of hUCMSC-exo in conjunction with SFD reduced cerebral infarction and diminished tissue damage in rats with MCAO.
