C. botulinum is capable of forming spores at low concentration; thus, regular detection methods must begin with an enrichment step before the main procedure. This additional step requires long time, complicated preparation, and might fail due to improper selective media. Therefore, this study aimed to establish a rapid detection procedure for C. botulinum serotype B directly from honey without enrichment culture by obtaining total DNA to use as template for the amplification of a fragment from botulinum neurotoxin type B (BoNT/B) encoding gene. Using 6 honey samples provided by the National Institute of Hygiene and Epidemiology with positive PCR results after enrichment, all three DNA extraction methods including Exgene extraction kit, freeze-thaw cycling and bead beating, showed amplification signals. Furthermore, when combined with the isothermal loop-mediated amplification, the total time could be shortened to less than two hours since collection. In addition, we observed PCR inhibition by the honey matrix and recommended dilution of the sample 5-10 times for the reaction. Methodological approaches to detecting and identifying chemical substances inadvertently present in foodstuffs were proposed In the Russian Federation. The developed methodological approaches include four stages: analytical identification of chemical substances; integrated assessment of chemical hazards employing additional selection criteria followed by the application of a score and summation of points; categorizing chemical substances with the assignment of potential hazard categories and final stage - health risk assessment for selected chemicals based on the integrated index. The presented methodological approaches were tested on the example of canned meat for infant nutrition consumed in the Russian Federation and the Socialist Republic of Vietnam. Nnitrosamines were assessed as a priority potentially hazardous inadvertently present chemical substance in samples of canned meat for infant nutrition for health risk assessment using methodological approaches developed in the Russian Federation. Assessment of the health risk to infants when consuming canned meat for infant nutrition containing Nnitrosamines on the example of products sold in the Russian Federation and the Socialist Republic of Vietnam revealed no excess hazard quotients (HQ < 1.0) in both territories. However, the calculation of carcinogenic risks has shown that when canned meat for infants sold in the Russian Federation from 6 months to 3 years of age is consumed, a carcinogenic risk may be formed in infants of the corresponding group. Most coughmedicines in Vietnam were extracted from Ivy leaves because of its 5 main active components, in which 4 Saponins included Hederacoside C (predominance), α-Hederin, Hederacoside D, Hederasaponin B are responsible for eliminating congestion (breaking up the phlegm and mucus) and Flavonoid: Kaempferol 3-rutinoside plays the role of reducing inflammations [1]. This study aimed to develop a HPLC-PDA method to simultaneously and fast analyze these 5 compounds in materials and dietary supplements containing hedera helix extract in Vietnam market. After the simple preparation procedure, the analytes were separated by using a C18 column (150 mm × 4.6mm, 5 µm) as stationary phase, and a mixture of 0.1% phosphoric acid and acetonitrile as mobile phase. The detection and quantification were in PDA detector at 205 nm. The method validation followed AOAC criteria. The calibration curves in the range of 0.5 - 200 mg/L for 4 saponins and 0.1 - 100 mg/L for the flavonoid with high correlation coefficient (R2 > 0.9999). The MDL (0.03 - 0.15 mg/kg) and MQL (0.15 - 0.50 mg/kg); RSDr (%) for repeatability (1.01 - 3.90%) and RSDR reproducibility (1.25 - 6.89%); recoveries (91.3 - 106%) for 5 compounds satisfied the AOAC requirements. The method was applied successfully for determining the content of the analytes in 10 real samples including dried ivy extract powder, dried leaves, and some cough relief products purchased from markets in Hanoi. This study has validated the method of determining resistant starch content according to AOAC 2002.02 method. The method has been validated with parameters of calibration curve, repeatability, intermediate reproducibility, and accuracy as required by AOAC. The robustness of the method was evaluated by comparing the content of total resistant starch and digestible starch with the total starch content according to the AOAC 996.11 method. This study has developed gas chromatography-mass spectrometry method GC-MS to determine the benzene content in foods containing benzoate salts and ascorbic acid simultaneously. The analyte sample was distilled to recover benzene and analyzed on GC-MS using a purge and trap injector, the analysis was controlled by benzened6 internal standard. The method has been validated for the parameters specified by AOAC. The method detection limit was 0.05 µg/kg, the method quantification limit was 0.15 µg/kg, the recovery ranged from 86.6 to 110.9%, Repeatability, RSDr in the range of 4.0 - 7.2%. The method has been applied to analyze the benzene content in 120 food samples including: soft drinks, fruit nectar, chili sauce and jam. National Institute for Food Control has implemented several studies, characterization of microbiology chemistry for food and feeding stuffs certified reference materials (CRM) in order to support laboratories in control and maintaining test results quality. CRM for Escherichia coli (E. coli) in food is our study’s first interest, because E. coli indicates unfavorable hygienic conditions and fecal contamination in foods. Milk and dairy products are used for any stage of human life from infancy to elder. CRM for E. coli in milk-based products were characterized by using a network of competency laboratories leading to certified values in accordance with ISO 17034:2016 requirements. Countries might utilize risk analysis frameworks to evaluate the impact as well as manage the risk of foodborne diseases from consuming contaminated foods in the food market. It is essential to obtain general information regarding hazard identification in the risk assessment process. Therefore, this literature review was conducted to identify common public health risks related to microbial contamination of food. Published studies with quantitative, qualitative, or mixed research methods were searched from peer-review journals. The chosen full-text articles were analyzed using an emergent thematic analysis approach to find key concepts and main results and then synthesized each item. The results of the literature review imply that food safety still has a significant impact on the health of consumers. Therefore, strict national risk-based food control systems should be considered to protect the health of the public. The method shows high specificity, and the calibration curve has a good correlation coefficient with R2 greater than 0.995; the limit of detection (LOD) and limit of quantification (LOQ) were 1.88 µg/kg and 6.26 µg/kg, respectively; The relative repeatability standard deviation (RSRr) was in the range of 2.46 - 7.34% and the recovery (R) was in the range of 85.7 - 100%. The method was applied to evaluate the mercury content in 39 health supplement samples taken in Hanoi city. Residues of chlorate and perchlorate in the sample were extracted with a methanolic solution acidified with 1% formic acid. Then, the extract was cleaned with C18 powder, used a normal phase column to separate chlorate and perchlorate, and quantified on an LC-ESI-MS/MS instrument. The method was validated for good specificity, linear calibration curve in the range of 0.0005 - 0.015 mg/L; the limit of quantification (LOQ) was 0.005 mg/kg; the recovery and relative standard deviation was from 91 to 109% and from 6.4 to 9.5% respectively and all met the requirements of SANTE. The method was applied to the real analysis, a total of 427 pangasius fillet samples collected from seafood processing companies. There were 76 samples were detected chlorate equivalent to 18%.
