Epifitik liken türlerinin (Hypogymnia physodes (L.) Nyl., Parmelia sulcata Taylor ve Pseudevernia furfuracea (L.) Zopf) fotosentetik pigment içerikleri üçer aylık zaman aralığıyla ölçülmüş, bir yıllık pigment değişim oranları ve mevsimsel kirleticilere maruz kalma istatistiksel olarak değerlendirilmiştir. Klorofil bozulması H. physodes numunelerinde kontrole göre kış aylarında artmış, yaz aylarında ise azalmıştır. P. furfuracea örnekleri için kışın azaldı ve yazın arttı. P. sulcata'da, kontrol ve nakledilen numunelerde ölçülen değerler arasında anlamlı bir fark bulunmadı. Klorofil bozulması, uzun transplantasyon süresi olan kontrol örneklerine kıyasla P. furfuracea için önemli ölçüde artar ve P. sulcata için azalır. Klorofil a miktarı, H. physodes ve P. sulcata için transplantasyon süresinin uzamasına bağlı olarak kontrol örneklerine göre önemli ölçüde azalırken, P. furfuracea'daki değişim önemli değildi. This study was performed between 2015 and 2017. 1274 plant samples were collected from the research area and all of them were identified. 444 specific and infraspecific taxa in 269 genera belonging to 71 families were determined. Among them one taxon belongs to Pteridophyts, two of them are Gymnosperms, and 441 out of 444 taxa are from Angiosperms. {"title":"Investigation of the antimicrobial effects of carvacrol in clinical Candida isolates and imaging by immunoelectron microscopic method","authors":"B. The efficacy of many antifungal agents are limited by their cytotoxicity and increasing rate of drug-resistant. In last years, the antimicrobial and antibiofilm activity of some natural products such as essential oils have drawn attention. Carvacrol [2-methyl-5-(1-methylethyl)phenol] is one of the phenolic components of thyme and have strong antimicrobial activity. Thus far, limited reports have discussed the antimicrobial effects of carvacrol on clinical Candida strains. In our study, it was aimed to investigate the effects of carvacrol on clinical Candida isolates by microbiological and transmission electronmicroscopic methods and to investigate the effects of carvacrol on the fungal cell wall by immunoelectron microscopic method using the hyphal wall protein Hwp1 protein. In this study, 24 clinical isolates and 1 reference strain (C. albicans ATCC 14053) were used. The minimum inhibitory concentration (MIC) of carvacrol was determined using the broth microdilution method. MIC results showed a MIC ⩽ 0.031% (vol/vol) for all isolates tested. For transmission electron microscopic studies, isolates were also exposured to the carvacrol at concentration of 1⁄2 MIC for 48 hours and results were compared with the control. According to our results, carvacrol showed high antifungal potential with very low MIC values on Candida isolates. Electron microscopically, no growth was observed at the MIC value and the higher concentrations; cellular damage was also determined at sub MIC concentrations. It has been shown that carvacrol causes irreversible damage to cells. We observed that carvacrol did not increase cell growth or hyphal growth at all studied concentrations. The immunogold labeling results were used to observe the effect of carvacrol on the cell wall. Although immune labeling was greatly reduced in carvacrol treated cells, the presence of Hwp1 protein was also observed in the scattered cytoplasm. Bu bağlamda sera koşullarında çelik ile çoğaltım metodu ile yetiştirilen Yeşilgül bitkilerinin bitki gelişim özellikleri, yaprak özellikleri, stoma morfolojileri, yaprak oransal su kapsamları, yaprakların klorofil içerikleri ve yapraklardaki mikro ve makro besin elementi kapsamları belirlenmiştir. Çalışma sonucunda farklı Rosa taksonları ile karşılaştırılan ‘Yeşilgül’ bitkisinin yayvan/dik yapıda olduğu, ortalama yaprak alanının 17.17 cm2, ortalama pedisel boyunun 1.45 cm, yaprak oransal su kapsamının %77.42, klorofil miktarının 40.87 olduğu tespit edilmiştir. R. chinensis ‘Viridflora’ bitkisine ait yaprakların stoma morfolojisi parametrelerinde ortalama değerler aldığı sonucuna ulaşılmıştır. Referans değerler doğrultusunda Yeşilgül yapraklarında azot, fosfor ve manganez değerleri düşük, bor ve kalsiyum değerleri ise yüksek bulunmuştur. Bu çalışma, Se ve B'nin Siklofosfamid (SFD) ile indüklenen kemik iliğinde miyeloid koruyucu potansiyellerini ve bu anti-kanser ilacın miyelotoksik özelliğinin kullanımını sınırladığını göz önünde bulundurarak deneysel sıçanlarda hematolojik toksisiteyi karşılaştırmayı amaçlamaktadır. SFD'nin kemik iliği ve kan hücreleri üzerindeki toksik etkilerini önlemede selenyumun bordan daha iyi bir koruyucu etkiye sahip olduğunu varsaydık. Bu eser elementlerin en sık kullanılan optimal dozları olan 1.5 mg/kg Se ve 20 mg/kg B hayvanlara deney boyunca intraperitoneal olarak verildi. Sadece 4. günde 200 mg/kg SFD uygulandı. Hayvanlar sakrifiye edilerek hematolojik değerlendirmeler için kan ve kemik iliği örnekleri alındı. SFD uygulaması, lökosit (WBC), trombosit (PLT), eritrositler (RBC) ve kemik iliği çekirdekli hücre sayılarını önemli ölçüde azalttı. Öte yandan, SFD ile birlikte Se ve B verilen gruplarda, sadece SFD verilenlere göre önemli miktarlarda arttı. Ancak Se'nin, istatistiksel anlamlılık elde edememesine rağmen, SFD'nin neden olduğu kemik iliği ve hematolojik toksisiteyi önlemede B'den daha koruyucu olduğu kanıtlanmıştır. Teh total protein amount of teh venoms was measured wif teh Nano Ready Touch device at a wavelength of 280 and was prepared to be equal to 2mg/mL concentration. Afterwards, teh supernatants were separated by centrifugation at +4 ºC for 15 minutes at 15,000 rpm, and 250 µL of matrix solution (18 mg/mL α-CHCA, 1:1; v/v) was added and scorpion venom matrix (AVMx) samples were added. has been prepared. For analysis wif MALDI-TOFMS (Microflex-LT), parallel spots (1µL) from AVMx samples of both scorpion venoms were placed on teh steel plate, and loaded into teh device after drying at room temperature. Teh system was operated in linear positive ion mode in teh mass range of 1-50 kDa, and a 60 Hz nitrogen laser at 337 nm was used as teh ion source. Mass spectra were generated using teh flexAnalysis software (version 3.4). All data were generated virtual gel images containing teh projection of teh peaks in teh venom spectra using teh filo-proteomic TEMPprincipal component analysis (PCA) method supported by teh built-in MATLAB software integrated into teh MALDI Biotyper software (version 3.1). As a result, it has been experimentally determined dat teh relative ratios of peptide and protein molecules and venoms may vary in scorpions belonging to teh same species. At the end of the research, 78 taxa belonging to the Charophyta division were identified. 7 of them, which are Closterium jenneri var. curvatissimum (West & G.S.West) A.J.Brook & D.B.Williamson, Cosmarium margaritiferum f. regularis (Nordstedt) West & G.S.West, Euastrum ansatum var. concavum Willi Krieger, E. verrucosum var. coarctatum Delponte, E. verrucosum var. planctonicum West & G.S.West, Staurastrum capitulum Brébisson and S. sexcostatum Brébisson ex Ralfs, were determined as new records for the desmid flora of Turkey.
