Methods Primary pelvic serous cancer tissues and ascites were obtained from 17 female patients without preoperative chemotherapy from July 2011 to July 2013.Side population(SP)cells and aldehyde dehydrogenase positive(ALDH+)cells were tested by Hoechst33342 staining and ALDH staining respectively to observe their expression difference and their relationship with pelvic metastasis. Results Among the detectable specimen,73.3%(11/15)were detected SP cells positive(percentage of SP cells0.15%),and 100%(10/10)ALDH+cells in either primary foci or ascites.Comparing the cells percentage from different sites in the same patients,73.3%(11/15)of the patients showed higher SP cells in ascites than those in primary tumors(P=0.01),whereas 90%(9/10)of the patiens showed higher ALDH+cells in ascites than those in primary tumors(P=003).Furthermore,patients with both SP positive and more ALDH+cells in ascites showed more metastatic sites than those with only SP positive or ALDH+cells(P0.05),however,they had little difference in metaststic rate(P0.05). Conclusions Higher percentage of SP cells and ALDH+cells in ascites indicates that pelvic serous CSC are related with peritoneal metastasis and disease progression.","PeriodicalId":12635,"journal":{"name":"Fudan University Journal of Medical Sciences","volume":"41 1","pages":"727-733"},"PeriodicalIF":0.0,"publicationDate":"2014-11-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"70185182","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods Immunohistochemistry(IHC),RT-PCR and Western blot were`used to identify moesin expression in breast cancer tissue samples and a series of human breast cancer cell lines with different metastatic potentials.RNAi technique combined with invasion and motility assay in vitro were also used to reveal the interrelationship between moesin deficiency and invasion and motility ability of breast cancer cells.Results IHC analysis showed that moesin were mainly distributed on the cell membrane and cytoplasm.Its expression was higher in metastatic breast cancer tissue samples than in non-metastatic ones(P=0.001,χ2=17.536).Western blot results revealed positive correlation between moesin expression and metastatic potential of breast cancer cells.The results of Transwell experiment displayed that moesin expression silencing remarkably inhibited invasion and motility ability of breast cancer cells,especially the cells with high metastatic potential.After moesin expression,the levels of MTDH mRNA and nm-23mRNA in breast cancer cells(P0.05)significantly decreased,while vimentin expression increased,and the expressions of E-cadherin and MMP-9decreased. Conclusions Moesin over-expression presents a positive correlation to breast cancer metastatic potential.Moesin may participate in the process of invasion and metastasis of breast cancer cells through influencing MTDH and nm-23,enhancing epithelial mesenchymal transition and increasing MMP-9secretion.","PeriodicalId":12635,"journal":{"name":"Fudan University Journal of Medical Sciences","volume":"40 1","pages":"679-684"},"PeriodicalIF":0.0,"publicationDate":"2013-11-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"70185078","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
