The aim of the current study is to explore the role of eugenol on γ-rays-induced acute kidney lesions in rats. The rats were separated into four groups (n=6). The control group received saline for two weeks. The eugenol group received i.p. 20 mg/kg Eugenol for two weeks. The γ-rays group was pretreated with saline for a week, then whole-body exposed to 8 Gy γ-rays and saline treatment continued for another week. The rats in γ-rays+ eugenol group received Eugenol for a week pre-irradiation, then were exposed to γ-rays and eugenol treatment continued for another week. The blood and kidney samples were collected for histopathological and biochemical investigations. It was observed that exposure to γ-rays induced atrophy of glomeruli with increasing capsular space, expanded renal tubules and congested inter tubular blood vessels. Histological changes were accompanied by significant decreases in the level of antioxidants: reduced glutathione content (GSH), and superoxide dismutase (SOD), and catalase (CAT) activities with a significant increase of malondialdehyde (MDA; end-product of lipid peroxidation), indicating oxidative stress. Significant increases in the levels of inflammatory markers, tumor necrosis factor-alpha (TNF-α) and interleukin-1 Beta (IL-1β) levels were recorded also. Kidney damage was substantiated by significant increases in urea, creatinine, lactate dehydrogenase (LDH), γ-glutamyltransferase (γGT), and disturbance of electrolytes balance; elevated sodium (Na + ), and decreased potassium (K + ). Eugenol treatment has significantly improved histological damage oxidative stress and inflammatory-biomarkers, which was accompanied by significant amelioration of kidney functions. Therefore, the purpose of the present study is to evaluate the histopathological consequence of gamma radiation (15 Gy) on submandibular glands as well as cytokeratin (CK) 17 and calponin expressions for deeper understanding of the mechanism of radiation –induced salivary gland injury. Rats were divided into control and irradiated groups. Rats’ heads in irradiated group were subjected to a single dose of gamma radiation (15Gy). The specimens of submandibular gland were examined for the histological changes and CK 17 and calponin expression. The intensity of CK 17 expression and also the area percent of CK 17 and calponin expression were measured. Data revealed acinar cytoplasmic vacuoles, loss of architecture and shrinkage in irradiated glands. Some areas showed degranulation of granular convoluted tubules and duct degeneration. The intensity of previously mentioned changes was increased by time. The intensity of CK 17 expression rose with time for both ductal and acinar cells while the area percent of expression decreased. The area of calponin expression decreased up to seven days after radiation and then increased at fourteen days. Rashed","doi":"10.21608/ejrsa.2023.194141.1152","DOIUrl":"https://doi.org/10.21608/ejrsa.2023.194141.1152","url":null,"abstract":"V ARIETIES of cellular stressors are reported to disrupt brain circuits or neuronal pathways that distribute neurotransmitter signals, resulting in subsequent genetic abnormalities, behavioral impairments, and/or mood disorders. For example, unfolded proteins accumulation in the endoplasmic reticulum (ER) induces ER stress-mediated cell death. Astringenin (AST) flavonoid functions as a neuroprotective agent and rescues neurons from various insults. However, the molecular mechanisms underlying the neuroprotective effects of Astringenin are still unclear. Rats received a single intraperitoneal (i.p.) injection of reserpine (RES) (5mg/kg) or exposed to a single dose of 10Gy head ionizing radiation (RAD) followed by AST treatment and sustained for 7 days. The expression of α-synuclein increased following head irradiation or reserpine administration, and endoplasmic reticulum stress factors were increased in brain tissue. AST administration results in upregulation of expression of AMPK and SIRT1, resulting in the inactivation of NF-κB p-65, FOXO1, and caspase-3. In addition, reduction of α-synuclein by AST improved the mobility-deficient behavior in rats. AST has also attenuated the decrease of dopamine, serotonin and BDNF levels. Moreover, AST upregulation of NRF2/HO1 levels was accompanied by enhancements of GSH content and SOD activity, and decreased inflammatory cytokines (TNF-α, IL-1β and IL-6).
