To explore the function of S6K in the ageing process of D. pulex, we cloned and analysed the full-length cDNA of S6K. With the growth of D. pulex, the expression of S6K mRNA first increased, then peaked at 10 days, and then decreased. Western blot analysis showed that the S6K protein expression increased from 1 to 5 days of age, but did not change significantly from 5 to 25 days of age. RNAi knockdown of S6K, reduced the expression of S6K mRNA and S6K protein, and it causes changes in other genes associated with ageing in both sexes. S6K may be involved in reproductive transformation, affecting reproductive capacity, thus significantly affecting the ageing of D. pulex. miRNA with potential regulation of the S6K gene were screened by the bioinformatics method and validated using a dual luciferase reporter gene system. The aim of the present study was to do an exhaustive review on the ectocommensals on Chilean inland-water decapods. The literature revealed the presence of species of Protozoa, viz., eight known only from Parastacus pugnax (Lagenophrys andos, Operculigera insolita, O. parastacis, O. seticola, O. striata, O. taura, Chilenophrys allendei, and Circolagenophrys shiftus), one for both P. pugnax and P. nicoletti (L. anticthos), one for Parastacus pugnax and Samastacus spinifrons (O. asymmetrica), and one reported for Aegla laevis (O. velata). In addition, the presence of flatworms was described, specifically Temnocephala chilensis, associated with A. laevis, P. pugnax, and S. spinifrons, and species of Polychaeta, i.e., Stratiodrilus platensis associated with A. abtao and A. bahamondei, S. aeglaphilus for A. laevis, and S. pugnaxi for P. nicoletti. These copepods turned out to belong to a new species of the freshwater genus Paracyclops. Paracyclops poppei, P. hirsutus, and P. chiltoni were the only species reported in Mexican freshwater ecosystems, and the record of P. fiersi sp. nov. is the fourth confirmed species of the genus from Mexico. The new species seems to be related to P. hardingi and P. bromeliacola. The new species differs in the ornamentation of the female antennary coxobasis, and in the posterior, subdistal, medial spinules on the coxa of the first–fourth leg in both sexes. The type series is mixed and includes specimens of two species. A syntype from Ponsee in Yunnan, P.R. China, is designated the lectotype of the species; those from Hotha in Yunnan are now referred to A. daiae (Zhang & Sun, in Zhang, Pan, Hao & Sun, 2020). Specimens which have previously been referred to as “A. tumidum” from other parts of Yunnan are here identified as a new species, Aiyunamon fatum sp. nov.. In this study, we compared the differences in metabolic enzymes and the mRNA expression of unfolded protein response (UPR)-related genes between the ammonia-resistant (LV-AT) and ammonia-sensitive (LV-AS) strains of Penaeus vannamei, aiming to investigate the different tolerance mechanisms of the two extreme strains underlying ammonia stress. The results showed that elevated ammonia concentrations mainly increased glutamine synthetase (GS) content while inhibiting transglutaminase (TG) content in the muscle of both strains and the contents were significantly higher in LV-AT than in LV-AS after 48 h of ammonia stress (). As a protective or adaptive strategy, the expression of the UPR related genes, including activating transcription factors ATF6, ATF4, and the spliced form of X box binding protein 1 (XBP1) were, respectively, either significantly induced, or inhibited () after ammonia stress. The mRNA expressions of the GRP78 (Bip) gene and the apoptosis gene (caspase 3) are then activated. In hepatopancreas and gills, the associated gene expressions gradually increase in LV-AT, while several related genes in LV-AS were first induced and then gradually inhibited. The expressions of key genes (GRP78, ATF6, XBP1 and ATF4) were significantly higher in LV-AT than that in LV-AS after 48 h of ammonia stress (). These results indicate that some enzymes involved in ammonia metabolism and UPR protection genes tended to be highly expressed in LV-AT. Hence, we speculate that LV-AT has a higher ammonia excretion capacity and a more effective adaptive mechanism after ammonia stress.
