The ameliorative potential of low-dose quercetin following cART-associated MOS-toxicities in cardio-hepato-renal organs was evaluated in in vivo model. Oral administration of cART (Dolutegravir 50 mg, Lamivudine 300 mg and Tenofovir disoproxil fumarate 300 mg [DLT]) at 9.29 mg/kg, was challenged against low-dose quercetin 25 mg/kg body weight (bw) in Wistar rats. Group 1, the normal control (NC) received distilled water (5 mL), while groups 2 to 4 received quercetin (25 mg), DLT (9.29 mg), and DLT + quercetin (9.29 mg + 25 mg respectively), per kg bw. All administrations lasted for 14 days, and thereafter animals were humanely sacrificed after intraperitoneal anesthesia injection with 100 mg ketamine /5 mg xylazine per kg bw followed by cervical dislocation. Blood and organs were harvested for analyses using standard protocols. The serum concentrations of lipid parameters [total cholesterol, triglycerides, LDL-cholesterol, and VLDL-cholesterol], liver biomarkers (total-bilirubin, direct-bilirubin, and transaminases], and kidney biomarkers [urea and creatinine] were significantly increased (p < 0.05) while electrolytes (Na+, K+, Cl− and HCO3−) were significantly decreased (p < 0.05) in DLT group but improved in DLT + Q group. Histopathology demonstrated distorted myocytes, hepatocytes and renal tubules, fatty liver with vacuolization, dystrophied glomeruli and distorted renal interstitium in DLT group, compared with normal appearing histoarchitectural features in NC and DLT + Q groups. The roots, leaves and seeds of this plant are used for different medicinal purpose. It principally carries flavonoids, triterpene glycosides, abrin and alkaloids. The plant had been mentioned for neuromuscular effects, neuro-protective, abortifacient, antiepileptic, anti-viral, antimalarial, antifertility, nephroprotective, immunomodulator, immunostimulatory properties, antiinflammatory activity, antidiabetic, and many others. The aim of this study was to investigate the anti-fertility activity of the methanol extract of Abrus precatorius seeds (APS) in female albino rats administered 45 and 50 mg/kg BW through oral gavage for 30 days. An Abrus precatorius seed (APS) was extracted (1:1 w/v) using methanol to obtain the crude extract. Liquid-liquid fractionation was performed on the crude methanol extract using solvents of different polarity. Eighteen female rats were divided into three groups each containing six animals was used for this experiment. Group one (control), received 1 ml of saline solution, group two received 45 mg/kg and group three received 50 mg/kg body weight by oral gavage daily for a period of 30 days. At the end of the study, Body weight, Organ Weight, Estrous cycle, Hormonal Concentrations, Oxidative Stress Markers, Enzymatic Activities, Biochemical indices (Liver and Kidney panel) and Hematological parameters were also evaluated. Consequent upon the administration of A.precatorius seed methanol extract 45 and 50 mg/kg BW into the female rats resulted in changes pertaining to Body weight, Estrous cycle, Hormonal Concentrations, Antioxidant Enzymes, Serum Biochemical indices and Hematological indices significantly (p < 0.05) compared to control rats. Initial body weights are significantly (p < 0.05) indicated, whereas final weights were decreased significantly (p < 0.05). Organ weight changes i.e. Ovary, Liver and Kidney weights were found to be significantly (p < 0.05) decreased in APS administered rats compared to control. Estrous cycle study i.e. time duration of Proestrus, Estrus, Metestrus and Diestrus (in days) was significantly prolonged. Hormonal Concentrations represented by FSH, LH, Prolactin, Estradiol were significantly (p < 0.05) decreased, whereas Progesterone levels were significantly (p < 0.05) increased in APS administered female rats compared to control group of rats. All the antioxidant enzyme assayed CAT, SOD, GPx, GR and GST were found to be significantly (p < 0.05) decreased, whereas MDA concentration was found to be increased significantly (p < 0.05). There is a medical need for alternative and safer therapies as LT4 is associated with special treatment considerations and adverse effects. Thyrogrit (THY) is a polyherbal formulation indicated for the treatment of hypothyroidism. The present study, describes the characterization of the phytocompounds present in THY and its in-vivo efficacy in rat model of hypothyroidism, in combination with a sub-optimal dose of LT4. Ultra High Performance Liquid chromatography was employed for the identification of the phytocompounds present in THY. For the evaluation of its in-vivo efficacy, female Wistar rats were administered THY orally, 15-days prior to disease induction, and continued throughout the experiment. Subsequently, hypothyroidism was induced by oral administration of propylthiouracil (PTU). From day 45 onwards, animals were administered orally with a sub-optimal dose of LT4 (2 μg/kg) till the end of the study. On day 79, animals were euthanized, blood was collected for measurement of thyroid hormones and other clinical chemistry parameters. Weights of liver, kidney and thyroid were recorded. Finally, the thyroid was subjected to histopathological evaluation through hematoxylin and eosin (H&E staining), immunohistochemistry as well as immunofluorescence. The principal phyto-components detected in THY by Ultra High Performance Liquid Chromatography included gallic acid, protocatechuic acid, corilagin, ellagic acid, piperine, guggulsterone E and Z, which are documented to exerted beneficial effects on thyroid function. In the in-vivo study, THY when supplemented with a low dose of levothyroxine restored the PTU-induced reduction in the serum levels of T3 and T4 and improved PTU-induced renal impairment. THY treatment ameliorated the hallmark histopathological changes associated with hypothyroidism and C-cell hyperplasia. Further, co-administration of THY and LT4 did not show any major non-clinical safety concerns even after the administration for more than twelve weeks. This study has demonstrated that co-administration of THY and LT4 improves the PTU-evoked alterations in the thyroid ultrastructure and function, abrogates hypothyroidism-associated renal impairment and exhibits an acceptable basic safety profile. Kaempferol have been reported to have several therapeutic benefits. In this study, the therapeutic value of kaempferol was investigated in relation to the cardio-nephrotoxicity in rats resulting from E. ocellatus envenoming. Fifty male wistar rats were allotted unbiased into five groups (n = 10) for this study. Group 1 was the control, while rats in groups 2 to 5 were envenomed with LD50 of E. ocellatus venom (0.22 mg/kg bw; i.p.). Group 2 was not treated after envenomation while groups 3, 4 and 5 were treated with polyvalent antivenom, 4 and 8 mg/kg of kaempferol, respectively. E. ocellatus envenomation caused considerable reduction in organ weight and relative organ weight in the envenomed untreated rats. The venom induced intense oxidative stress, inflammation, apoptotic damage to the cardiac and renal tissues accompanied with severe histomorphology in the organ tissues of untreated envenomed rats. In contrast, kaempferol treatment post-envenomation attenuated the venom-induced cardio-nephrotoxic responses in a dose dependent effect. Kaempferol substantially (p < 0.05) decreased malondialdehyde levels while enhancing reduced glutathione levels and superoxide dismutase and glutathione peroxidase activities in the heart and kidney of envenomed treated rats. Treatment of envenomed rats with kaempferol successfully decreased nitric oxide levels and myeloperoxidase activity. Overexpression of apoptotic caspase 3 and caspase 9 in cardiac and renal tissues were suppressed by kaempferol (p < 0.05). The histopathological result supports kaempferol’s ameliorative ability by convalescing the severe morphological alterations of cardiac and renal tissues induced by the venom. Although many diseases have been successfully treated with herbal treatments for a long time, the majority of the herbs utilized have unclear mechanisms. Therefore, this study aimed to examine the modulation effects of green tea oil (GTO) and pumpkin oil (PO) on hyperlipidemia, oxidative stress, and hematological abnormalities in an experimental multiple sclerosis rat model. Forty albino male Wistar rats (weighing 120–140 g) were divided into four groups of six each: group 1, the control group; group 2, the myelin oligodendrocyte glycoprotein (MOG)-injected group; and groups 3 and 4, the MOG-injected groups treated with GTO and PO at 5 mg/kg b.w., respectively. At the end of the experiments, animals were anesthetized with diethyl ether inhalation, and blood samples were collected from the jugular vein. A Beckman Coulter was then used to determine the differential complete blood counts. The obtained serum was rapidly collected and stored at 20 °C to assess the lipid profile and oxidative stress and antioxidant biomarkers. Our findings showed that GTO and PO treatment produced a significant reduction in total cholesterol (TC), triglycerides (TG), high-density lipoprotein-cholesterol (HDL-C), low-density lipoprotein-cholesterol (LDL-C), and very low-density lipoprotein-cholesterol (VLDL-C) levels. Furthermore, GTO and PO treatment alleviated the elevated cardiovascular risk indices 1 and 2. Thiobarbituric acid reactive substance (TBARS) concentration significantly decreased and glutathione (GSH), superoxide dismutase (SOD), and glutathione peroxidase (GPx) levels significantly increased in rats injected with MOG and treated with GTO and PO. Furthermore, after GTO and PO treatment, the reduced red blood cells (RBCs) count, hemoglobin content (Hb%), lymphocyte percentage, and hematocrit (HCT) of MOG-injected rats increased, while the elevated white blood cells (WBCs), platelet, and neutrophil percentage substantially declined. To assess the efficacy of Costus speciosus ointment as a potential alternative to commercially available ointments, a study was conducted using a cohort of 16 healthy male mice, aged between 3 and 4 months. The ointment formulation was prepared utilizing ethanol extracts from both the leaves and seeds, presented in a dosage form for application. This investigation seeks to provide insights into the comparative effectiveness of Costus speciosus ointments in relation to conventional market preparations, with a focus on its potential applicability as an alternative therapeutic option. The parameter measured was wound contraction and epithelization period. The mice groups treated with seed & leave extracts ointment showed better wound size changes than the negative and standard groups. However, the leaf extract ointment promotes the formation of granulation in tissue, allowing the reepithelization phase to occur more rapidly than seed extract. From the 1st day to the results of the 9th day of the extract indicate that there is a significant increase (P < 0.05) in the percentage of wound contraction in the group. After 21 days the wound was healed fully. These results suggest that ointment of ethanol leaf extract of Costus speciosus could be an answer to facilitate wound healing compared to seed extract, to approve the traditional claims of the plant in wound healing activity. The study concludes that ethanol leaf and seed extracts of Costus speciosus exhibit potential wound healing activity in mice excision wound models. The topical formulation of these extracts significantly enhances wound closure, reduces wound size, and increases tensile strength compared to the control group. The efficacy of the extracts is comparable to the standard drug, Neomycin sulphate ointment. Therefore, the use of Costus speciosus extracts in topical formulations can be considered a promising alternative for the treatment of wounds. This study elucidates the ameliorative potential of the tunic of onions against Wistar kidney rats toxified with sodium oxalate. Ethylacetate extract of the tunic of onions otherwise regarded as Onions peel extract (OPE) in this study was prepared to get the flavonol-rich extracts. Adult male Wistar rats received 70 mg/kg body weight sodium oxalate with or without co-treatment with OPE, quercetin or cystone. Biochemical analyses were carried out on the plasma and urine, followed by a histopathological assessment of the kidney. Intoxication with sodium oxalate brought about electrolyte imbalance, nephrotic syndrome (high concentrations of total protein and albumin in the urine and low concentrations in the plasma) reduced renal function (low renal clearance of creatinine and urea) and damage to the kidney as well as fluid accumulation. Treatment with flavonol extract from onion tunic mitigated these deleterious changes as a result of sodium oxalate intoxication. The finding suggests that onion peel has the potential to prevent damage arising from oxalate toxicity in the kidney. This study which deals with age-related wound healing, investigates the healing effect of Nishamalaki, a classic antidiabetic Ayurvedic formulation comprising turmeric (Curcuma longa) and Indian gooseberry (Phyllanthus emblica), on cutaneous wounds in aging rats. Rats with excision wounds of 7 mm created on their dorsal side received Nishamalaki (500 mg/kg p.o) daily, or a combination of Nishamalaki (500 mg/kg p.o) with 1% Nishamalaki gel applied on the wound or the reference standard metformin (2 μmol) applied on the wound daily till the scabs fell off. All treatments enhanced the rate of formation of granulation tissue and wound contraction. All treated rats showed lower blood glucose levels compared with their 1st-day values and significantly lower blood glucose levels when compared with the Aged Control rats. A significant restoration of the aging-depleted L-hydroxyproline, hexosamine, ascorbic acid, PDGF, AMPK, and mTOR levels, and attenuation of the aging-elevated IL-6 and TNF-α levels was elicited by all treatments. The treatments significantly restored the aging-depleted endogenous antioxidants. The Nishamalaki combination treatment of the oral extract and topical gel displayed a better wound-healing effect than the oral treatment alone. The histopathological studies on skin ulceration, hair follicles, granulation tissue, and collagen fiber formation of the wound tissues corroborated the biochemical findings. Curcumin and other antioxidant polyphenolic components of Nishamalaki may be responsible for its wound-healing effect. For the first time, the present study has investigated the action of PDGF, AMPK, and mTOR on cutaneous wounds. They seem to be acting together to promote wound healing and repair. Artocarpus altilis possesses antioxidative, anti-inflammatory, and anti-proliferative properties. Aluminum (Al) is extensively utilized for consumer products, cooking utensils, pharmaceuticals, and industries. Indication for the neurotoxicity of Al is investigated in various studies, notwithstanding the precise mechanisms of Al toxicity are yet to be fully elucidated, and, which requires novel therapy. In this study, we determined the ameliorative role of Artocarpus altilis on aluminum chloride-induced neurotoxicity in Drosophila melanogaster. Varying concentration of the extract were used to formulate diets for 6 groups of flies. Group 1 contained basal diet, group 2 contained basal diet and aluminium chloride (AlCl3), group 3 contained basal diet + 0.1% unseeded breadfruit (UBF), group 4 contained basal diet + 1% unseeded breadfruit, group 5 and 6 contained basal diet + AlCl3 + 0.1% and 1% unseeded breadfruit. Assays such as acetylcholinesterase activity, malondialdehyde (MDA) concentration level, catalase activity, and superoxide dismutase (SOD) activity were carried out after 7 days of exposure respectively. The results showed low activity of acetylcholinesterase activity and MDA level and high catalase and SOD activity in the pretreated and post-treated flies with Artocarpus altilis compared to the normal and negative control respectively. Taken together, Artocarpus altilis is a promising prophylactic, antiacetylcholinesterase, and antioxidant plant in the prevention, management and treatment of neurodegenerative diseases.
