Masseyeff","doi":"10.1016/S0980-9090(88)80005-3","DOIUrl":"10.1016/S0980-9090(88)80005-3","url":null,"abstract":"<div><p>Bien que le processus de remplacement progressif des anticorps polyclonaux (ACP) par des anticorps monoclonaux (ACM) apparaisse irréversible, les auteurs ont voulu faire le point sur les avantages et inconvénients réciproques des deux variétés d'anticorps.</p><p>Au plan de la spécificité, les ACM ont nettement amélioré le pouvoir discriminant par un meilleur ciblage de l'épitope (ex. molécules à fortes homologie, énantiomères, antigènes en présence d'anticorps, etc.), cependant il faut se garder des conséquences d'un excès de spécificité (en bactériologie, virologie ou en cancérologie).</p><p>En regard de l'affinité, les ACM présentent des constantes d'équilibre généralement inférieures à celles des ACP. Les raisons invoquées tiennent dans les protocoles d'immunisation ou dans la nature des schémas réactionnels. L'importance du paramètre affinité n'est pas la même dans les méthodes par compétition ou par extraction-saturation.</p><p>L'article aborde également le rôle (moins connu) des parties constantes de l'anticorps (fragment-Fc) au niveau de la fixation sur les phases solides et de la stabilité à la conservation.</p></div><div><p>Even though the progressive replacement of polyclonal antibodies by monoclonal antibodies appears to be irreversible, the authors would like to take an observation of their reciprocal advantages and disadvantages.</p><p>The specificity has been greatly improved by monoclonal antibodies, thanks to the epitope-specificity (structure-like molécules, enanliomers, antigens complexed with endogenous related antibodies).</p><p>However, one has to be very careful that a very high specificity may induce unwanted consequences (in bacteriology, virology, oncology).</p><p>As far as affinity is concerned, the equilibrium constants of the monoclonal antibodies are generally lower than the one of polyclonal antibodies. This results from the immunisation procedures and the schemas of the reactions themselves. Affinity is not similarly important in competition techniques or in techniques involving extraction-saturation.</p><p>This paper also deals with the role played by the constant fragment of the antibody (Fc), concerning the binding on solid phase and the stability for conservation.</p></div><div><p>Obwohl die allmähliche Ablösung der polyklonalen Antikörper (PAK) durch monoklonale Antikörper (MAK) irreversibel erscheint, beabsichtigen die Autoren, hier noch einmal eine Bestandsaufnahme der Vor- und Nachteile der beiden Antikörperarten zu machen.</p><p>Bezüglich der Spezifität haben die MAK's die Unterscheldungsfähigkeit durch eine höhere Zielgerichtetheit auf das Epitop (stark homologue Verbindungen, Enantiomeren, Antigene im Beisein von Antikörpern usw.) deutlich verbessert ; vor den Folgen eines Spezifitätsüberschusses wird jedoch gewarnt (betreffend : Bakteriologie, Virologie und Kanzerologie).</p><p>Im Hinblick auf die Affinität besitzen die MAK's allgemein schlechtere Gleichgewichtskonstanten als die PAK's. Those can be used as detoxification means, make possible the detection of endogenous molecules corresponding to the synthetic drugs, and allow for a study of those very macromolecules which are the targets of the drugs.</p><p>The reader will be given examples of application for each previously listed section. Those will detail: intoxications due to digoxin, colchicine, aflatoxin, the use of antibodies to benzodiazepine in the regulation of human psychism, and the action of pharmacologic agents on receptors.</p></div><div><p>Der Autor stellt weniger bekannte, gegen Medikamente gerichtete monoklonale Antikörper und deren Anwendungsmöglichkelten vor : als Detoxikationsmittel, zum Nachweis endogener Verbindungen, die synthetisch hergestellten medikamentösen Verbindungen entsprechen, sowie zur Untersuchung von Makromolekülen, die Medikamenten als Zielorte dienen.</p><p>Zu jedem dieser Punkte findet der Leser Anwendungsbeispiele, z.B. On assiste au développement d'automates intelligents qui prennent en charge le pilotage des mesures, leur traduction en concentration, parfois même une partie des tâches administratives du laboratoire.</p><p>L'immuno-analyse est un domaine particulier de la biologie médicale puisqu'il n'existe pas de relation simple entre la réponse physique du système de dosage (radioactivité, densité optique, fluorescence…) et la concentration du produit à doser dans l'échantillon testé. Le passage de l'un à l'autre est réalisé grâce à une courbe de calibration. Les premiers logiciels apparus en immuno-analyse ont réalisé l'interpolation des concentrations. Deux autres tâches sont venues en complément : estimation de l'intervalle de confiance de chaque résultat et aide à la validation des dosages.</p><p>Après avoir présenté les différentes méthodes d'ajustement pour décrire les courbes d'étalonnage, l'auteur attire notre attention sur le fait que l'ordinateur « peut se tromperet propose une explication cohérente pour l'origine d'erreurs qu'il importe à tout prix d'éviter.</p></div><div><p>The introduction of computers in the laboratory has the advantage of suppressing part of the routine work. Automation are made in charge of system control, data acquisition, measurements and final concentration estimation. They sometimes deal with a part of the administrative work in the laboratory. Immunoassay in a singular field in medical biology because there is no simple connection between the physical response of the assay system (radioactivity, optical density, fluorescece…) and the concentration of the substance of interest in the tested sample. A calibration curve makes a bind between both of them. The first specific software in the field of immunoassay have performed the interpolation of the concentrations. They were completed by two other means which proceeded in the determination of the confidence interval for each result and the help to validation of assay results.</p><p>The author first presents the various techniques to fit the calibration curves.</p><p>Afterwhich, he points out the fact that the computer can be wrong and explains the reasons why some errors may occur and therefore be awaided.</p></div><div><p>Die Zunahme eines EDV-Einsatzes im Laboratorium ist positiv zu bewerten, weil sie dem Anwender die Routinearbeiten abnimmt.
