Estrus Cycle indices are good parameters for evaluating reproductive changes, while actual ova shed represents evidence of ovulation. This study is aimed at investigating how different doses of CC can affect the Estrus Cycle (EC), Estrus Cycle Ratio, (ECR), and ova shed (OS) in adult female rats. Methodology: Twenty-five (50) female Sprague Dawley (SD) rats were divided into 5 groups. Group A was controlled while groups B, C, D, and E were treated groups. Group A received 0.5 ml of sterile water. Groups B, C, and D were given 0.2 mg/kg, 2 mg/kg, and 4 mg/kg respectively at diestrus. Group E was given 6mg/kg/day of CC dissolved in sterile water. The administration was done orally. A vaginal smear was evaluated for various estrus phases. ECR was calculated, and ova shed at the estrus phase was evaluated at autopsy. All data were presented as mean ± standard error of the mean (SEM). Statistical significance was taken at P < 0.05 Findings: There was a significant decrease (p<0.05) in the number of proestrus, estrus, metestrus, and diestrus in treated groups compared with control. ECR value in all treated groups also decreased significantly (p<0.05) when compared with control. This study further shows that all the animal groups (A-E) released ova but there was a significant decrease (p<0.05) in the number of ova released in groups B, C, D, and E against the control group. Conclusion: CC decreases EC. It reduced ECR, hence prolonging luteogenesis. It decreases the number of eggs released. With a large dose and prolonged use, eggs were still released even when the ova released and counted were fewer compared with when this drug was not in use. Recommendation: Infertile couples should copulate more at the luteal phase of the menstrual cycle when on CC to increase their chances of pregnancy. CC decreases the number of eggs released but makes the few eggs released stay longer for fertilization.","PeriodicalId":9711,"journal":{"name":"Central European Journal of Biology","volume":"60 2-3","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-10-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"136232764","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

For the treatment of CCHFV, there is still a lack of vaccine or any antiviral medicine. For this purpose, a study was performed to design a multiepitope based subunit vaccine (MESV) for effective prevention against CCHFV infection. Methodology: The study contains immunoinformatic and docking methodologies to obtain a MESV by choosing highly antigenic and overlapping epitopes comprising of 8 epitopes of both MHC class I and II from viral proteins. Epitopes were chosen which were conserved within the epitopes of IFN- gamma, T-cell and B-cell. Then these epitopes were joined to final peptide by GPGPG and AAY linkers. An adjuvant was added at N terminal of vaccine via EAAAK linker for the improvement of vaccine’s immunogenicity. Findings: Our final construct was comprised of 278 amino acids. To validate the vaccine’s immunogenicity and safety, its physiochemical properties, allergenicity as well as antigenicity were checked and it was predicted to be non-allergenic and antigenic. The construct was further analyzed by molecular docking within vaccine and TLR3 receptor to assure its molecular interaction and binding affinity. In the end, In-silico cloning was also carried out for ensuring its expression efficiency. Recommendations: Nonetheless, the designed construct is proposed to be tested in laboratory settings to confirm its immunogenicity and safety.","PeriodicalId":9711,"journal":{"name":"Central European Journal of Biology","volume":"3 3","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-10-21","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135512711","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

A strict gluten-free diet (GFD) is the only treatment that can eliminate CD complications. It was aimed to evaluate the effect of a gluten-free diet on serum total glutathione (tGSH) level, superoxide dismutase (SOD), myeloperoxida (MPO), paroxanase (PON-1) and aryl esterase (ARE) activity in patients with celiac disease, an autoimmune disease. Materials and Methods: The study was conducted with 68 participants, 39 of whom were celiac and 29 were healthy. Two groups were formed in patients with celiac disease as newly diagnosed and previously diagnosed and following a gluten-free diet. Blood samples were taken from all participants and tGSH, SOD, MPO, PON-1, and ARE measurements were made spectrophotometrically from serum samples. Results: While no significant change was observed in tGSH, SOD, and ARE levels, MPO activity was observed to be significantly lower in celiac patients compared to healthy controls, while this decrease was found to be higher in the newly diagnosed group. While PON-1 activity was significantly lower in newly diagnosed patients compared to the control group, it was higher in the gluten-compatible diet group. Conclusion: Low MPO values in celiac patients may be insufficient to function by creating oxidative stress in inflammation. While PON-1 values are significantly lower in newly diagnosed celiacs, it can be said that they reach normal values with adherence to a gluten-free diet.","PeriodicalId":9711,"journal":{"name":"Central European Journal of Biology","volume":"25 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-10-13","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135918825","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

They are responsible for higher morbidity and mortality rates resulting from dysentery every year across the world. Antibiotic therapy of this disease plays a critical role in decreasing the prevalence as well as the fatality rate of this infection. However, the management of this disease remains challenging, owing to the overall increase in resistance against many antimicrobials. Hence, it has become important to identify as well as develop therapeutic methods presenting novel avenues for infections. In the current study, proteome based mapping was utilized to find the potential drug targets for dysentery. There is need to identify novel drug and vaccine targets to control this disease. This study is designed to identify new drug targets to develop drug and vaccines to battle bacillary dysentery. Subtractive genomics approach was used in this study to find novel drug targets. Methodology: The proteomes of Shigella, E. coli, Campylobacter and Salmonella were retrieved from Uniprot. Paralogs in these proteomes were removed by CD-HIT. Gene essentiality was screened by Geptop server 2.0. Host-pathogen interaction was analyzed through HPIDB database. To identify non-homologous proteins, the essential proteins were analyzed in Blastp against Homo sapiens (H. sapiens). The unique metabolic pathways were recognized by comparison of metabolic pathways of selected strains of bacteria and H. sapiens using the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway. Subcellular localization and functional classification of proteins involved in unique metabolic pathways were analyzed through PSORTb and SVMProt. Druggability potential of unique essential proteins was investigated using the DrugBank database. Findings: Over all analysis showed 7 proteins that were common in all four bacteria. These proteins were essential to pathogens. Out of 7 proteins 6 proteins MURA, MURG, DAPA, MURB, DAPE, DnaA are reported as a drug target in different bacteria and one protein DAPD is novel. The study will enable the development of natural and cost-effective drugs against dysentery infections. Recommendation: However, further validations are necessary to confirm their drug effectiveness and biocompatibility.","PeriodicalId":9711,"journal":{"name":"Central European Journal of Biology","volume":"15 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-10-08","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135251043","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
