The study was carried out from August 2014 to May 2017. Zooplankton was collected from hauls at 20 m and 40 m depth contours off Mumbai and Ratnagiri coast. The results indicated that Species Richness d, Shannon H and Simpson 1-λ at Mumbai stations were low compared to Ratnagiri stations. The values were higher in post-monsoon (POM) and pre-monsoon (PRM) than in monsoon (MON) season. Two-way ANOVA revealed a significant difference (P<0.05) for values of ‘d’, ‘H’ and 1-λ ’ between Mumbai and Ratnagiri. Post-Hoc Tukey test showed a significant difference (P<0.05) between values of ‘d’, ‘H’ and 1-λ ’ during MON and POM, MON and PRM. However, no significant difference (P>0.05) was observed between POM and PRM. Cluster analysis showed a 35% similarity in diversity between MON and the other two seasons. SIMPER analysis of similarity showed average dissimilarity of 44.77 between the diversity of Mumbai and Ratnagiri. The study indicated seasonal variations in the zooplankton diversity in coastal regions of Mumbai and Ratnagiri. The impact of pollution and anthropogenic activities could have reduced zooplankton diversity and abundance in the coastal environment of Mumbai compared to the less polluted waters of Ratnagiri.","PeriodicalId":93791,"journal":{"name":"Journal of the Marine Biological Association of India. This is the first confirmed record of the species from the seas around India after 80 years of its original record. Four specimens were collected from the Arabian Sea from a depth of 450 m, during the fishery surveys onboard the FORV Sagar Sampada, using a mid-water trawl net. The dorsal mantle length (DML) of the individuals ranged from 27 to 36 mm. Random sampling was performed for a period of six months and conducted interviews with bait diggers, traders and local fishers (n=30). Polychaete annelids were found to be the most extensively harvested taxa in the intertidal areas due to market demand from the aquaculture industry. The collected polychaetes were identified as Marphysa spp. During the survey, each pair (two partners) on a typical day is reported to collect around 1-2 kilogram of live worms, which earns them ` 1000 per kg. It is recorded that the average collection of worms from various locations in intertidal areas of Mumbai city is around 150-200 kg/day. Last decade, the annual consumption of polychaetes by shrimp hatcheries ranged from 16 to 20 tonnes. Shrimp farming in India is intensified and currently, there are 315 hatcheries registered with the Government of India for seed production of Litopenaeus vannamei. The demand for polychaete worms has therefore increased many folds and could be estimated in the range of 252 to 337 tonnes annually and nowadays, this demand is being met by an unreported wild-caught polychaete resource as well as import of Specific Pathogen Free (SPF) polychaete from overseas suppliers. There is scope for polychaete culture to satisfy the country’s growing market demand. Moreover, polychaete fishing in coastal areas removes substantial biomass and hence can cause major impacts on coastal habitats. In this study, the water samples from Thumba Arattuvazhi Beach in Kerala were screened for l-asparaginase producing microorganisms. This was followed by colourimetric screening using modified M9 media with 0.009% Phenol red dye and using l-asparagine as a sole nitrogen source. Then, the Nessler assay was performed to quantify the enzyme. Molecular identification was made by 16SrRNA sequencing and aligned the sequence with GeneBank for phylogenetic tree construction using BLAST. Seawater was serially diluted for 10<sup>-1</sup> to 10<sup>-6</sup> using nutrient agar plates. A total of 19 bacterial colonies were isolated. The colonies were evaluated to produce l-asparaginase according to the pink zone around the colonies on the modified M9 medium using a red phenol indicator. The KB1 sample was selected for further studies according to plate colour assay. Nessler assay of L-asparaginase quantified as 2.537 IU/ml. Molecular characterisation showed the sequence association with <i>Bacillus altitudinis</i> the sequence submitted in Genebank as <i>B. altitudinis KB1</i> strain. The l-asparaginase II gene (AnsB) was amplified based on the entire length of the hypothetical protein of annotated genome with accession number CP022319.2. The l-asparaginase activity in this study was 57% higher than the reference organism <i>B. altitudinis</i> BITHSP010. The l-asparaginase producing bacterium <i>B. altitudinis</i> KB1 from a marine source in Kerala can produce asparaginase, which can be utilised for biotechnology applications.</p>","PeriodicalId":93791,"journal":{"name":"Journal of the Marine Biological Association of India.
