Tennekoon","doi":"10.9734/bjpr/2016/23317","DOIUrl":"https://doi.org/10.9734/bjpr/2016/23317","url":null,"abstract":"Aims: The present study was aimed to isolate active anti proliferative compound/s from the hexane extract of leaves of the mangrove plant Scyphiphora hydrophyllacea C.F.Gaertn. and evaluate their cytotoxic properties. Place and Duration of the Study: At the Institute of Biochemistry, Molecular Biology and Biotechnology and HEJ Research Institute of Chemistry, between January 2015 to November 2015. Methodology: The hexane extract of the leaves of S. hydrophyllacea which was found to be cytotoxic was fractionated using normal phase column chromatography and fractions obtained from Original Research Article Samarakkon et al.; BJPR, 10(1): 1-6, 2016; Article no.BJPR.23317 2 column chromatography were tested for anti proliferative effects. Active fractions obtained were further fractionated using normal phase column chromatography to obtain compound 1. Structure of the isolated compound was elucidated with combination of Hand C-NMR spectroscopy, and mass spectrometry techniques. Anti proliferative properties of compound 1 in three cancer cell lines {MCF-7 (estrogen receptor positive breast cancer), HepG2 (hepatocellular carcinoma) and AN3CA (endometrial cancer)} was evaluated by MTT assay after 24 and 48 h incubations. Results: Isolated compound was found to be hopenone-I, a triterpenoid and this is a novel finding for the presence this compound in the mangrove plant S. hydrophyllacea. Hopenone-I showed some promising anti proliferative effects in all the cancer cells in a time and dose dependent manner after 24 and 48 h incubations {MCF-7: 14.98 μM (24 h) and 7.76 μM (48 h), HepG2: 29.24 μM (24 h) and 11.60 μM (48 h), AN3CA: 12.15 μM (24 h) and 4.99 μM (48 h)}. It is used ethno medicinally in ulcer, jaundice and splenomegally. Phytochemical screening was carried out on the aqueous ethanol extract of the whole plant by using standard phytochemical methods. Antibacterial studies were also carried out on the aqueous ethanol extract by using diffusion method and broth dilution methods. The aqueous ethanol extract of the plant was found to contain alkaloids, flavonoids, anthraquinones, saponins and cardiac glycosides. The aqueous extract was found to be active against Staphylococcus aureus, Streptococcus faecalis, Escherichia coli, Helicobacter pylori and Shigella dysenteriae and had minimum inhibitory concentrations (MIC) of 10, 5, 5, 5, and 5 mg/ml and also minimum bactericidal concentration (MBC) of 20, 20, 20, 10, and 10 mg/ml respectively. The extract was also found to be inactive against Corynbacterium ulcerans and Salmonella typhii. Zones of inhibition produced by aqueous ethanol extract of plant were less than those produced by Original Research Article Shehu et al.; BJPR, 11(2): 1-6, 2016; Article no.BJPR.23046 2 ciprofloxacin (5 μg/ml). Endosulfan, a chlorinated cyclodiene insecticide, is widely and very liberally used in agricultural sector. But it has created deleterious effects on human health. Objective: In the present study amelioration effect of Emblica officinalis (E. officinalis) extract on ovary in Endosulfan induced Swiss albino mice was studied. Materials and Methods: Three groups were formed, each with six mice. The control group of mice received distilled water as drinking water. The second group was exposed to Endosulfan @ 3 mg/kg body weight daily for 6 weeks by Gavage method. The third group was exposed to Endosulfan @ 3 mg/kg body weight daily for 6 weeks followed by 6 weeks exposed to E. officinalis (100 mg/kg body weight) extract. Animals were sacrificed after the schedule treatment. Results and Discussion: Light microscopic observations showed various histopathological deformities in sections of treated ovary and improper development of follicle such as detached ova Original Research Article Sinha and Kumari; BJPR, 11(2): 1-9, 2016; Article no.BJPR.17669 2 and degeneration of follicular cells due to Endosulfan exposure were also found. The present studies also indicate that alterations were found to be significantly ameliorated in E. officinalis extract treated group. {"title":"Formulation Development and Optimization of Ibuprofen Poloxamer Melt Granules Using Hydrophilic Excipients","authors":"B. Gajera, R. Dugar, R. Hydrophilic excipients like xylitol and lactose anhydrous were added to the binary mixture of conventional low melting surfactant Poloxamer 407 and Ibuprofen. Physical mixtures of Ibuprofen and Poloxamer 407 were prepared in ratios of 1:0.25, 1:0.5 and 1:0.75 using a water-jacketed high shear mixer. For each ratio of Ibuprofen and Poloxamer 407, xylitol and lactose anhydrous were added separately at two levels (75 mg and 150 mg) per unit dose containing 200 mg drug. Phase solubility studies revealed linearity in drug solubility enhancement with Poloxamer 407 concentration. In vitro dissolution studies were carried out for drug, physical mixtures (PM) and melt granules (MG) for all ratios in de-ionized water and 0.1 N HCl (pH=1.2). Solid state characterization was performed using Fourier transform infrared spectroscopy (FTIR), modulated differential scanning calorimetry (mDSC) and Original Research Article Gajera et al.; BJPR, 13(6): 1-19, 2016; Article no.BJPR.29048 2 powder X-ray diffraction (PXRD) methodologies. Powder rheology studies were performed conventionally by measuring Carr’s index and Hausner’s ratio. Basic flowability energy values were calculated using a powder rheometer to corroborate flowability data measured by conventional methods. Particle morphological studies were done by Scanning electron microscope (SEM) and Fluid imaging technologies. In-vitro dissolution studies showed approximately 7 fold drug release in water and 19 fold drug release in acidic media for MG 1:0.75 at hydrophilic excipient level of 150 mg compared to that of neat Ibuprofen in respective dissolution media. mDSC and PXRD data confirms crystalline nature of drug in the formulations. FTIR data confirms no interactions between drug and excipients used during processing. Particle morphology analysis confirms absence of rhombic Ibuprofen crystals in formulations.
