However, to date, magnesium oxide (MgO) and its effect on odontogenic differentiation of human dental pulp cells (HDPCs) has not been investigated and the underlying regulatory mechanisms of MgO remain undefined. This study was designed to evaluate the stimulatory effect of different concentrations of MgO on odontogenic differentiation with the expression of extracellular matrix-related proteins (dentin sialoprotein (DSP), dentin matrix protein 1 (DMP-1), dentin sialophosphoprotein (DSPP), and collagen-Type I (COL-I). An aqueous MgO suspension was prepared. HDPCs were cultured with 0.5 mM, 1 mM, 2 mM, 4 mM, 8 mM concentrations of supplemental MgO, 0mM as the negative control group, lignin sulfonic acid sodium salt and xanthan gum as the vehicle control groups. Odontoblastic differentiation was assessed by evaluating expression of odontogenic-related proteins: DSP, DMP-1, DSPP, and COL-I. Statistical analysis was done using Multi-Way Analysis of Variance (MANOVA) with Wilks’ lambda test. 0.5 mM MgO elicited the highest stimulatory effect on odontogenic differentiation with significant upregulation of expression of odontogenic-related proteins (DSP, DMP-1, DSPP, COL-I) compared to the negative control ( P <0.0001). This is the first report revealing that MgO-enriched micro-environment promoted odontogenic differentiation in HDPCs. Magnesium-based biomaterials might provide an innovative therapeutic potential to substantially enhance regeneration of dental tissues. Magnesium (Mg 2+ ) has been considered for its potential ability to accelerate proliferation and differentiation of human osteoblasts. However, to date, magnesium oxide (MgO) and its dentinogenic effects on human dental pulp cells (HDPCs) has not been investigated. This study was designed to evaluate the stimulatory effect of different concentrations of MgO on dentinogenesis of HDPCs. HDPCs were cultured with 0.5 mM, 1 mM, 2 mM, 4 mM, 8 mM concentrations of supplemental MgO, 0 mM as the negative control group, lignin sulfonic acid sodium salt and xanthan gum as the vehicle control groups. Cell attachment efficiency was assessed at 16 h. Proliferation rate was evaluated at 3, 7, 10, 14 days. Both attachment efficiency and proliferation rate were assessed by crystal violet staining. Cell viability was determined by activity of mitochondrial dehydrogenase enzyme. Alkaline phosphatase (ALP) activity was assessed using fluorometric assay at 7, 10, and 14 days. Mineralization of cultures was measured by Alizarin Red staining. Statistical analysis was performed using multi-way ANOVA with Wilks’ lambda test. Higher cell attachment efficiency was shown with 0.5 mM at 16 hours compared to negative control ( P <0.001). Cells with 0.5 mM supplemental MgO showed significantly higher proliferation rates than negative and vehicle controls at 7, 10, and 14 days ( P <0.001). Higher levels of ALP activity and mineralization were also observed in 0.5 mM supplemental MgO at 10, and 14 days ( P <0.001). In conclusion, optimal MgO (0.5 mM) group significantly upregulated HDPCs attachment, proliferation, ALP activity, odontogenic differentiation and mineralization. Magnesium oxide containing biomaterials could be a potential novel material for pulp and dentin repair in regenerative endodontics.","PeriodicalId":92689,"journal":{"name":"Madridge journal of dentistry and oral surgery","volume":" ","pages":""},"PeriodicalIF":0.0,"publicationDate":"2021-11-03","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"46378825","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Materials and methods: A total of 72 two-stage sinus lift were performed in 54 patients during 2007 to 2010. The autologous bone was harvested from the mandibular ramus and mixed with the β-tricalcium phosphate Poresorb® TCP sized 1-2 mm. The materials were used in a proportion ranged between 1:1 and 1:3. After the healing period a total of 119 implants were placed and 10 samples of the regenerated bone were collected for the histomorphometric analysis. CBCT or panoramic X-rays were performed presurgically, before the implant placement, six months after implant placement and then yearly to evaluate the bone formation and marginal bone loss. The implant success rate was determined using the Albrektsson et al. Criteria. Results: The mean of the residual bone was 4.07 mm ± 1.87 mm. The bone gain in the sinus was 11.91 mm ± 2.80 mm. The implant success rate was 94.95%. The histomorphometric measurements on the biopsies showed a bone area mean of 39.7 ± 9.71%. The residual allograft area was 16.21 ± 8.78%. The connective tissue was 44.16 ± 5.85%. Conclusion: Within the limit of this study, the osteoconductive β-tricalcium phosphate associated with autologous bone is a viable grafting material for sinus lift procedures. The use of composite grafts can help to reduce the morbidity and aggressivity of the bone harvesting. It is characterized by a paucity of clinical symptomatology and can reach large sizes before final diagnosis. Degenerative cystic changes and haemorrhagic areas are typical, and the most common clinical manifestation is a cystic pancreatic tumour, palpable mass and uncharacteristic abdominal pain. Although resection of the tumour provides a 5-year survival rate of almost 90%, local recurrence or distant metastases can occur in a significant number of patients. Patients with solid pseudopapillary cystic tumours have a much better prognosis, therefore it is important to distinguish it from other pancreatic neoplasms.","PeriodicalId":92689,"journal":{"name":"Madridge journal of dentistry and oral surgery","volume":"2 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2019-07-18","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"89971643","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

They are relatively rare and have a particular tropism for the nail system. Pain is the master symptom but the diagnosis confirmation is based on histology. The use of nail plate and intraoperative dermoscopy facilitates the diagnosis and helps in the localization and demarcation of the tumor. Complete surgical excision is the curative treatment of choice for these tumors. The transungual method is safe and effective, allowing better visualization, easy exploration, and minimal long-term complications. We report a case of glomus tumor developed in the matrix dermis, and treated transungually with partial proximal nail avulsion. Through this case, we emphasize the pre and intraoperative dermoscopic features, and we detail operative procedure.","PeriodicalId":92689,"journal":{"name":"Madridge journal of dentistry and oral surgery","volume":"23 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2019-04-29","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"85098608","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
