In October 2010, following widespread human disease within the Busoga sub-region of Eastern Uganda the Ministry of Health sought to verify the cause. Tungal extraction was therefore performed to provide specimens for diagnosis. Aim: To identify the organisms enucleated from the feet of residents in two affected districts. Method: The formalin-preserved enucleate was macroscopically described, processed and embedded in paraffin wax. Sections four micrometers thick were then stained with haematoxylin and eosin and microscopically examined. Results: Histology showed cystic bodies with internal structures. At the periphery a multi-layered cuticle overlay a stratum of hypodermal cells. At the centre, distended globular sections lined by columnar cells characteristic of digestive epithelia had speckled content representing ingested human blood. Eccentric bipolar sections had convoluted microvillous epithelia typical of filtration-excretory surfaces. Eosinophilic rings formed sub-cuticular chains and central clusters, describing tracheal routes. Numerous eosinophilic anisocytic spheres were enclosed in circular sections lined by cuboid cells characteristic of ovarian epithelia. Fat globules and striated muscle were noted; the head, thorax and terminal abdomen were not seen. Conclusion: The structures described were distinct from those observed in cutaneous helminthiasis, cutaneous myiasis and acariasis. The organisms were thus reported as tissue-embedding haematophagous oviparous arthropods of the genus Tunga. We examined the use of a self-contained, vacuum-assisted biopsy system and a novel muscle freezing technique to improve, simplify, and standardize human muscle biopsy collection and cryopreservation in older adults. The VACORA vacuum-assisted biopsy system was deployed in muscle biopsies of 12 individuals ranging in age from 57 to 80 years. This office-based approach was well tolerated as it is minimally invasive, uses only local anesthetic, and has a quick recovery. To maximize biopsy sample quality and reproducibility, we developed a novel muscle sample freezing protocol. Fresh muscle biopsy samples were placed into readily available tissue cassettes followed by direct freezing in liquid nitrogen. After this modified snap freezing protocol, frozen muscle samples were enrobed in embedding medium for cryosectioning. We examined the effect of this freezing approach in histological sections of rodent and human muscle samples. The VACORA Biopsy System provided as many as four skeletal muscle core samples from a single biopsy site. Biopsy samples from 12 older adults weighed an average of 147.5 ± 11 mg each and had a consistent size and shape. There were no complications, and the residual scar is less than 10 mm. The freezing method using standard tissue cassettes with direct freezing in liquid nitrogen yielded high quality cryopreserved muscle tissue suitable for histological analysis without the need for isopentane and with little to no freeze-thaw damage. This results in neuropathic pain with or without neurogenic inflammation which is a major health problem regularly seen in trigeminal neuralgia. Activation of the trigeminal system results in the release of vasoactive neuropeptides substance P and Calcitonin Gene-related Peptide (CGRP) which contribute to nociception, pain and neurogenic inflammation in injured tissues.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Aim: &lt;/strong&gt;To study the alterations in the neuronal neuropeptides expressions in models of tissue injury associated with either nerve injury or with inflammation and to determine if denervation would alter the neuronal response to inflammation.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Material and methods: &lt;/strong&gt;Experiments were performed on rat mandibles to produce three models. Firstly, denervation model by sectioning one of the mandibular nerve branches (inferior alveolar nerve). Secondarily, inflammation model by intra-gingival injection of lipopolysaccharide (LPS). Thirdly, combined denervation and inflammation model by sectioning the nerve with subsequent LPS injection. The animals were sacrificed seven days postoperative. Trigeminal ganglia on the operated sides were processed for &lt;i&gt;in situ&lt;/i&gt; hybridisation for neuropeptides; substance P and CGRP mRNAs. Images were analysed for morphological and morphometric analysis using Image J software.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Results: &lt;/strong&gt;substance P and CGRP mRNAs were expressed in small and medium-size primary afferent neurons in the mandibular division of the trigeminal ganglia. Both the denervation and the inflammation models showed alteration in neuropeptides expression in the sensory primary afferent neurons innervating the affected mandibular tissues. While, denervation resulted in a significant (substance P=P&lt;0.04, CGRP=P&lt;0.01) downregulation contrarily, inflammation resulted in a significant (P&lt;0.001) upregulation of neuropeptides' mRNAs. Interestingly, denervation prior to induction of inflammation resulted in insignificant changes in neuropeptides levels. There was a strong correlation (Pearson Correlation=0.8) between substance P and CGRP expression.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Conclusion: &lt;/strong&gt;We show that tissue damage associated with nerve injury or inflammation results in alteration of neuropeptides levels in the innervating primary afferent neurons. Tissue destruction associated with chronic inflammatory condition such as arthritis and periodontitis are believed to be due to the production of neuromodulators causing neurogenic inflammation. Here we show that denervation abolishes the neuronal response to inflammation. Therefore, tissues denervation could relieve neurogenic inflammation associated with chronic disorders through regulation of neuronal neuropeptide production. It is very important to find the etiologic factors of oral SCC and its early diagnosis. Copper is one of the most useful metals that have been used by men since ancient time. The level of copper increases when the cancerous changes occur in the body, saliva analysis can be used to discover various illnesses because of its non-invasive methods. In addition to biopsy, an alternative method for examining oral lesions is exfoliative cytology. The aim of this study was to determine the salivary copper level and cytomorphologic changes of oral mucosa among three study groups.Materials and Methods: In this cross-sectional study, 15 individuals with oral SCC, 15 workers exposed to copper and 15 healthy individuals were included. Saliva samples were collected and analyzed by Atomic Absorption Spectrophotometer. The exfoliative smears were prepared by brush biopsy and stained by Papanicolaou and AgNOR staining methods. Data were analyzed by One-way ANOVA and Kruskal-Wallis test. P value <0.05 was considered significant.Result: The results showed a significant difference in mean salivary copper (P=0.008), cytomorphology of oral mucosa and AgNOR among the three groups (P<0.001).Conclusion: Occupational exposure to copper increases the salivary levels of this element and causes changes in mucosal cells. The present study was conducted to assess the ultrastructural changes of RBCs, WBCs and platelets in SLE and to corelate these changes with the disease activity. Comet assay and 8- hydroxydeoguanosine (8-OHdG) were used to confirm cellular dysfunction. Ninety subjects were recruited and equally divided into 3 groups: Group Ι; SLE (normal CBC), Group ΙΙ; SLE (abnormal CBC) and Group ΙΙΙ; healthy control. Disease activity was evaluated by systemic lupus erythematosus disease activity index score (SLEDAI). Ultrastructure examination of blood cells were done by electron microscope, DNA damage was assessed by Comet assay and serum 8-OHdG levels. CBC and serological tests including serum C3, C4, ANA and anti-dsDNA were evaluated. There was statistically significant negative correlation between RBCs and WBCs elements with SLEDAI score in Group II. There was significant statistical difference in RBCs and WBCs cell membrane defects by electron microscope between Group I and Group II. There was no statistically significant correlation between blood cell membrane defects and SLEDAI score in both Group I and Group II. There was a significant increase in percentage of tail DNA damage (p<0.05) in Comet assay and serum 8-OHdG levels in Group I and Group II. In conclusion, there are ultrastructural changes in blood cells in SLE that could play a crucial role in the thrombotic and inflammatory effects of blood cells. In our country, there are cases of women with inflammatory disease of reproductive organ too. Although the researches on ovarian mast cells have been common so far, the researches on mesovarium mast cells, which have good blood and nerve supply, are pretty uncommon. The researches on cold stress induced changes in mesovarium mast cells’ number, size and structure in relation with menstrual cycle hasn’t been done yet in our country. In our study, we have chosen 16 rats in control group and 48 rats in cold stress group, a total of 64 rats. The cold stress group was divided again into 3 groups (16:16:16) and underwent daily cold stress (refrigerator-15°C) from 8:00 am-11:00 am for 7, 14, 21 days in order to generate cold stress model. The number, shape, structural changes, size, perimeter, length and width of the mesovarium mast cell were determined. In the prodromal phase, symptoms such as depression, social withdrawal and mutism occur during about 7 years, till an acute manifestion with symptoms as hallucinations, illusions and paranoia appears [1]. Since the proposal of the dopamine hypothesis, a multi-neurotransmitter system in different brain regions has been suggested. An increased dopamine release, via the dopaminergic D2 receptor, and an augmented serotonin release via the serotonergic 5-HT2A receptor occur in in the involved brain areas, e.g. ventral tegmental area. In preclinical studies, ketamine, an antagonist of the NMDA (N-methyl-D-aspartate) receptor can cause schizophrenia-like symptoms, which can be ameliorated with 5-HT2A antagonists, but not by first-generation antipsychotic drugs which block D2 receptors. The search for susceptibility genes has been developed, and some risk genes are known [2,3]. In most cases, schizophrenic patients are treated with first-generation antipsychotic drugs and more often with newer antipsychotic drugs which block, with different affinities, the dopaminergic D2 receptor and the serotonergic 5-HT2A receptor [4]. A variety of neoplasms have been discovered to have genomic alterations (GAs) and loss of immunohistochemical (IHC) expression of chromatin remodelers <i>ARID1A (BAF250A), SMARCA2 (BRM), SMARCA4 (BRG1),</i> and <i>SMARCB1 (INI1). SMARCA1 (SNF2L)</i> is another member of the chromatin remodelers, and has not yet been studied in neoplasia. As <i>SMARCA1</i> is located on chromosome X, could be potentially inactivated by a single hit. We aimed to evaluate GAs and protein expression of <i>SMARCA1</i> in soft tissue tumors.</p><p><strong>Method: </strong>The publically available cBioPortal.32e34 platform was queried to analyze data on soft tissue tumors from The Cancer Genome Atlas project (TCGA) related to <i>SMARCA1</i> GAs. Our institutional archives were queried to collect 26 cases of soft tissue tumors including 10 undifferentiated sarcomas, 5 leiomyosarcomas, 6 liposarcomas, and 5 malignant peripheral sheath tumors (MPNST). IHC for SMARCA1 with an SNF 2C4 monoclonal antibody was performed on whole tissue sections.</p><p><strong>Results: </strong><i>SMARCA1</i> GAs were present in 8/261 soft tissue sarcomas (3%) in the TCGA dataset. Leiomyosarcomas had most common <i>SMARCA1</i> GAs in 6/99 cases. <i>SMARCA1</i> deletions existed in 1/56 dedifferentiated liposarcomas and 1/48 undifferentiated sarcomas. No <i>SMARCA1</i> GAs occurred in other sarcoma subtypes. SMARCA1 IHC was studied in the sarcoma subtypes with potential <i>SMARCA1</i> alterations in our institutional cases. SMARCA1 nuclear expression was lost in 3/10 cases (30%) of undifferentiated sarcoma, and 2/5 cases of MPNST (40%). SMARCA1 expression was intact in all cases of leiomyosarcoma and liposarcoma.</p><p><strong>Conclusion: </strong>This is the first study to demonstrate loss of expression of SMARCA1 in soft tissue sarcomas subtypes, including undifferentiated sarcoma.
