Regulatory T (Treg) cells may participate in mediating a suppressive microenvironment in tumor tissue and contribute to tumor evasion from immune response. Specifically, some studies have indicated that Treg cell infiltration in malignant ascites and peripheral blood has been associated with poor clinical outcome. \u0000 Methods: The number of CD4+, CD25+, forkhead box p3+ (Foxp3+) lymphocytes was assessed via flow cytometry in ascites and peripheral blood from 24 women with epithelial ovarian cancer (EOC). Overall survival was associated with Treg levels. \u0000 Results: The study indicated an increase in the Treg cell subset from peripheral blood of EOC patients compared with those in healthy controls. Treg infiltration in ascites was higher than in blood into EOC patients. The patients with high infiltration of Treg in blood revealed less overall survival compared to a counterpart with normal levels of Treg. Recently, it has been demonstrated that malignant cells could build upon inflammation and detour it to their own profit, in reprogramming T helpers lymphocytes on an immunosuppressive mode by interacting with checkpoint proteins pathways like PD1/PD-L1 [7]. Therefore tumor microenvironment elements have been more closely investigated especially pro-inflammatory cytokines. Among them, the interleukin 17 family members, that include IL-17A secreted by Th17 lymphocytes, have shown interesting properties though conflicting [8]. While some authors have reported anti-tumor qualities, other teams like ours have observed proliferation, invasion and chemotherapy resistance enhancing effects [9-11]. Knowledge in the fields of immunology is rapidly improving, particularly in oncology, but not only, and almost every medical specialty has benefited of these discoveries in the past 20 years. In their quest for progress, physicians and researchers must remember that the immune system is multifaceted, having positive effects and their opposite at the same time, balanced by complex mechanisms. Gerli","doi":"10.15406/MOJI.2016.04.00136","DOIUrl":"https://doi.org/10.15406/MOJI.2016.04.00136","url":null,"abstract":"In order to investigate the effects of some dietary phytoderivates (Origanum vulgare L. and Rosmarinus officinalis L.) on rabbit immune system, 100 New Zealand mixed-sex rabbits weaned at 30 days of age were split into homogeneous groups submitted to the following dietary treatments: 1) Standard diet (C); 2) C +150 ppm Vit E (E); 3) C +0.2% oregano (O); 4) C +0.2% rosemary (R) and 5) C +0.1% oregano + 0.1% rosemary (OR). Blood samples were drawn from rabbits at 30 and 90 days of age (ten rabbits/diet group at Time 0 and 1, respectively) to investigate the blood lymphocyte subset evolution, the in vitrolymphocyte proliferation (in presence or absence of mitogens) responses and the Immunoglobulin M(IgM) concentration in the supernatants of lymphocyte cultures. A diet effect was observed in the O and OR groups, where the lymphocytes proliferation responses to pokeweed mitogen (PWM; ***P<0.001, in the O group) or the interleukin-2 production (IL-2; ***P<0.001, for both groups) at Time 1 were significantly higher. The IgM levels were systematically higher in the O, OR and E cells culture supernatants (**P<0.01). Age did not affect the rabbit lymphocyte subset evolution nor the in vitrolymphocyte proliferation. One of the approaches to treat APL is differentiation therapy which works by inducing differentiation of leukaemic promonocytes into mature non-replicative cell types that eventually undergo apoptosis. Plasma Membrane-derived Vesicles, (PMVs) are a heterogeneous group of small, membrane-coated vesicles with phospholipid rich particles, membrane receptors as well as other proteins inherent in their parental cells. There is evidence that PMVs are involved in differentiation in the hematopoietic environment in the bone marrow which suggests a possible involvement in leukaemia myeloid development, and possibly monocytic haematopoiesis. This work aimed to show principally whether PMVs can initiate differentiation/stop proliferation in HL-60 cells (promonocytes) and cause their differentiation into macrophages. This way, PMVs can be considered for differentiation therapy. \u0000 \u0000 Methods: PMVs were induced from cells using differential centrifugation. We also employed flow cytometry, ELISA, fluorescent microscopy analysis, SDS-PAGE, western blotting, growth inhibition & differentiation assays, nitroblue tetrazolium assay and cell cycle analysis to conduct our experiments. \u0000 \u0000 Results: HL-60 cells treated with PMVs showed a dose-dependent increase in adherence and increased expression of CD11b and CD14 as a sign of differentiation. Morphological changes were induced in HL-60 as well as phosphorylation of the TGF-?/Smad signaling pathway upon treatment with PMVs. Again, TGF-?1-bearing PMVs reduced the proliferation of HL-60 cells and caused them to exit the cell cycle at G0/G1. It was also shown that PMVs from HL60 or THP-1 cells but not from MCF-7 cells significantly reduce the growth rate of HL-60 cells and the reduction in proliferation was dependent on TGF-?1 delivered by the PMVs. \u0000 \u0000 Conclusion: This work showed that PMVs (isolated from HL-60 and THP-1 cells) carry TGF-β1 on their membrane surface. These TGF-β1-bearing PMVs can modulate the growth rate of HL-60 promonocytes without inducing apoptosis, the cells exiting the cell cycle at G0/G1. In addition, dramatic changes in the morphology of PMV-treated cells were observed as they became increasingly irregular in shape with membrane extensions resembling pseudopodia. The discovery of 1p and 19q chromosomal arms deletion in glial tumour influence the diagnosis and more accurate prediction of chemotherapy response. As a result, an attempt has been made to detect deletion using fluorescent insitu hybridization (FISH) and to determine its prognostic value in a cohort of glial tumour patients from Hyderabad population. The FISH analysis was carried out on 90 FFPE tissue sections by using Vysis LSI 1p36/LSI 1q25 and LSI 19p13/LSI 19q13 dual colored FISH probe sets. Signals were scored from 150-250 non-overlapping, intact nuclei. The analysis for 1p and 19q deletions was observed in (21/35) 60% of oligodendroglyomas which included (8/21) 38.1% of grade II and (13/21) 61.9% of grade III. Isolated 19q deletion was seen in (1/21) 4.7% & lone 1p loss was not observed in oligodendroglyomas. In mixed oligoastrocytomas combined 1p/19q loss was observed in (7/16) 43.75% cases, including one grade II and 6 grade III tumors and 1/16 (6.25%) showed isolated 1p loss & 19q deletion. This disorder was not observed in astrocytomas. The oligodendroglial phenotype was found to be significantly associated with a loss of 1p (p< 0.05), a loss of 19q (p<0.05) and a combined loss of 1p and 19q (p< 0.05). Frontal location of a tumor occurred to be a statistically significant factor unfavorable for prognosis, p<0.05. A study published in the Journal of Clinical Oncology examined the relationship between eosinophils in peripheral blood and the incidence of colorectal cancer [2]. The researchers found that a higher number of peripheral blood eosinophils were associated with a reduced risk of dying from colorectal cancer, especially in patients who never smoked and in males. Although the study could not identify the mechanism to explain this relationship, one plausible theory is that a more active immune system reduces the risk of developing colorectal cancer. Another study published in the journal Modern Pathology in 2016. Prizement et al. [3] looked at how eosinophils might predict outcomes for patients with colorectal cancer. Taghizadeh et al. [4] found no associations between objective allergy markers and cancer in a total population but reported positivity and a high number of eosinophils with association to reduced risk to die of cancer in specific subgroups. Although staging of colorectal cancer typically is based on information about the tumor itself, lymph node involvement, and the presence of metastases (cancer spread to other sites), often two patients with the same staging will have dramatically different outcome. The authors of this study examined whether eosinophils levels in or around a colorectal tumor helps predict outcome.
