Multinucleate cells were frequently observed allowing description of the cell structure and details of the reproduction process. Nuclear staining revealed the position of the multiple polymorphic nuclei between the chloroplast lobes. Ultrastructure of coenocytic cells showed no signs of cleavage of the protoplast in the cytoplasm, although basal bodies were already present within the multinucleate cells. In the further course of the reproduction, biflagellate zoospores were developed that retained their flagella during sporulation. Sites were near vertical or steep, wetted by trickles and drips, but subject to marked changes in extent of wetting and with parts of the gen- eral area liable to desiccation. Water chemical data are given for four sites, where the alga oc- curred in base-rich, low nutrient waters and with fi lterable organic phosphorus the principal fraction of total fi lterable orthophosphate. Chroothece was associated with a range of cyanobac- teria, especially species of Schizothrix, but also colonial, heterocystous and presumed nitrogen- fi xing genera, and calcicolous diatoms (e.g. Epithemia and Mastogloia). Nannochloropsis oculata is a small microalgae with a thick cell wall. In vivo techniques to es- timate cell density, chlorophyll a and neutral lipids are reported. A calibration curve of cell density versus optical density was obtained and validated at 540 nm, under different growth phases. Intracellular neutral lipid storage was evaluated with fl uorometry and epifl uorescent microscopy employing fl uorochrome Nile Red. The addition of 5 % dimethyl sulfoxide en- hanced 12.5 times the fl uorescence signal effi ciency. In situ fl uorescence measurements al- lowed estimating the neutral lipid content (NR-FI). Besides, no signifi cant differences were found in the lipid neutral content between gravimetric and triolein methods. The relationship between NR-FI and chlorophyll fl uorescence signals was used as a neutral lipid accumulation index, which is useful in order to establish the optimum harvesting time.
