Its value is increasing in food industry because of its nutritional and medicinal properties. Besides this, it also contains certain volatile chemicals, phenolic acids, flavonoids and carotenoid like substances which play a major role in antioxidant properties. The composition of honey depends on the plant species visited by the honeybees. The present study was carried out to evaluate the antioxidant activity of Srilankan honey and its role in protecting DNA and erythrocytes from free radical induced oxidative damage. Kalla","doi":"10.3923/AJB.2015.117.124","DOIUrl":"https://doi.org/10.3923/AJB.2015.117.124","url":null,"abstract":"This study was undertaken to monitor the effect of 0.1 mM Mn2+ on the glutathione s-transferase activity (GST) in nicotine treated and untreated spermatozoa samples and to calculate the Michalis-Menten kinetic parameter of the reaction. Semen samples were collected from healthy volunteer donors after semen analysis, sample was subjected for preparation of 10,000 g supernatant. The enzymatic activity has been performed using 1-chloro, 2-4 dinitrobenzene (CDNB) as a substrate. The 0.1 mM Mn2+ supplementation was found to decrease the GST activities significantly (p<0.001). CDNB-GSH conjugate formation were almost significantly (p<0.05) lowered down with the addition of 0.1 mM Mn2+ to nicotine treated samples. The substrate affinity constant (km) of the reaction was found to be 44.44 μM. It remained unchanged throughout the Vmax which was considerably lowered upon Mn2+ addition to the spermatozoal samples. Among them, culture RGSS-09 was adjudged as the best protease producer and was identified as Pseudomonas aeruginosa RGSS-09 from MTCC Chandigarh. The isolate showed maximum protease production (1256 U mLG) with 2.5 g LG biomass production in the presence of wheat bran and yeast extract at 37°C pH 6.5 within 48 h of incubation. Pseudomonas aeruginosa RGSS-09 also showed enhanced activity and stability in presence of Ca, Mg, Tween-20, Tween-40, triton-X-100 and SDS at 5 and 10 mM concentration. The enzyme was also stable in the presence of n-dodecane, isooctane, n-decane, xylene, toluene, n-haxane, n-butanol and cyclohexane, respectively. The presence of benzene, methanol and ethanol marginally reduced the protease stability, respectively. The enzyme was showed it 100% activity at 75°C and pH 7.5 with 110 and 100% stability at 70°C and pH 7.0, respectively. The purity of the enzyme was confirmed by the presence of a single band on SDS-PAGE and its molecular weight was approximately 42 kDa. The 100 and 200 mg kg-1 b.wt. of Camellia sinensis and 100 mg kg-1 b.wt. Silymarin was administered orally to separate groups of rats for 7 days prior to CCl4 administration. A significant decrease (p<0.05) was observed in both the groups treated with 100 and 200 mg kg-1 b.wt. of the leaf extract and Silymarin on the levels of the enzymes and non-enzyme markers of tissue damage and lipid peroxidation, with no significant changes in the relative organ weight of the treated groups.
