Therefore, retroviral vectors containing the human interleukin (IL)-2 or IL-4 gene have been constructed to transduce human melanoma cells to explore whether their immunogenicity can be increased both in vitro and in vivo. Our preliminary results indicate that retroviral vectors can efficiently transduce the IL-2 and or IL-4 gene into melanoma clones, inducing production of either cytokine in the range of 0.5-2 ng/ml/10(5) cells in 48-72 h. In both ovarian and lung tumor lines, but more markedly in the latter, subcutaneous (s.c.) tumor progression of the IFN-gamma-producing cells was profoundly suppressed in the normal syngeneic as well as in athymic nude mice. In addition, experimental metastasis via the tail vein of the IFN-gamma producers was also suppressed. Lung tumor suppression was abolished by X-irradiation of the syngeneic mice or by the administration of antiasialoganglioside GM1 antibodies into the nude mice. These results suggest that tumor suppression is due to the effect of the tumor-derived IFN-gamma on the host antitumor mechanisms including natural killer cells. A-NK cells have also been shown to exert antimetastatic therapy in animal models and in the clinic. Transfection of genes for cytokines or possibly other molecules has the potential to improve the therapeutic potency and efficacy of the effector cells. Gene transfection to induce autocrine production of IL-2 and/or other cytokines is expected to augment their antimetastatic activities, while avoiding toxicity from systemic administration of high doses of cytokines. Here we discuss some of the cytokines used in gene transfer, the importance of MHC class I expression on tumor cells used as vaccines, the relevance of experimental tumor systems to human tumors, and the use of live versus irradiated vaccines. Possible methods for effective gene transfer and possible protocols for immunotherapy studies are presented. The ability of clones releasing IL-2, IL-4 or IFN-gamma to form tumors after s.c. challenge was compared to the TS/A parental cells (TS/A-pc) and to cells transduced with the neomycin resistance gene alone. Cytokine-gene-transduced clones activated a strong inflammatory reaction. The elicited by IL-2 and IL-4-gene-transduced cells efficiently led to tumor rejection. This reaction depended on the activation of several cell mechanisms, those classed as nonspecific being predominant. The repertoire of reactive leukocytes recruited in the reaction varies as a function of the secreted cytokine. Experiments with multiple cytokines have demonstrated that an array of different host effector cells can be recruited by different cytokines in vivo, but more studies are necessary to distinguish cytokine-specific effects from as yet uncharacterized influences of different tumor models. A technically feasible clinical application of this approach is to be seen in the generation of vaccines by introducing such immunostimulatory genes into cancer cells and boosting systemic immune reactions against the unmodified cells. These synthetic agents consist of two covalently linked functional components: a DNA-binding domain and a domain that is a ligand for a cell surface receptor. Recognition of the ligand by its appropriate cell surface receptor allows cellular internalization of the complex via receptor-mediated endocytosis. Since conjugates are limited because they lack a specific mechanism to accomplish cell vesicle escape, and since adenoviruses possess such a mechanism, we derived conjugates that incorporate adenovirus into their functional design. The adenovirus-polylysine conjugates mediated high-efficiency gene transfer. In this configuration, the viral moiety functions both as an endosomolysis agent and also as the ligand domain of the conjugate. To determine whether the adenovirus component of the conjugate could function exclusively in the former capacity, combination conjugates were constructed containing an alternative ligand in conjugation with an incorporated adenovirus. Internalization of these conjugates via the receptor for the alternative ligand in cells lacking adenovirus receptors demonstrated that the adenovirus moiety could function to enhance conjugate entry in this context. This has stemmed from clinical considerations based on the evidence that high-dose interleukin-2 (IL-2) may be effective in a proportion of patients, that foreign genetic material may be successfully inserted into the genome of normal and neoplastic cells, and by extensive studies in experimental models which have shown that cytokine-gene-transduced tumor cells may reveal a diminished or abolished tumorigenicity. Evidence has also been provided that the IL-2 released constitutively by engineered tumor cells may induce the generation of specific cytotoxic T lymphocytes and of an antitumor memory response. Here, we will review the data which have led to the activation of the first pilot vaccination protocols with IL-2 gene-transduced allogeneic cell lines for advanced-stage melanoma and renal cell carcinoma patients.
