In the case of premature rupture of the membranes it is appearing in the vaginal fluid. The level of AFP was measured by the ELISA technique in the vaginal fluid of pregnant women. {"title":"Cell membrane fluidity in blast cells of children with acute leukaemia.","authors":"C Kiss,&nbsp;M Balázs,&nbsp;I Kovács,&nbsp;L Karmazsin","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Plasma membrane fluidity has been investigated by determining steady-state fluorescence polarization (FP) of the apolar stain 1,6-diphenyl-1,3,5,-hexatriene in intact blast cells, separated from peripheral blood and bone marrow of children with various types of acute leukaemia. FP-values of blast cells taken before antileukaemic therapy were compared with FP-values of peripheral blood and bone marrow mononuclear cells separated from patients in complete remission as well as from control patients and from healthy volunteers. Moreover, fluorescence polarization measurements were also performed using blast cells of leukaemic patients on short-term single-drug prednisolone pretreatment. The results have shown that untreated blast cells have significantly lower FP-values than normal mononuclear cells of peripheral blood or bone marrow. No compartment difference has been observed within blast cells, while normal mononuclear cells from peripheral blood have significantly higher FP-values than bone marrow cells. Baseline values of ANP were elevated being 51.7 +/- 21.2 fmol/ml. Volume depletion by withdrawal of 10 ml blood did not cause changes in systolic blood pressure (79.4 +/- 4.3 vs 71.4 +/- 5.6 mmHg) and heart rate (115 +/- 5.2 vs 115 +/- 2.4 l/min). ANP levels in plasma remained unaltered (53.4 +/- 24.9 fmol/ml). Volume load of 10 ml blood transfusion increased central venous pressure by 33 per cent and ANP concentration in the plasma nearly by 30 per cent, while heart rate and blood pressure remained unchanged. Measurements were made prior to and 1 and 2 hours following insulin administration. Before insulin treatment, the sodium level in the red blood cells of the diabetes patients was significantly higher that the control level, while the potassium level was significantly depressed. These differences were not observed 1 and 2 hours following insulin administration. Attention is drawn to the rapid and extensive electrolyte changes in the red blood cells of diabetics in response to insulin. The potassium content of the serum of the diabetics was significantly decreased by insulin administration. No correlation was found between the electrolyte levels and the fasting blood glucose levels. A weak negative correlation (p less than 0.02) was observed between the potassium content of the red blood cells and the duration of the illness. None of the 8 children developed VZV antibodies by the end of the 5-day ACV treatment. All VZV relapses were successfully treated with ACV or Vidarabine, but were stopped only after the appearance of VZV antibodies in the patients' sera. The possible role of ACV treatment in pathogenesis of early VZV relapses could be excluded by comparing the VZV antibody production of patients treated with ACV from the first day of varicella on with the antibody response of those, who received ACV as late as on the 5th day of varicella. Ten premature newborn infants (birth weight 850-1850 g), all ventilated artificially for severe hyaline membrane disease categorized radiologically as stage III-IV, received a single dose of Curosurf (200 mg/kg) via the tracheal tube. This treatment resulted in a rapid improvement of oxygenation, similar to that observed by other teams collaborating in the study and, in an astonishingly fast resolution of the radiological changes. It was measured in parallel with the level of ceruloplasmin and apotransferrin in sera of 35 healthy children and 20 premature babies (gestational age 28-33 weeks). Antioxidant capacity was very low in premature babies with a nadir on the 4th postnatal day. Lipid-peroxidation of bovine-brain homogenate could be inhibited in vitro by the addition of exogen antioxidant. The degree of saturation (expressed as activity product = AP) with respect to this phase was therefore calculated in urines of 36 hypercalciuric children (20 absorptive, 16 renal subtype) with isolated hematuria and 30 healthy controls. The effect of thiazide treatment on the urine saturation and on the evolution of hematuria was also investigated. The results were compared to the urinary calcium excretion (expressed as Ca/cr ratio). Urines of both hypercalciuric groups were saturated on basal conditions (AP above 3.5 x 10-6 mol2/l2; -lgAP below 6.4), the values differed significantly from those of the controls (-lgAP = 6.78 +/- 0.4 in the control-; 6.1 +/- 0.25 in absorptive-, 6.03 +/- 0.34 in renal hypercalciuria; p less than 0.001). Thiazide normalized the activity product in all groups. During thiazide therapy significant decrease in the occurrence of hematuria was noted (p less than 0.001 in both hypercalciuric groups). These data furnish further evidence on the relation of hypercalciuria and postglomerular hematuria.
