This process leads to pronounced alterations in the types and concentrations of metabolites present in the bodily fluids of cancer patients via liquid biopsy approaches. Plasma and serum, owing to their minimally invasive collection, repeatability, and ability to reflect systemic metabolic status, have emerged as optimal sample types for clinical metabolomics. These metabolic changes serve as valuable indicators for inferring disease progression and predicting patient prognosis. Lung cancer, particularly non-small cell lung cancer (NSCLC), with its high global incidence and mortality, represents a critical area where plasma metabolomics can address unmet clinical needs in prognostic prediction and therapeutic stratification. This review focuses primarily on lung cancer. However, the scarcity of studies investigating the prognostic value of metabolite alterations in lung cancer promoted the inclusion of research from other malignancies as well. This review first summarizes the current liquid biopsy metabolomics detection technologies and associated biological materials, followed by an overview of tumor-related metabolic pathway alterations. It then discusses the clinical applications of these principles in prognostic prediction and therapeutic evaluation. The aim is to provide a comprehensive overview and inspire future research directions in this field.</p>","PeriodicalId":73245,"journal":{"name":"Holistic integrative oncology","volume":"5 1","pages":"33"},"PeriodicalIF":0.0,"publicationDate":"2026-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC13109134/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147791072","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This guideline, developed by the Committee of Cancer Genetic Diagnosis of the Chinese Anti-Cancer Association (CACA), provides a comprehensive framework for integrating genetic testing into a holistic approach to cancer care. Addressing key aspects of genetic testing—including patient selection, testing protocols, interpretation of genetic results, and integration into treatment plans—this document is designed to support clinicians in making informed decisions that align with each patient’s unique genetic profile and overall well-being. By emphasizing evidence-based practices and incorporating input from experts in oncology and genetics, these guidelines aim to optimize the effectiveness of genetic testing in cancer care, ultimately enhancing outcomes and quality of life for patients. Our approach promotes the use of genetic insights as a complement to integrative oncology, underscoring the potential of genetic testing to improve holistic management strategies across various cancer types.","PeriodicalId":73245,"journal":{"name":"Holistic integrative oncology","volume":"4 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2025-03-17","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://link.springer.com/content/pdf/10.1007/s44178-025-00149-5.pdf","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147900503","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In a recent landmark study, Wang et al<i>.</i> introduced an innovative internal Z-score based approach to comprehensively characterize the transcription factor (TF) landscape in prostate cancer progression, uncovering distinct TF profiles associated with adenocarcinoma and NEPC lineages. Notably, the study proposes a three-phase model of NEPC transdifferentiation-comprising de-differentiation, dormancy, and re-differentiation-revealing dynamic shifts in TF expression that underpin lineage plasticity and therapeutic resistance. This commentary critically evaluates the methodological advancements, the functional significance of the identified TF signatures, and the broader implications of these findings for developing novel therapeutic strategies. By delineating the molecular events driving the transition from androgen receptor (AR)-dependent adenocarcinoma to treatment-resistant NEPC, this work underscores the potential of targeting early and dormant phases of transdifferentiation to improve patient outcomes.</p>","PeriodicalId":73245,"journal":{"name":"Holistic integrative oncology","volume":"4 1","pages":"65"},"PeriodicalIF":0.0,"publicationDate":"2025-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12552323/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145373291","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
