Shuttleworth","doi":"10.4404/HYSTRIX-00128-2018","DOIUrl":"https://doi.org/10.4404/HYSTRIX-00128-2018","url":null,"abstract":"Viral infections globally threaten wild and captive mammal populations, with surveillance options limited by a lack of non-invasive diagnostics; especially when infection is asymptomatic in nature. We explored the potential for hair samples collected from red (Sciurus vulgaris) and grey (Sciurus carolinensis) squirrels to provide a means of screening for adenovirus (ADV) and squirrelpox virus (SQPV) using evolving polymerase chain reaction (PCR) assays. An initial pilot study phase utilised samples opportunistically harvested from grey squirrels controlled in Gwynedd, United Kingdom (UK). The screening of 319 grey squirrel carcasses revealed 58% spleen ADV DNA qPCR and 69% SQPV antibody enzyme linked immunosorbent assay (ELISA) positives. We developed new nested ADV and SQPV qPCRs and examined tail hair samples from a sub-set of 80 of these 319 sampled squirrels and these assays amplified ADV and SQPV DNA in a higher proportion of animals than the original qPCR (94% and 21% respectively). Tail hair samples obtained from six Cumbrian red squirrels which had died from squirrelpox disease also revealed 100% SQPV and 50% ADV DNA positive by the nested qPCR assays. These findings indicate enhanced sensitivity for the new platform. In order to study the importance of this type of prey, we analysed the species’ diet in five colonies across the Iberian Peninsula using amplicon metabarcoding in order to describe its composition at the species level, and analyse its geographic variability within the peninsula. We identified 138 prey species, belonging to 11 different arthropod orders. Among them, 45 species of spiders were identified, mostly of the orb-web building guild, as consumed by 82 out of 106 studied bats, corresponding to every colony and season sampled. Besides, lepidopterans and dipterans were also consumed in every colony. The crested porcupine and the European badger are semifossorial mammals sharing dens in central Italy. Both species localise their setts mainly in densely vegetated areas, providing them with cover and protection from local predators and poachers. This is particularly evident for the porcupine, widely poached in central and southern Italy, whereas badgers may locally exploit burrows also in open and periurban areas. Wood-cutting and mowing of riparian vegetation surrounding den setts force both porcupines and badgers to leave their burrows. We evaluated the probability of den re-occupancy in the years following the vegetation removal, through intensive camera-trapping at 14 den setts monitored for 9 years. We performed GLMMs to test the annual probability of sett occupancy by the two species after vegetation disturbance events. The probability of re-occupying the burrow by porcupines increased with increasing time from the disturbance cessation. A similar pattern was also observed for the badger, which probability of den occupancy was also negatively correlated with the porcupine presence at the same den, confirming the aggressive behavior of this rodent. We also tested whether, since the first year after vegetation removal, the proportion of years of occupation by porcupines on the total of years has been affected by the disturbance repetition. This effect was found to be significant only for the badger. The crested porcupine, protected by international and national laws, is more sensitive than the badger, protected according to the Italian national law, to vegetation removal. A single disturbance event is sufficient to force it to abandon the den sett, followed by a slow recolonisation with growing vegetation.
