Many features of these antibodies, including their dynamics and neutralization capacities, are still unclear. Understanding the immune response of the host is very important for the development of appropriate treatment methods, vaccines, and epidemiological control strategies. The present study aimed to monitor the change in antibody levels over time in individuals diagnosed with SARSCoV- 2 infections and to determine their neutralization capacity. Material(s) and Method(s): Anti-nucleocapsid and anti-spike antibody titers were measured using different kits on monthly obtained serum samples of patients of patients with SARS-CoV-2 infection. The neutralizing antibodies were evaluated using a microneutralization assay. Result(s): A total of 134 serum samples taken from 43 patients with a mild-moderate disease course were analyzed. Anti-spike antibody positivity was detected on day seven at the earliest and day 334 at the latest following a positive polymerase chain reaction (PCR) test. The mean antibody levels were observed to increase gradually to a peak after three months, and then started to decrease after month six. Anti-nucleocapsid IgM and IgG antibodies were detected alone or in combination. The highest neutralizing antibody titer was 1/80 in the first month, which was seen to drop below 1/10 after four months. Conclusion(s): The combined use of kits for the detection of antibodies against different antigens or testing total antibodies would result in a more accurate and earlier detection of the antibodies that start to emerge on the seventh day and decrease six months after SARS-CoV-2 PCR positivity. In addition, the dramatic decrease in neutralizing antibody titers after four months may be one of the causes of early reinfections. Demographic data, laboratory parameters, and the severity of thorax computed tomography findings were recorded. The relationship between the viral load and these data was compared.Results: The mean age of the patients was 63.4 +/- 9.8 years, mean body mass index (BMI) was 28.6 +/- 5.4 kg/m2, and mean cycle threshold (Ct) values were 21.4 +/- 5.2 cycles. No correlation was found between Ct value and gender, age, and BMI. There was a significant relationship between radiological severity and Ct value, age, and gender. There was a significant correlation between the Ct value and C-reactive protein, leukocyte, neutrophil, lymphocyte, neutrophil-lymphocyte ratio, ferritin, albumin, and calcium levels. In contrast, no significant correlation was found betweenConclusion: The viral load amount shown by PCR during the early period predicts the condition of the patient's lung in the advanced immunological phase. Vaccine studies have been initiated worldwide to end the pandemic, and the CoronaVac (R) vaccine was first administered to healthcare workers at high risk of COVID-19 in Turkey. In our study, we aimed to investigate serum antibody levels after vaccination. Materials and Methods: Volunteer healthcare workers without COVID-19 disease who received two doses of CoronaVac (R) vaccine 28 days apart and were at least 14 days after the last dose of vaccine were included in this study. Assessment of antibodies against Severe acute respiratory syndrome-Coronavirus-2 (SARS-CoV-2) in blood samples from participants was performed using the Elecsys (R) anti-SARS-CoV-2 electrochemiluminescence immunoassay. Samples with a cut-off index (COI) (COI;signal sample/cut-off) <1.0 were considered negative;whereas samples with COI >= 1.0 were deemed positive. Results: A total of 269 healthcare workers, 168 women (62.5%), were included in our study. The mean age of the participants was 37.7 +/- 8.6 (minimum-maximum: 21-62). Antibody levels were positive in 188 (69.9%) of the participants. The median antibody level was 9.2 COI (interquartile ranges=3-34.7). In terms of mean age, the mean age of participants with negative antibodies was higher with a statistically significant difference (p=0.001). The antibody positivity rate of women was higher than that of men (p<0.001). No statistically significant association was found between the time elapsed after vaccination, presence of comorbidities, development of post-vaccine side effects, and antibody levels. It was found that one or more side effects developed in 45.7% of the participants after vaccination. Conclusion: Our study showed that seropositivity developed significantly in healthcare workers after the CoronaVac (R) vaccine.
