One of the most useful methods for suppressing bitterness in solid dosage forms is coating drug particles with a polymer to physically prevent contact between the drug and the taste buds. Because ODFs are extremely thin, the volume available for particle incorporation is limited; therefore, it is necessary to prepare fine coated drug particles with a size smaller than 200 µm, which also helps mitigate particle roughness in the oral cavity. In this study, we designed and prepared ODFs incorporating taste-masked drug particles for easy administration and improved adherence to drug therapy.

Methods: We used quinine hydrochloride (QH) as a model bitter active pharmaceutical ingredient (API), polyvinylacetal diethylaminoacetate (AEA), a pH-dependent polymer, as a coating agent, and hydroxypropyl methylcellulose (HPMC) as the film-forming agent. The ODFs containing taste-masked QH granules and the corresponding granules themselves were evaluated using a taste-sensing system. The disintegration, drug dissolution profiles, and physical properties of the ODFs were also evaluated.

Results: We prepared QH granules with the targeted median diameter (d50) and geometric standard deviation (σg), and the initial release of QH from these granules was less than 5% within 2 min. We also demonstrated, using the same system, that ODFs containing these granules delayed and suppressed the emergence of QH-induced bitterness. The disintegration time of the ODFs was less than 30 s, and their tensile strength remained above the 2 MPa threshold required for handling.

Conclusion: We successfully prepared ODFs that suppressed the bitterness of QH while maintaining a short disintegration time and practical mechanical strength.
","PeriodicalId":524122,"journal":{"name":"Die Pharmazie","volume":"81 8","pages":"53415"},"PeriodicalIF":0.0,"publicationDate":"2026-08-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148893331","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Conventional treatments are mainly based on tyrosinase inhibition; however, many depigmenting agents have limitations, including low stability, poor skin permeation, prolonged treatment durations, and adverse effects such as irritation and dermatitis. In this context, nanotechnology-based delivery systems have emerged as promising alternatives for the topical administration of depigmenting compounds. Therefore, this review discusses recent advances in nanotechnology-based strategies for treating skin hyperpigmentation, with a primary focus on lipid-based nanosystems, including liposomes, nanoemulsions, solid lipid nanoparticles, and nanostructured lipid carriers. A total of 33 studies published between 2000 and 2025 were analyzed. The reviewed studies demonstrated that nanoencapsulation may improve the physicochemical stability, enable controlled release, enhance skin permeation/retention, and increase the therapeutic performance of depigmenting agents while reducing toxicity and adverse effects. In vitro studies also reported enhanced tyrosinase inhibition, high cell viability, and increased accumulation of active compounds in different skin layers. Overall, nanotechnology-based delivery systems represent a promising and versatile approach for improving the efficacy and safety of topical treatments for skin hyperpigmentation.
","PeriodicalId":524122,"journal":{"name":"Die Pharmazie","volume":"81 8","pages":"52238"},"PeriodicalIF":0.0,"publicationDate":"2026-08-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148893362","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The objective of this study was to develop a simple quantitative method using the Child-Pugh classification scoring system to predict the area under the curve (AUC) of drugs in subjects with mild, moderate, and severe hepatic impairment.

Methods: In this study, from healthy subjects, the AUC of 64, 76, and 51 drugs in subjects with mild, moderate, and severe hepatic impairment, respectively, were predicted. The drugs used in this analysis are those that are extensively metabolized in the liver by the cytochrome P-450 system (phase I metabolism). Child-Pugh scores were used to classify varying degrees of hepatic impairment. The scores <5 (no hepatic impairment), and 5, 7, and 10 are the scores from where the Child-Pugh score begins for mild, moderate, and severe hepatic impairment, respectively. In this analysis, a Child-Pugh score of 4 was assigned to healthy subjects. The AUC of a drug in subjects with mild hepatic impairment was predicted as follows: AUC in healthy subjects × (5/4)^0.75, where 5 is the Child-Pugh score in subjects with mild hepatic impairment, and the exponent 0.75 is the exponent of basal metabolic rate or clearance. A similar approach for the prediction of the AUC of drugs was taken in subjects with moderate and severe hepatic impairment. The predicted AUC values in subjects with mild, moderate, and severe hepatic impairment were compared with the observed values obtained from dedicated clinical trials.

Results: In subjects with mild, moderate, and severe hepatic impairment 96.9%, 97.4%, and 82.4% drugs were within 0.5-2-fold prediction error, respectively. No drug was predicted with >2-fold prediction error.

Conclusions: Overall, the results of the study indicated that the proposed method, in its predictive performance, was fairly accurate for the prediction of the AUC of a drug in subjects with varying degrees of hepatic impairment. The proposed method will be helpful in the selection of appropriate dose(s) of drugs in subjects with varying degrees of hepatic impairment for clinical trials or, in some cases, in clinical settings (especially, in mild or moderate hepatic impairment).
","PeriodicalId":524122,"journal":{"name":"Die Pharmazie","volume":"81 7","pages":"52625"},"PeriodicalIF":0.0,"publicationDate":"2026-07-22","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148623714","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This disease process is primarily driven by inflammation and oxidative stress (OS), leading to excessive deposition of the extracellular matrix (ECM). Current treatments provide limited efficacy and are associated with significant side effects.
