These disorders are characterized by progressive protein aggregation and cellular dysfunction, yet no effective therapies exist. Emerging evidence indicates that microbial communities influence host proteostasis, giving rise to the concept of the microbiota-proteostasis axis. Microbes and their products can modulate host proteins by engaging with host proteostasis. As such, microbial dysbiosis has been linked to proteostasis disruption through the production of metabolites, extracellular vesicles, functional amyloids, and toxins or effector proteins capable of seeding or destabilizing host proteins associated with PCDs. These microbial factors have been shown in experimental models to converge on key proteostasis pathways, including protein synthesis, folding and clearance, thereby potentially reducing cellular buffering capacity and lowering the threshold for proteotoxic collapse. In parallel, microbiota-driven influence on inflammatory responses and immune signalling further amplifies systemic proteostasis disruption. In this review, we synthesize emerging evidence defining the microbiota-proteostasis axis and highlight how microbial factors influence host proteostasis. Importantly, these host-microbe interactions often precede neurodegeneration, suggesting potential for early detection and intervention. Together, these insights support targeting the microbiota as a potential strategy to enhance proteostasis and delay or prevent neurodegenerative disease. Yet many proteins can populate multiple states, including native conformations, liquid-like condensed assemblies, and aggregated states, reflecting the metastability of the proteome. As a framework for understanding how cells preserve proteome function under such conditions, we discuss protein rheostasis as the system that regulates thermodynamic driving forces and kinetic barriers to control the flux between alternative states over time. Framing proteome maintenance in terms of rheostatic control over the multiple states helps rationalize how ageing, stress, and mutations redistribute populations towards condensed and aggregated states by eroding kinetic buffering capacity, and it suggests therapeutic opportunities that restore control by tuning the transitions between metastable states. They are an essential capacity for humans' potential to care for fellow humans and non-human entities. How do such emotions evolve, and what type of caring environments and mechanisms are needed for their development? In this review, we provide insights into how children's kind emotions emerge, develop and are nurtured by caring environments. We begin by reviewing the development of kind emotions across the early years and identify consequences of kind emotions on prosociality and well-being. Next, we summarize research on the mechanisms and functions of caring environments in shaping the development of kind emotions, including caring relationships and practices of care across diverse relationships (e.g. with parents and peers) and layers of the ecosystem (e.g. familial, educational, societal and natural). Finally, we provide recommendations for future developmental research in this area. We ask how infants and adults influence each other's expressions of positive affect by examining the timing of transitions among dyadic states defined by the presence and absence of each partner's smiles. Fifty-eight infants interacted with their mothers, fathers and a stranger at 4 and 8 months. A survival analysis of the time course of infant (and adult partner) smiles and non-smiles indicated that infants initiated smiles and reciprocated smiles at lower rates than their adult partners, yet ended smiles at higher rates. Nevertheless, adult smiles were associated with higher infant smile rates, while infant smiles prolonged adult smiles, suggesting a dyadic path to mutual positivity. Eight-month-olds initiated smiles at higher rates than 4-month-olds when neither partner was smiling, suggesting increasing infant smiling initiative. Partner-specific patterns revealed selective infant responsiveness. Although strangers initiated and sustained smiles at higher rates than mothers, infants were more probably to reciprocate mother than stranger smiles. In the first year of life, infants showed a tendency to down-regulate positive affect expressions across age and context, as well as partner-specific smile reciprocation proclivities and a developmental increase in smile initiation. Perhaps owing to this, play faces are mimicked in social interactions across many species. Despite growing interest, few works have directly compared play face mimicry across species and age groups within the same experimental framework, limiting our understanding of its developmental and evolutionary origins. In this study, we examined play face mimicry in human infants (n = 63), human adults (n = 36) and orangutans (Pongo pygmaeus; n = 6), using a tightly controlled design. We found that both human infants and adults reliably mimicked play faces, however, infants produced these robustly more than adults. Additionally, while adult humans did not produce as many play faces as infants, they produced reliably more smiles in response to the play face stimuli. When considering the orangutans, we found no evidence of play face mimicry, with only one instance observed across the entire experiment. This suggests that while our paradigm reliably induced mimicry across both human groups, our set-up may have been suboptimal to evoke mimicry responses in orangutans. Previous work suggests that self-distancing can reduce negative emotional responses, whereas self-immersion can amplify them. This study quantified how these emotion regulation strategies, applied during exposure to negative and neutral videos, shaped self-rated emotional experience across valence, arousal and intensity dimensions, influenced the free production of emotion-descriptive labels and altered measures of respiratory and cardiovascular physiology. A total of 45 participants took part in a task during which they were prompted to simply watch, or to self-distance from or self-immerse in, the emotional content of negative or neutral videos, which were presented in 12, 30-s blocks. Their rated emotional experience (valence-arousal-intensity ratings and free emotion labels) was recorded, along with cardiovascular and respiratory activity. Overall, self-distancing and self-immersing respectively attenuated and amplified aspects of subjective emotion experience relative to comparator conditions as predicted. Self-distancing reduced freely generated emotion labels corresponding to fear, but had less impact on labels corresponding to disgust, sadness and anger. The effects of emotional regulation strategies on cardiorespiratory physiology (e.g. heart rate) potentially also differ between individuals. Together, this work validates a framework for dissecting components of induced emotion, highlighting commonalities and variability in experiential and physiological reactivity and effects of regulation. Temperament is known to influence the regulation and expression of emotions. However, how animal temperament affects their vocalizations and particularly their vocal expressions of emotions remains largely unexplored. Sociability is often measured as the behavioural reactivity to social separation and is a temperament trait intrinsically linked to emotional reactivity. Most social species respond to such challenging situations using contact calls. Here, we investigated whether the acoustic structure of these calls reflects sociability, emotions or both. Among 152 recorded female lambs, 42 belonged to two diverging sheep lines selected for high or low sociability. High bleats were recorded both in isolation (social challenge, all lambs) and before receiving a food treat (non-social challenge, selected lambs) to investigate the link between vocalizations, emotions and heritable sociability. The acoustic features of isolation bleats differed between lines, which was not the case for pre-food treat bleats. However, surprisingly, the genetic selection index and social behaviour explained the structure of isolation bleats better than emotional arousal per se. Last, encoding of individuality in isolation bleats was impaired by the genetic selection. Our findings show that selecting for sociable animals affects vocal signatures in calls produced during a social challenge, leading us to hypothesize a socio-acoustic co-selection. They are thought to emerge in the first years of life, with children's developing cognitive capacities and their growing appreciation of social norms and rules. Recent years have seen a rapid increase in research on self-conscious emotions in early life, supported by advanced methods including manual and automated micro-coding of non-verbal behaviour, depth sensor imaging and physiological measures. In this narrative review, we summarize this emerging evidence from developmental science, which suggests that self-conscious emotions may be observed earlier than previously thought-within the first months and years of life-and that they seem to serve both motivational and communicative functions from early on. Specifically, they motivate socially appropriate behaviours, such as helping others, while communicating adherence to social norms and rules. Further, we highlight current limitations in the field and recommend more experimental and longitudinal studies especially in infancy and toddlerhood, the integration of multi-modal data and the use of naturalistic tasks to advance our understanding of self-conscious emotions in early development.
