Extracellular vesicles (EVs) have emerged as promising biomarker carriers for brain disorders due to their ability to cross the blood-brain barrier, yet their clinical translation has been hindered by complex isolation and detection requirements. Here, we develop a novel wash-free analytical platform leveraging liposome-EV fusion mediated by membrane lipid fluidity and integrated with Förster resonance energy transfer (FRET) technology. This innovative approach enables direct, rapid quantification of both EV concentration and EV-encapsulated miRNA in native biofluids without requiring prior EV separation, overcoming key limitations of conventional methods. When applied to clinical plasma samples, our method demonstrated high sensitivity with detection limits of 4.272 × 1011 particle/mL for EVs and 0.779 nM for miRNA. While no significant difference in total EV concentration was observed between ischemic stroke patients and hypertensive controls, EV-derived miRNA-21 levels were markedly elevated in patient samples, showing exceptional diagnostic performance (AUC = 0.915). This fusion-mediated FRET platform represents a significant advancement in EV-based diagnostics, offering a rapid, sensitive tool with substantial potential for point-of-care stroke diagnosis and timely intervention.","PeriodicalId":517965,"journal":{"name":"Biomedical Analysis","volume":"2 4","pages":"179-185"},"PeriodicalIF":0.0,"publicationDate":"2025-12-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147914917","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This study explored the underlying mechanisms of Tripterygium wilfordii (TWH) through network pharmacology, molecular docking, and experimental validation. Active compounds were identified from the TCMSP database using oral bioavailability ≥ 30 % and drug-likeness > 0.18, while osteoporosis-related targets were collected from GeneCards, OMIM, and TTD. Key targets were screened through protein–protein interaction analysis, and molecular docking assessed binding affinities. In vitro assays using mouse bone marrow-derived macrophages evaluated cytocompatibility, osteoclast inhibition, and changes in gene and protein expression. Eighteen active compounds and 121 putative targets were identified, with kaempferol and triptolide emerging as core constituents. Thirty-seven overlapping targets were enriched in IL-17, TNF, and estrogen pathways, and PTGS2, STAT3, and ESR1 were identified as key nodes. Molecular docking revealed strong binding affinities between major compounds and core proteins. TWH extract inhibited osteoclast formation dose-dependently and significantly downregulated PTGS2 and p-STAT3 expression at mRNA and protein levels. These findings suggest that TWH exerts anti-osteoporotic effects by modulating inflammation-related and estrogen-mediated signaling pathways, supporting its potential as a multi-targeted therapeutic candidate for osteoporosis.","PeriodicalId":517965,"journal":{"name":"Biomedical Analysis","volume":"2 3","pages":"89-99"},"PeriodicalIF":0.0,"publicationDate":"2025-09-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147897713","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The adsorbent material was prepared by mechanically integrating three key components: Fe₃O₄ nanoparticles for magnetic separation, hydroxylated multiwalled carbon nanotubes (MWCNTs-OH) for enhanced adsorption capacity, and citric acid (CA) serving as an intrinsic pH modulator. The synthesized material exhibits exceptional properties, including an extensive surface area and rapid magnetic response, facilitating the selective capture of the target analyte. A distinctive feature of this system is its ability to self-regulate the extraction environment to achieve optimal acidity, thereby eliminating manual pH adjustments and significantly simplifying pretreatment procedures. When coupled with high-performance liquid chromatography with ultraviolet detection (HPLC-UV) analysis, the composite demonstrated high extraction efficiency—with extraction recoveries ranging from 57.12 % to 78.56 % for five NSAIDs—and sensitive quantification of NSAIDs in human plasma. Validation studies revealed satisfactory method performance with good linear correlation (R²>0.99), acceptable accuracy, and reproducible precision. Successful application to clinical specimens confirmed the practical utility of this approach. This integrated methodology represents a significant advancement in sample preparation technology, offering an efficient and economical solution for therapeutic drug monitoring, with promising applications in clinical diagnostics and pharmaceutical research. • A one-pot magnetic adsorbent with integrated pH regulation was synthesized. • Citric acid in the composite enables automatic acidification during extraction. • The method enables rapid MSPE of NSAIDs from plasma without pH adjustment. • High extraction efficiency and reproducibility were achieved using HPLC-UV. • The approach simplifies therapeutic drug monitoring of NSAIDs in biofluids.","PeriodicalId":517965,"journal":{"name":"Biomedical Analysis","volume":"2 3","pages":"120-129"},"PeriodicalIF":0.0,"publicationDate":"2025-09-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147905971","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Here, a nanozyme referred to as CuS/Fe 3 O 4 @BC has been developed by integrating copper sulfide (CuS) and iron oxide (Fe 3 O 4 ) onto a biochar (BC) matrix. This material exhibits biomimetic hierarchical porosity and enhanced peroxidase-like activity, making it specifically tailored for biomedical analysis applications. This multifunctional platform achieves ultrasensitive mercury(II) detection in biological fluids through structural optimization of mass transfer kinetics combined with a 29.3-fold enhancement in catalytic activity compared to conventional CuS nanozymes. To bridge laboratory diagnostics with point-of-care applications, the nanozyme is integrated into smartphone-coupled hydrogel spheres, enabling portable colorimetric quantification of Hg 2 + with a detection limit of 0.28 μM.
