All achievements in this area not only are directly related to the possibility of answering the question of how life was created on this planet but also impose great risks on the creation of objects that can be directed to the detriment of humanity or individual groups of people. Since modern technologies make it possible to synthesize the genomes of many organisms (bacteria and viruses) in vitro without complex equipment, only the moral and ethical qualities of the researcher and his foresight and deep study of the consequences of his actions can ensure the correct development of synthetic biology as a new and very promising science, without fear of possible negative consequences.","PeriodicalId":509940,"journal":{"name":"Russian Journal of Genetics","volume":"61 11","pages":"1394-1401"},"PeriodicalIF":0.0,"publicationDate":"2025-11-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147881249","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The LUSC transcriptome data were downloaded from the TCGA and GEO databases. The immune and stromal scores as well as tumor purity of LUSC samples were analyzed. The differentially expressed genes (DEGs) in high vs. low stromal score and high vs. low immune score were respectively screened. The important TME-related genes were screened using univariate/multivariate cox regression, and LASSO regression analyses. The correlation of important TME-related genes and immune microenvironment was analyzed. Finally, the important TME-related genes were validated through GEO dataset and clinical samples. The LUSC samples with low estimate, immune and stromal scores, and high purity score had better prognosis. 1241 and 798 DEGs were respective lyscreened in two groups, with 642 intersection DEGs. Pathway analysis identified cytokine-cytokine receptor interaction as the most significantly correlated pathway. From these intersection genes, STARD6, FGA, TRIM58 and HPR were considered prognosis-correlated important TME-related genes. High expression of STARD6 had a better prognosis, while low expression of FGA, TRIM58 and HPR had a better prognosis. STARD6 was upregulated in tumors, while FGA, TRIM58 and HPR were downregulated in tumors. TME may be related with cancer progression and prognosis of patients in LUSC. Immune and stromal scores-based genes may serve as treatment biomarkers for LUSC.","PeriodicalId":509940,"journal":{"name":"Russian Journal of Genetics","volume":"61 1","pages":"108-123"},"PeriodicalIF":0.0,"publicationDate":"2025-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147894680","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Melatonin (MLT) is a natural hormone secreted by pineal gland associated with circadian rhythm and epilepsy. This study intended to figure out the molecular mechanism of IS therapy by analyzing the influence of CLOCK, SIRT1, HDAC2 and MLT on glucocorticoid receptor α (GRα). In the present study, the results showed that the gene of CLOCK, SIRT1 and HDAC2 were successfully knockdown by small interfering RNAs in GT1-7 cells. In addition, gene knockdown of CLOCK, SIRT1 and HDAC2 significantly inhibited the GRα expression and decreased the GRα concentration, which this inhibitory effect was significantly enhanced by MLT intervention. The similar results found that the expression of BMAL1, C-Cbl, BDNF and TrkB were also markedly decreased by MLT intervention, which is consistent with gene knockdown. Therefore, all data provided that CLOCK, SIRT1 and HDAC2 knockdown synergistic melatonin intervention decreased the concentration of GRα, while MLT intervention strengthened this effect, which might provide a molecular mechanism for treating CRH-induced IS.","PeriodicalId":509940,"journal":{"name":"Russian Journal of Genetics","volume":"58 1","pages":"85-93"},"PeriodicalIF":0.0,"publicationDate":"2022-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147919073","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Dorsomorphin 2HCl (DOR) and AICAR were used to inhibited and activated AMPK expression, respectively. And then, the myoblasts apoptosis and differentiation were established by dexamethasone (DEX) and 2% pregnant mare serum gonadotropin (PMSG) induction. In the present work, DEX succeed promoting the apoptosis of duck myoblasts and the expression of marker genes Caspase 3, LC3 and Caspase 7. Further, the mRNA expression of AMPKα1 and FOXO3 and the protein expression of p-AMPK and FOXO3 significantly increased in the 1000 and 10 000 ng/mL group but AMPKα2 only in 10 000 ng/mL group. However, the positive effects of DEX on apoptosis of duck myoblasts could be inhibited by DOR. We found that the activator of AMPK/FOXO3 pathway could increase the expression of Caspase 3, LC3 and Fas and promoted the G0/G1 phase arrest. On the other hand, the AMPK/FOXO3 signal pathway could also be activated by 2% PMSG. Similarity, the expression increased of MyHC was induced by 2% PMSG could also be inhibited by DOR. Corresponding, the activation of AMPK/FOXO3 signal pathway significantly increased the expression of MyHC. Therefore, our results indicated that the activation of AMPK/FOXO3 signal pathway could promote the duck myoblasts apoptosis and differentiation.","PeriodicalId":509940,"journal":{"name":"Russian Journal of Genetics","volume":"57 1","pages":"97-109"},"PeriodicalIF":0.0,"publicationDate":"2021-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147914260","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
