Its prevalence is on the rise as the population ages worldwide. There are many drawbacks to the current clinical approaches to treatment, and traditional chinese medicine (TCM) has long had a unique advantage in the prevention and treatment of osteoporosis. As research in the field of ferroptosis continues to intensify, the study of TCM has become a hot area in the treatment of the disease through multimethod interventions for ferroptosis. Based on this, this study searched the Cochrane Library, PubMed, Embase, CBM, CNKI, WanFang Data, and VIP databases using osteoporosis, TCM, and ferroptosis as keywords. Up to April 2023, research literature on the relationship between TCM, osteoporosis, and ferroptosis was collected. The results found a strong relationship between the main regulatory mechanisms of ferroptosis and osteoporosis. TCM methods with good therapeutic effects on osteoporosis, such as Zanthoxylum bungeanum, Curcumae longae, Salvia miltiorrhiza, Astragalus membranaceus, Bushen Huanjing Recipe, and external treatment of TCM, can all achieve interventions on ferroptosis under different pathways. It provides a new theoretical basis and research direction for the future treatment of osteoporosis through multiple methods of TCM to intervene in ferroptosis. This study aimed to evaluate the mechanisms of TXD against myocardial ischemia and provide evidence for its subsequent clinical application. Materials and Methods: Active components and mechanisms of action of TXD against myocardial ischemia were predicted and analyzed by network pharmacology and molecular docking. The chemical constituents in TXD were determined by HPLC-MS. The oxidative damage model was established using H2O2, which caused myocardial ischemia damage. The MTT assay and Hoechst 33342 staining were used to evaluate cell viability, while cleaved caspase-3 immunofluorescence staining was used to determine cell apoptosis. The fluorescent probe method detected ROS and intracellular Ca2+, while spectrophotometry was used to measure SOD, MDA, and NO levels in myocardial cells. Western blotting was used to detect the expression levels of ERɑ, PI3K, p-PI3K, AKT, p-AKT, and eNOS in cells. Results: It was found that TXD plays a protective role in myocardial ischemia through the estrogen pathway. The TXD drug-containing plasma exhibited increased cell survival rates and suppressed MDA levels, elevated SOD and NO levels, and significantly suppressed ROS levels as well as intracellular Ca2+ levels. Moreover, the TXD drug-containing plasma pretreated cells had significantly suppressed PI3K, p-PI3K, AKT, and p-AKT expression levels, as well as elevated ERɑ and eNOS expression levels. Conclusion: TXD activates estrogen receptor ERɑ, enhances NO release of cardiomyocytes through the estrogen signaling pathway, improves intracellular Ca2+ overload, oxidative stress injury, and apoptosis, and plays a protective role in myocardial ischemia injury.","PeriodicalId":508089,"journal":{"name":"Pharmacognosy Magazine","volume":"37 4","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-02-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139856619","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Materials and Methods: The T2DM model was established by a high-fat high-sucrose diet (HFSD) for 6 weeks and streptozotocin (STZ) administration. The rats were randomly divided into control, model, metformin (100 mg/kg/day), and T. officinale extracts (1,500 mg/kg/day) treatment groups. Fasting plasma glucose (FPG) levels, 2-h postprandial plasma glucose (2hPPG) levels, and sucrose preference rates (SPR) were ascertained. Enzyme-linked immunosorbent assay (ELISA) was used to quantify the levels of superoxide dismutase (SOD) and malonaldehyde (MDA) in the serum. Finally, the expression levels of UPRmt and mitophagy-related proteins were determined via Western blot analysis. Results: Rats in the model group exhibited significantly higher levels of FPG and 2hPPG but lower SPR than those in the control group. Rats in the Taraxacum group had lower FPG and 2hPPG levels and higher SPR than those of the model group. Moreover, clpP, HSP60, PINK-1, Parkin, P62, Beclin-1, and LC3-II/I proteins were significantly upregulated in the rats of the model group, relative to the control group. UPRmt and mitophagy-related proteins were markedly downregulated in rats of the Taraxacum group, relative to the model group. Conclusion: T2DM rats exhibited certain levels of anhedonia and depression, which were effectively alleviated by T. officinale extracts through the downregulation of UPRmt and mitophagy, and also by conferring protection to mitochondrial function. The adults are the major victims of gliomas, and the men are mostly affected. At present, gliomas are treated with surgical resection followed by radiation and the administration of chemodrugs such as temozolomide. The invasive and high recurrence rate of gliomas often reduces the effectiveness of treatment, which leads to poor recovery. Hence, more potent drugs without side effects on long-term treatment need to be discovered to treat gliomas. Morin is one such promising phytochemical that possesses immense pharmacological properties. It is proven to exert anti-inflammatory, antioxidant, anticancer, antibacterial, antidiabetic, and neuroprotective properties. The current research focuses on examining the growth inhibition and apoptosis-inducing potency of morin against glioma C6 cells. Materials and Methods: Rat C6 glioma cells were treated with different concentrations of morin and analyzed for cytotoxicity using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. The intracellular reactive oxygen species (ROS) production and alteration of mitochondrial membrane potential (MMP) by morin in glioma cells were examined with dichlorodihydrofluorescein diacetate (DCHF-DA) and rhodamine 123 staining. The apoptotic induction in glioma cells was analyzed with dual staining and it was confirmed by quantifying the apoptotic protein using the enzyme-linked immunosorbent assay (ELISA) technique. The anti-inflammatory property of morin was assessed by the inflammatory cytokines tumor necrosis factor-α (TNF-α), nuclear factor-kappa B (NF-κB), cyclooxygenase-2 (COX-2), and interleukin-6 (IL-6) using the ELISA technique. Results: Our MTT results show that morin treatment significantly reduced the cell viability of rat C6 glioma cells. It also significantly increased ROS generation and decreased MMP in rat C6 glioma cells. Morin treatment also effectively increased caspase-3 and caspase-9, the proapoptotic protein Bax, and decreased the antiapoptotic protein Bcl2. The induction of apoptosis by morin in glioma cells was evident with our acridine orange/ethidium bromide (AO/EtBr) staining results. Morin also decreased the inflammatory cytokine levels in rat C6 glioma cells. Conclusion: Our findings have proven that morin significantly induces apoptosis by increasing the levels of apoptotic protein via the generation of ROS. Results: It was shown that the appearance and content varied in RRS, GRS, and WRS. In addition, the method of PLS-DA and LDA can differentiate RRS, GRS, and WRS. Conclusion: RS is diverse, and the appearance and chemical characteristics can be used to differentiate RRS, WRS, and GRS.","PeriodicalId":508089,"journal":{"name":"Pharmacognosy Magazine","volume":"269 14‐18","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-02-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"139799628","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
