Since its first outbreak in China in August 2019, LSD has spread widely across mainland China, posing significant threats to the cattle industry. This study aimed to isolate and identify a clinical strain of LSDV via Vero cells. Skin tissue samples from lump lesions were homogenized and inoculated onto cell cultures. After 7 passages, the inoculated cells exhibited typical cytopathic effects (CPEs). PCR amplification of the LSDV132 gene confirmed the presence of LSDV nucleic acid. In addition, quantitative PCR (qPCR) demonstrated a significant increase in viral copy number over time. Transmission electron microscopy (TEM) revealed typical brick-shaped viral particles. Furthermore, an indirect immunofluorescence assay (IFA) of infected Vero cells exhibiting CPEs produced a positive reaction with antiserum from cattle naturally infected with LSDV. Additionally, nucleotide similarity analysis of 123 LSDV strains revealed a high degree of similarity (98.4%-100%) among different geographic lineages. Nucleotide sequencing and recombination analysis of the LSDV011 gene from LSDV/China/HB01/2020 revealed close similarity to Asian strains and revealed a recombination event. Furthermore, similarity plot analysis confirmed two genomic exchange sites at nucleotide positions 120 and 762 within the LSDV011 gene. Recombination events between 65 Asian LSDV strains and 13 goatpox virus (GTPV) strains have raised safety concerns regarding the use of attenuated goatpox vaccines, highlighting the need for novel and safer LSDV vaccines. In summary, this study successfully isolated a clinical LSDV strain, demonstrating its evolutionary status and providing crucial insights for LSD control in the cattle industry.","PeriodicalId":504399,"journal":{"name":"Animal Diseases","volume":"5 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2025-06-27","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://animaldiseases.biomedcentral.com/counter/pdf/10.1186/s44149-025-00182-x","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147900430","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The mAbs were generated via the use of a KLH-conjugated peptide and purified equine IL-1β protein as separate immunogens. Notably, the generated mAbs (3G8 and 5G3) demonstrated no cross-reactivity with other major inflammatory mediators, including IL-1α, IL-1Ra, TNF-α, and SAA. The IL-1β assay, which is based on the screened mAbs, exhibits a detection range of 200–10,000 pg/mL, meeting clinical detection requirements. The coefficients of variation for the repeatability and reproducibility of the assay were both less than 5%, indicating an acceptable level of variation. Subsequently, 84 equine and 24 asinine serum samples were collected, and the IL-1β concentration was measured with both our assay and a commercial kit in parallel. Our results revealed no significant difference between the in-house and commercial ELISA kits for the detection of IL-1β concentrations in horse sera. Moreover, our ELISA method demonstrated superior sensitivity for IL-1β detection in donkey samples compared to existing commercial assays. These findings suggest that the newly developed ELISA provides a reliable analytical method for detecting IL-1β in both equine and asinine samples.","PeriodicalId":504399,"journal":{"name":"Animal Diseases","volume":"5 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2025-06-18","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://animaldiseases.biomedcentral.com/counter/pdf/10.1186/s44149-025-00175-w","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147898965","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

A recent report of PDCoV infection in Haitian children also highlights potential public health implications. In this study, two monoclonal antibodies (mAbs), 1C2 and 5H5, were generated and showed high specificity for the PDCoV S protein. Both mAbs displayed high-titer neutralizing capabilities, suggesting their potential for passive immunotherapy. Epitope mapping revealed that the mAbs likely recognized conformational epitopes in the S1 subunit domains A and B of the native S protein, thereby blocking the interaction between the S1 receptor-binding domain and the cellular receptor, which could inhibit viral entry into host cells. This study offers new biological tools for PDCoV detection and lays the groundwork for the future development of porcine-specific antibodies for the prevention and treatment of PDCoV in piglets.","PeriodicalId":504399,"journal":{"name":"Animal Diseases","volume":"5 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2025-01-26","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://animaldiseases.biomedcentral.com/counter/pdf/10.1186/s44149-025-00156-z","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147909939","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Here, we report that histone methyltransferase G9a is elevated in the muscle of muscular atrophy model mice and that muscle-specific deficiency of G9a ( Ehmt2 Ckmm− KO) alleviates muscular atrophy in both aged and denervated mice. Moreover, increased nerve-to-myofiber ratios and increased Agrin-Lrp4-MuSK signaling, which maintains NMJ, are found in aged Ehmt2 Ckmm− KO mice. Together, these data suggest that G9a promotes muscular atrophy and disrupts NMJ; thus, inhibiting the G9a level may be a potential therapy for muscular atrophy.","PeriodicalId":504399,"journal":{"name":"Animal Diseases","volume":"4 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-11-05","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://animaldiseases.biomedcentral.com/counter/pdf/10.1186/s44149-024-00147-6","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147906257","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Taurodeoxycholic acid (TDCA) is a metabolite of bile acids and has anti-inflammatory and protective effects on the intestinal tract. However, whether TDCA can alleviate oxidative stress in the intestine is still unclear. Here, we investigated the effects of TDCA on diquat-induced oxidative stress in the jejunum and its mechanism. The results revealed that TDCA increased the concentrations of antioxidant enzymes in the serum, jejunal tissue and intestinal epithelial cells of the mice, as did the expression of tight junction-associated proteins and the Nrf2 protein in the jejunal epithelial tissue and intestinal epithelial cells. We then explored the mechanism of Nrf2 with ML385 (a specific Nrf2 inhibitor). The results showed that after ML385 treatment, the levels of antioxidant enzymes were significantly decreased in the serum, jejunum, and intestinal epithelial tissues of the mice. The expression of tight junction proteins in jejunum epithelial tissues and intestinal epithelial cells was also decreased. In conclusion, our study suggests that TDCA alleviates oxidative stress to improve intestinal barrier function through the Nrf2-mediated signaling pathway. These findings help elucidate the role of TDCA in protecting the intestinal barrier and its mechanism of action, providing insights for the prevention and treatment of intestinal diseases caused by oxidative stress.","PeriodicalId":504399,"journal":{"name":"Animal Diseases","volume":"4 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-09-27","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://link.springer.com/content/pdf/10.1186/s44149-024-00139-6.pdf","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147886774","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, high-fat diets have always been associated with a variety of digestive disorders. This study aimed to investigate the impact of high-fat diets on the gut microbiota and the mechanisms of gut damage in Nyctereutes procyonoides . 16S rRNA sequencing demonstrated that high-fat diets caused diarrhea and intestinal damage through alterations in the gut microbiota: a decrease in the abundance of Firmicutes, an increase in the abundance of Proteobacteria and Actinobacteria , and an increase in the abundance of Enterococcaceae , Escherichia coli-Shigella , Clostridium and Lactobacillus . Subsequently, changes in metabolic pathways, such as amino and fatty acid pathways, were identified by KEGG and COG enrichment analysis, and the TLR4/NF-κB/NLRP3 inflammatory signaling pathway was shown to be activated by high-fat diets. In addition, high-fat diets lead to the accumulation of ROS and MDA and reduce the activity of the antioxidant enzymes GSH-PX and SOD. Correspondingly, the levels of proinflammatory cytokines (IL-6, IL-1β and TNF-α) were significantly increased, and the apoptosis and necrosis signaling pathways of colonic cells were detected, causing a dramatic decrease in the expression of intestinal tight junction proteins (Occludin, E-cadherin, ZO-1 and ZO-2). In conclusion, high-fat diets altered the structure of the Nyctereutes procyonoides gut microbiota community and led to colon damage. This study provides new insights into the intestinal health of Nyctereutes procyonoides . In the present study, we identified the ESX-1 secretion-associated protein EspL, which promotes Mtb survival by inhibiting phagosome maturation and autophagy initiation. EspL knockout decreased Mtb intracellular survival, while EspL overexpression increased bacterial survival by interfering with phagocytosis and autophagy. EspL interacts with ULK1 and promotes its phosphorylation at Ser 757 , leading to the inhibition of autophagy initiation. Additionally, overexpression of EspL reduced antigen presentation and T-cell responses both in vitro and in vivo. Our findings revealed that EspL interferes with autophagy and antigen presentation by suppressing ULK1 activation. These insights provide a novel understanding of Mtb pathogenicity.","PeriodicalId":504399,"journal":{"name":"Animal Diseases","volume":"4 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-06-25","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://animaldiseases.biomedcentral.com/counter/pdf/10.1186/s44149-024-00128-9","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147892849","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
