Genetic assimilation provides an alternative hypothesis to explain adaptation, in which phenotypic plasticity is first triggered by environmental factors, followed by selection on genotypes that reduce the plastic expression of phenotypes. To investigate the evidence of genetic assimilation in a high-altitude dweller, the toad-headed agama Phr ynocephalus vlangalii, we conducted a translocation experiment by moving individuals from high-to low-altitude environments. We then measured their gene expression profiles by transcriptome sequencing in heart, liver and muscle, and compared them to two low-altitude species P. axillaris and P. fors ythii. The results showed that the general expression profile of P. vlangalii was similar to its viviparous relative P. fors ythii, however, the differentially expressed genes in the liver of P. vlangalii showed a distinct pattern compared to both the low-altitude species. In particular, several key genes (FASN, ACAA2 and ECI2) within fatty acid metabolic pathway were no longer differentially expressed in P. valgnalii, suggesting the loss of plasticity for this pathway after translocation. This study provides evidence of genetic assimilation in fatty acid metabolism that may have facilitated the adaptation to high-altitude for P. vlangalii.","PeriodicalId":49236,"journal":{"name":"Asian Herpetological Research","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2021-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"67548037","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Nanorana parkeri is an anuran endemic to the southern Tibetan Plateau where it has an extensive altitudinal range(2850 to 5100 m asl). Low oxygen concentration is one of the main environmental characteristics of the Tibetan Plateau. Hypoxia-inducible factor α subunits(HIF-1α and HIF-2α, encoded by Endothelial PAS domain protein 1(EPAS1)) and associated genes(e.g., vascular endothelial growth factor(VEGF) and Erythropoietin(EPO)) play crucial roles in maintaining oxygen homeostasis. In this study, we compared the expression of HIF-1A, VEGF, EPAS1 and EPO mRNA between two populations of N. parkeri: one population inhabiting the native high altitudes, and the second living in, and being acclimated to, the lower plains(70 m asl). The expression of HIF-1A, VEGF and EPAS1 mRNA in the high altitude population were significantly higher than in the acclimated population, whereas there was no significant difference for EPO between two groups. Our results indicated that gene expression plasticity may make significant contributions to local adaptation of species that have broad altitudinal distributions. In addition, we deepen our understanding of the adaptive potential of this species by evaluating the experiments in the scope of its evolutionary history.","PeriodicalId":49236,"journal":{"name":"Asian Herpetological Research","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2016-03-25","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"67547917","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
