The heat stress coupled with freezing and thawing processes generates reactive oxygen species (ROS), which further reduce post-thaw spermatozoa quality. Application of cysteine as a semen additive have been reported to alleviate such damaging effect in canine, buck, bull, human, boar, and rooster. Therefore, objective of this study was to investigate ameliorating effect of cysteine on summer stress on buffalo spermatozoa. For the experiment, 24 ejaculates with individual progressive motility >70, volume >1mL and sperm concentration 1000 million/mL from four Murrah buffalo bulls were collected and divided into four groups as Control (Group-I), and Group-II, III, and IV (4, 8, and 12 mmol cysteine, respectively) containing 80 million spermatozoa per mL. Semen was filled (French mini straw), sealed and equilibrated at 40⸰C and frozen into LN2. The group containing L-cysteine at 4 mmol concentration had higher significant (p<0.05) difference in post thaw motility, viability, plasma membrane integrity and acrosome integrity, TAC, DNA integrity and lower significant (p<0.05) difference in spermatozoa abnormality, LPO and intracellular ROS. Moreover, cysteine concentration at 8 mmol has no beneficial effect compared to the Control, whereas Group-IV had greater adverse effect. This may be due to its toxicity at this concentration. It is evident from the study that the cysteine molecules at 4 mmol concentration produced more pronounced improvement in the freezability potential of buffalo bull spermatozoa.","PeriodicalId":488871,"journal":{"name":"The Indian Journal of Animal Reproduction","volume":"9 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135400197","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Thirty-one CTVT confirmed dogs presented to University Veterinary Hospitals were categorised into three groups. The group I (n=13) dogs were treated with vincristine sulphate, while group II (n=9) and III (n=9) dogs were treated with vincristine along with ivermectin and cyclophosphamide respectively. Haemato-biochemical evaluations were performed on the day of presentation and at weekly intervals till complete tumour regression. The haemato-biochemical parameters like TEC, Hb, VPRC, TP, BUN, creatinine, albumin, globulin and ALT were within normal range. Significant differenceswere observed in DLC, TC, MCV, AST, ALP and bilirubin levels (p<0.05) during treatment. Various blood dyscrasias like leucocytosis was observed in group I and group II, and thrombocytopenia in group III. Highly elevated AST, ALP and bilirubin were observed in all three groups. But combination therapy with ivermectin was more efficient with fewer adverse effects and decreased treatment duration.","PeriodicalId":488871,"journal":{"name":"The Indian Journal of Animal Reproduction","volume":"156 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135400231","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Buffaloes (n=8) were superstimulated twice using 2x2 design at an interval of 30-35 days using 200 mg and 300 mg FSH divided into four and six tapering doses (each dose of 50 mg) given over 2 and 3 days at 12 hourly intervals. Prostaglandin F2α analogue (500 μg Cloprostenol; i/m) at 72 and 84 hrs after start of superstimulatory treatment. Oocytes were retrieved from > 6 mm follicles at 48 h after the last FSH injection by ovum pick up. Oocyte maturation was assessed by the degree of cumulus cell expansion and by Orcein staining of denuded oocytes. OPU of 60 and 91 follicles resulted in recovery of 38 and 50 cumulus oocyte complexes (COCs) in 2-Day and 3-Day group, respectively. Significantly higher proportion of expanded oocytes was recovered in 3-Day protocol compared to 2-Day protocol (36.0% vs. 13.2%) and higher number of compact COCs were recovered in 2-Day protocol. In 3-Day protocol, significantly higher percentage of oocytes retrieved was at Metaphase-II stage compared to 2-Day protocol (40% vs.15.8%, respectively). The preliminary trial demonstrated that 3-day superstimulatory protocol yielded higher percentage of in vivo matured oocytes than 2-Day protocol in buffaloes.","PeriodicalId":488871,"journal":{"name":"The Indian Journal of Animal Reproduction","volume":"232 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135401826","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Ejaculates were collected by AV method from Sahiwal bulls. X-sperm enrichment was done by percoll density gradient method i.e. 7 layers (70-10%). Centrifugation was done at 750 g (22-24°C) for 15 min. The pellets obtained were diluted in EYC medium. Semen quality was evaluated in fresh semen (Control), in pellet of normal centrifugation (Group I), supernatant of centrifugation in percoll density gradient (Group II) and pellet of centrifugation in percoll density gradient (Group III). To assess the quality of enriched semen pH, mass motility, progressive motility, live spermatozoa %, abnormal spermatozoa %, HOST % and intact acrosome % were evaluated. Number of progressively motile sperms in pellet of X- enriched semen were non-significantly increased and significantly (P<0.05) decreased in supernatant. The abnormal spermatozoa (%) were decreased in G III as compared to G II Live spermatozoa (%) were increased in enriched semen (pellet). Number of Intact sperms decreased significantly (P<0.05) in supernatant of percoll density gradient centrifuged Sahiwal semen. HOST responsive sperms number was not affected after percoll density gradient centrifugation. Thus, the semen quality of X sperm enriched semen by percoll density gradient method (7 layer 70%) was not affected hence it can be used to increase female calves’ birth after A.I.","PeriodicalId":488871,"journal":{"name":"The Indian Journal of Animal Reproduction","volume":"108 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135400238","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
