{"title":"Characterization of a highly diverged mitochondrial ATP synthase F<sub>o</sub> subunit in Trypanosoma brucei.","authors":"Caroline E Dewar, Silke Oeljeklaus, Christoph Wenger, Bettina Warscheid, André Schneider","doi":"10.1016/j.jbc.2022.101829","DOIUrl":"10.1016/j.jbc.2022.101829","url":null,"abstract":"<p><p>The mitochondrial F<sub>1</sub>F<sub>o</sub> ATP synthase of the parasite Trypanosoma brucei has been previously studied in detail. This unusual enzyme switches direction in functionality during the life cycle of the parasite, acting as an ATP synthase in the insect stages, and as an ATPase to generate mitochondrial membrane potential in the mammalian bloodstream stages. Whereas the trypanosome F<sub>1</sub> moiety is relatively highly conserved in structure and composition, the F<sub>o</sub> subcomplex and the peripheral stalk have been shown to be more variable. Interestingly, a core subunit of the latter, the normally conserved subunit b, has been resistant to identification by sequence alignment or biochemical methods. Here, we identified a 17 kDa mitochondrial protein of the inner membrane, Tb927.8.3070, that is essential for normal growth, efficient oxidative phosphorylation, and membrane potential maintenance. Pull-down experiments and native PAGE analysis indicated that the protein is both associated with the F<sub>1</sub>F<sub>o</sub> ATP synthase and integral to its assembly. In addition, its knockdown reduced the levels of F<sub>o</sub> subunits, but not those of F<sub>1</sub>, and disturbed the cell cycle. Canadian researchers have made significant contributions to the scientific understanding and treatment of depression. For example, Canada ranks first in the world on the number of articles published on depression (and second on papers determined to be Highly Cited in Field) when considering the proportion of researchers in each country. Canadian psychologists have played a critical role in advancing our knowledge of the epidemiology, vulnerability, treatment, and prevention of depression. This special issue presents a series of state-of-the-art empirical reviews that highlight many of the important contributions-covering cognitive vulnerability, perfectionism, epidemiology, cognitive behavioral therapy (CBT), mindfulness, and emotion-focused therapy-that Canadian psychologists have made to our understanding of depression. Some of the key themes from these reviews are summarized in this editorial. Aside from featuring outstanding scholarship, this special issue also serves as a tribute honoring the monumental contributions that many Canadian psychologists have made and as a catalyst to inspire next generation of depression researchers in Canada.Alternate :La dépression majeure (aussi appelée « dépression clinique ») est un trouble très répandu, périodique et débilitant qui touche plus de 264 millions d'individus de par le monde. Les chercheurs et les chercheures du Canada ont grandement contribué aux connaissances scientifiques sur la dépression et sur ses traitements. Par exemple, le Canada est au premier rang dans le monde pour le nombre d'articles publiés sur la dépression (et au deuxième rang parmi les plus cités dans le domaine) lorsqu'on tient compte de la proportion de chercheurs dans chaque pays. Les psychologues canadiens ont joué un rôle déterminant dans l'avancement des connaissances sur l'épidémiologie, la vulnérabilité, le traitement et la prévention de la dépression. Over the past 30 years, our work, as well as the work of other Canadian and international researchers, has attempted to understand the cause and maintenance of perfectionism and the role that this pernicious personality style plays in predisposing individuals to various problems, such as depression. In the present article, we outline our multidimensional and multilevel descriptive model of perfectionism and summarize several models of perfectionism and depression that we have focused on over the years, both from a diathesis–stress perspective and from a more complex developmental and relational perspective. We, then, outline the extant research that we and others have conducted based on these models and conclude by providing a description of and evidence for a dynamic-relational treatment of perfectionism that functions to reduce depressive symptoms and vulnerability to depression.
