Polymorphic CYP2D6 activity has been shown to be a determinant of the pharmacokinetics and pharmacodynamics of tramadol via hepatic phase I O-demethylation of (+)-tramadol to (+)-O-desmethyltramadol. Several studies have demonstrated the impact of CYP2D6 polymorphism on the pharmacokinetics of tramadol. Hence, the aim of this study was to determine if the well documented pharmacokinetics of tramadol regarding CYP2D6 could be verified in a group of Egyptian abusers. The genotype-phenotype relationships were also assessed. A total of 83 tramadol intoxicated subjects who were referred to the Poison Control Center (PCC), Ain Shams University Hospitals, were enrolled in the present study. Urinary concentrations of tramadol (TMD), and its metabolites, O-desmethyltramadol M1 (ODT) and N-desmethyltramadol M2 (NDT) were determined using the Trace GC-TSQ mass spectrometer. CYP2D6 genotyping was performed using PGX-CYP2D6 Strip Assay. Through the use of CYP2D6 phenotyping, 10 patients (12.1 %) were classified as CYP2D6 poor metabolizers (PMs), and 73 (87.9 %) were genotyped as CYP2D6 extensive metabolizers (EMs), including 57 (68.7 %) homozygous EMs and 16 (19.2 %) heterozygous EMs. Median TMD level didn’t differ significantly between PMs and EMs (p = 0.356). Median M1 level was significantly higher in EMs than that in PMs (p = 0.001), while median M2 level didn’t differ significantly between PMs and EMs (p = 0.597). There were statistically significant differences in TMD/M1, TMD/M2 and M1/M2 ratios between PMs and EMs (p = 0.001). Mahmoud","doi":"10.21608/ejaps.2019.175301","DOIUrl":"https://doi.org/10.21608/ejaps.2019.175301","url":null,"abstract":"In this study, Manganese dioxide decorated graphene oxide (GO-MnO 2 composite) was prepared via fixation of MnO 2 on the surface of graphene oxide (GO) and used as an adsorbent for separation of europium and cerium ions from aqueous solutions. GO, MnO 2 and GO-MnO 2 composite were characterized using scanning electron microscope (SEM), Thermal analysis (TGA), X-ray diffraction (XRD), Raman spectroscopy and Fourier transform infrared spectrum (FT-IR). The role of different variable as: contact time, solution pH, metal ion concentration, adsorbent dosage, ionic strength and temperature on sorption efficiency was studied using batch technique. It was found that sorption of Eu(III) and Ce(III) ions on synthesized adsorbents could be described more favorably by pseudo-second-order kinetic model. Sorption data have been interpreted in terms of both Freundlich and Langmuir isotherms. Desorption of Eu(III) and Ce(III) ions from loaded samples was studied using different eluents. The present work investigates the synthesis of hematite (Fe2O3) nanoparticles through a simple chemical precipitation method to be used as failure resistance modifier for water-based drilling fluids (WBFs) at high temperature/high pressure (HTHP) conditions. The surface morphology revealed by transmission electron microscopy (TEM) strategy suggested that the employed iron oxide nanoparticles (IONPs) are mainly present as big spherical shaped particles and are well crystallize in nature with an average diameter less than 50 nm. Scanning electron microscopy (SEM) technique confirmed the shape and size of the produced nanoparticles. The chemical composition of the synthesized Fe2O3 nanoparticles was confirmed by energy dispersive analysis of X-rays (EDX). Three water-based drilling fluids (WBFs) samples; blank unmodified, surface modified 1 g iron oxide nanoparticles (IONPs-A) and modified 2 g (IONPs-B) were prepared and studied. The rheological properties and filtration ability were investigated for the utilized samples at both 500 psi and 350°F (HTHP). The results of this study demonstrated that the addition of 1 g of IONPs to the drilling fluid sample displayed that the plastic viscosity (PV) was recorded at 8 cP and thixotropy at 2 lb/100 ft2 under HTHP conditions without mud failure due to establishing highly stable multiple bentonite-IONPs bridging effect.
