{"title":"Protective Effect of Lepidium Sativum Seeds and Buttermilk on Osteoporosis in Female Rats","authors":"رحاب محمد مراد, سحر سلطان عبد المجيد, اميلي توفيق حنا, داليا رفعت حسن","doi":"10.21608/sjseas.2022.221947","DOIUrl":"https://doi.org/10.21608/sjseas.2022.221947","url":null,"abstract":"Glucocorticoid-induced osteoporosis (GIO) is one of the serious side effects which have become the most common secondary osteoporosis. The study aimed to evaluate the effect of different levels of Lepidium sativum seeds and buttermilk on glucocorticoidinduced osteoporosis in rats. Methods: The present study was carried out in the National Nutrition Institute NNI Cairo, on forty-eight female rats. The rats were divided into eight groups (six rats each). The first group was fed on a basal diet and represents the negative control, while the other seven groups were injected subcutaneously with betamethasone at a dose of 4 mg/ kg BW three times a week. One group of them represents the positive control. The other six groups were fed on a basal diet containing 10%, 20% Lepidium sativum seeds (LSs), 10%, 20% buttermilk (BM), 10% and 20% mixture of LSs and BM for a period of eight weeks. Results: The positive control group showed a significant decrease in serum Estradiol (E2), Calcium (Ca), Phosphorus (P) levels and Ca and P in femur bone and a significant decrease in total Bone Mineral Density (BMD), and a significant increase in serum Alkaline phosphatase ةيـــقيبطتلا مولـعلاو ةـــيعونلا ةــــيبرتلل ةيــــملعلا ةلــجملا es The Scientific Journal of Specific Education and Applied Scienc 87 (ALP). On the other hand, all osteoporosis groups administrated with different levels of LSs and BM (10 and 20%) had a significant decrease in (ALP) and a significant increase in Ca, P in serum and Ca, P in bone and BMD, compared with the positive control group. The pathological examination of bone confirmed these results. Conclusion: Lepidium sativum seeds and buttermilk showed bone protection against glucocorticoid-induced osteoporosis in rats. Lepidium sativum seeds had a potent protective effect more than Buttermilk due to its content of isoflavones. Thirty-five albino rats of Sprague-Dawley strain were randomly distributed after the adaptation period into two main groups (7 of each), the first main group was kept on the basal diet as negative control (-ve), The rest of the animals (n = 28) were subcutaneously injected with a single dose of CCl4 for the induction of necrosis. After injection of CCL4, rats were divided into 4 subgroups. The first subgroup of rats was as a positive control group (+ve). The other subgroups were fed on the experimental diets containing different levels of corn silk 5%, 7.5% and 10%, respectively. At the end of the experimental period (6 weeks) liver and kidney functions were determined. Also, antioxidants enzymes in addition MDA were determined. Histopathological examination for liver and kidneys were done. The results indicated that liver enzymes including ALT, AST, ALP in addition albumin, total and direct bilirubin were decreased in groups 3,4 and 5, respectively compared with the positive control group. Urea nitrogen and creatinine were significantly improved in all tested groups compared with the positive control one. Moreover, antioxidant enzymes (SOD and CAT) were significantly improved in all examined groups compared with the positive control group. However, serum MDA decreased significantly in tested groups which were treated with 5%, 7.5%, and 10% corn silk when compared with the positive control group.
