Matrix metalloproteinase 13 (MMP-13) has the ability to activate osteoclasts, leading to bone resorption. Regenerative treatments have been widely used in periodontology. When combined with Platelet-rich fibrin (PRF), xenografts will give better results in bone regeneration. The aim of this study was to evaluate the effect of xenograft combined with PRF on MMP-13 expression in a bone defect using an experimentally created bone defect. Material and Methods: eighteen New Zealand rabbits were assigned to three groups. Each group consisted of six New Zealand rabbits. A critical bone defect with a diameter size of 5 mm was created in the right tibia of each rabbit in group 1 (application: xenograft), group 2 (application: PRF), and group 3 (application: xenograft and PRF). The PRF was produced from 5 ml of blood taken from each rabbit’s ears. After 30 days, the rabbits were euthanized. The tissue samples were evaluated by immunohistochemical staining. Results: group 3 showed the lowest mean expression of MMP-13 (4.50) compared to group 1 (20.50) and group 2 (11.70). Group 3 showed a significant difference in the MMP-13 expression compared to group 1 and group 2 (P = 0.000) (P < 0.05). Conclusion: this research showed that the combination of xenograft and PRF had the lowest expression of MMP-13. Calcifying odontogenic cyst (COC), or Gorlin’s cyst, is a well-recognized example of these lesions. We describe a case of COC with AOT-like areas and highlights its morphological diversity. A 60-year-old pheoderma man presented with a large swelling in the anterior buccal region of the mandible. Panoramic radiography revealed a well-defined, unilocular, radiolucent lesion associated with important root resorption. Complete enucleation of the lesion was performed and the histopathological findings met the criteria for the diagnosis of COC, although the cyst exhibited unusual AOT-like features. The patient has been recurrence free for 6 months after surgery. COCs with AOT-like features are rare, and reflect the multipotentiality and complexity of the inductive effects of the odontogenic epithelium with the ectomesenchyme. Enucleation seems to be the most indicated treatment, similar to classical COC. Material and Methods: a double blinded randomized control trial was conducted in 45 children, who were indicated for pulpectomy in any one of the primary mandibular molars. The canal preparation was done using either hand K-files, Kedo-S files, or Kedo-S Square files based on the groups assigned. The time taken for instrumentation was recorded using a stopwatch. The quality of obturation was evaluated using post-operative radiograph and post-operative pain was assessed with modified Wong-Baker Faces Pain scale. Results: instrumentation time was minimum in rotary Kedo-S Square files (53.23 ± 9.60 seconds) followed by Kedo-S files (82.70 ± 11.86 seconds). The preparation time was maximum with manual K-files (121.43 ± 20.18 seconds). Kedo-S square files provided a higher number of optimally filled canals (66.4%). All the three instrumentations equally showed the tendency to produce voids in the obturation. Rotary files Kedo-S Square followed by Kedo-S showed less post-operative pain compared to K-files.
