Morphological changes of\u0000the skeletal muscles in the first stage are characterized by presence of dystrophic-necrotic processes and reflect\u0000the compensatory-adaptive reaction of the organism to hypoxia. In the third stage one can see the progress of\u0000morphological damages, which develop during the ischemic period against a background of exhaustion of\u0000proteinase inhibitors. This indicates the intensity of endogenous intoxication of the organism with the products\u0000of disturbed metabolism and determines the irreversibility of destructive processes and probability of multiple\u0000organ failure. Proceeding from the character of the pathomorphological changes and the state of proteinase-inhibitor system one can suppose, that the optimal time for medical measures is the first stage and the first hours of the second stage (to increase the ischemic tolerance of the skeletal muscles). Taking into account the direct relation between the intensity of pathomorphological injuries and the imbalance of proteinase-inhibitor system, the usage of proteinase-inhibiting medicines for correction of RS-development and reduction of the destructive changes during first and second stages is substantiated. With the assistance\u0000of virological methods was found that the TBEV are absorbed by macrophages and replication in them. An\u0000ultrastructural study has shown that the virus enters into the cytoplasm by local destruction of plasmalemma\u0000and newly synthesized virus particles exited from the cell by same. Simultaneously there is a seal of perinuclear\u0000cytoplasm space, where found in a large number of ribosomes, microfilaments, ribonucleoprotein fibers and viral\u0000special structure: nucleocapsids, tubular formations and viral layers (fabrics). On the surface of last structures\u0000the newly synthesized virus particles were visualized. Thus, the evidence shows that macrophages play a role\u0000in the spread of TBEV, being for their the target cell. Direct contact of the most microsporidia species with host cytoplasm suggests that these parasites may control physiological processes of infected cells by secretion of various proteins. In previous experiments, secretion of significant amounts of microsporidia Paranosema locustae alpha/beta-hydrolase into infected cells of Locusta migratoria fat bodies was demonstrated using polyclonal antibodies against the enzyme. However, heterologous expression of microsporidian hydrolase in yeast Pichia pastoris cells was not accompanied by its secretion. In this study, we have constructed library of recombinant single chain antibodies (scFv-fragments) against proteins of fat bodies of infected locusts and isolated mini-antibody specifically recognizing the studied enzyme using phage display technology. Immunoblotting and immunofluorescent microscopy with selected scFv-fragment confirmed secretion of two different in size forms of P. locustae alpha/beta-hydrolase into infected host cell. The present study is focused on ITLN1, PPARã and TNFá gene expression in intra-abdominal adipose tissue and its effect on the serum levels of omentin 1 and TNFa in individuals with different body mass. It has been shown that serum TNFa level is significantly higher in the subgroup of patients with overweight and obesity (BMI ≥ 25 kg/m2) as compared to individuals with normal body weight (BMI < 25 kg/m2)( p < 0.03). We have demonstrated that the expression level of the PPARã gene is positively correlated with the ITLN1 gene expression level in the intra-abdominal adipose tissue (r = 0.516, p = 0.020). Serum level of omentin 1 positively correlates with PPARã mRNA and protein levels in intra-abdominal adipose tissue (r = 0.550, p < 0.05 and r = 0.581, p < 0.03, respectively). For the subgroup of patients with overweight and obesity, we have shown negative correlation of the level of TNFá mRNA with PPARã and ITLN1 mRNA levels was shown (r = –0.549, p < 0.05 and r = –0.475, p < 0.05, respectively). Here we studied the effects of histone deacetylase inhibitor, sodium butyrate, and MEK/ERK-pathway inhibitor, PD0325901, on cells with modifications in genes involved in anti-cancer therapy response, Wip1 phosphatase and p53. We have investigated the effect of these agents on cell cycle of wild-type cells, Wip1 knockout cells and cells with double deletion of Wip1 and p53. Our results showed that more severe changes in S and G2/M phases were observed in response to sodium butyrate in Wip1-defecient cells than in wild-type cells. Meanwhile, PD0325901 treatment led to G1 arrest. At the same time, a «sodium butyrate type» response dominated the response to combined treatment with both drugs in Wip1-deficient cells, while the response of Wip1–/–/p53–/– cells to combined treatment was similar to the single use of PD0325901. Wip1–/– and Wip1–/–/p53–/– cells were more sensitive to the use of PD0325901 than wild-type cells. Obtained results suggest that Wip1 deficiency sensitizes cells to sodium butyrate and to MEK/ERK inhibitors independently from Wip1 main target protein — p53. We have identified several morphological changes in tubulin cytoskeleton that appear during the acclimation to salt and osmotic stress in the cells of different root tissues: 1) decreased density of cortical microtubule network, 2) random orientation of cortical microtubule bundles, 3) non-uniform density of the bundles, 4) thickening of the bundles, 5) fragmentation of the bundles, 6) formation of centers of converging microtubule. Reduced density of the microtubule network and thickening of the bundles was detected during osmotic and salt stress, yet random orientation of cortical microtubules was observed under osmotic stress and not found during salt stress. Fragmentation of microtubule bundles was apparent during salt stress and less evident at high concentration of mannitol. Formation of centers of converging microtubule was common under prolonged action of sodium sulfate, less common under sodium chloride and not found after mannitol treatment. Our data show that cortical microtubules in alfalfa root cells rearrange not only in response to different ions, but also to osmotic pressure. Thus, the signaling pathways and molecular mechanisms inducing reorganization of the microtubule system may be triggered not only by sodium cations but also by sulfate and chloride anion at the concentrations that do not cause irreversible cell damage. It was previously considered\u0000that the nuclear lamina predominantly plays a structural role, however, its involvement have been recently\u0000described in the regulatory processes such as chromatin organization and gene transcription. It is known that\u0000mutations in the LMNA gene lead to the development of a large number of diseases, laminopathies, which mainly\u0000affect mesenchymal tissue. Nowadays, the mechanisms by which the lamina can regulate cell differentiation\u0000remain incompletely understood. In the present work, we have studied the effect of LMNA gene mutations on\u0000the process of muscle differentiation of primary satellite cells and Ñ2Ñ12 cell line. The genome of satellite cells\u0000and Ñ2Ñ12 cell line was modified by the introduction of lentiviral constructs encoding LMNA G232E associated\u0000with the development of muscular dystrophy Emery—Dreyfus and LMNA R571S associated with the development\u0000of dilated cardiomyopathy. The morphology of the cells was estimated using immunofluorescence, the\u0000expression level of myogenic genes were analyzed by qPCR. We have shown that the analyzed mutations reduce\u0000the ability of cells to differentiate, to fuse and to form myotubes.
