{"title":"黏连蛋白调节DNA复制时序以维持基因组稳定性","authors":"Jinchun Wu, Yang Liu","doi":"10.1360/tb-2023-0936","DOIUrl":"https://doi.org/10.1360/tb-2023-0936","url":null,"abstract":"人类细胞的细胞核中, 高达3.2×10个碱基对组成的染色 质在细胞中高度折叠, 形成染色体区室(compartments)、拓扑 相关结构域(topologically associating domains, TADs)以及染 色质环(chromatin loop)等结构层级. 黏连蛋白(cohesion)复 合体是由蛋白亚基SMC1、SMC3、RAD21和STAG1/2组成 的环状结构, 与CTCF (CCCTC-binding factor)蛋白一起通过 染色质环挤出(loop extrusion)过程调节染色质环的形成, 介 导远距离染色质间的相互作用. 此外, 黏连蛋白还介导姐妹 染色单体的黏连和分离. 近年来, 黏连蛋白复合体蛋白亚 基的功能缺失型突变在结直肠癌、胶质细胞瘤、白血病和 黑色素瘤等多类癌症中被广泛发现. 然而, 黏连蛋白突变与 癌症等疾病发生、进展的关系尚无定论. 为了探究黏连蛋白功能缺失的影响, 本课题组在人源 K562细胞和小鼠胚胎干细胞(mouse embryonic stem cells, mESCs)中构建了生长素诱导的RAD21蛋白的急性降解体系, 利用高通量测序和细胞成像等方法发现了黏连蛋白在DNA 复制和基因组稳定性维持中的重要功能, 进一步揭示了黏连 蛋白功能丧失在癌症发生过程中的可能机制, 相关工作发表 于Nature Genetics. 该体系通过在培养基中添加吲哚乙酸 (indole-3-acetic acid, IAA)诱导内源RAD21蛋白快速降解, 从 而破坏黏连蛋白的功能. 值得注意的是, 为避免残留RAD21 的干扰, 我们还结合细胞分选以得到RAD21完全降解的细胞 进行后续实验. 首先, 我们发现, γH2AX的免疫荧光信号水 平在RAD21降解后的细胞中显著上升, 提示RAD21的降解可 能导致了DNA双链断裂(DNA double-stranded breaks, DSBs) 的产生. 接着, 利用可定量检测DSB水平及发生位点的PEMseq (primer-extension-mediated sequencing)方法, 发现 RAD21降解导致细胞内DSBs增加了3~5倍, 且在白血病中发 现的RAD21功能缺失型突变体不能回补RAD21降解导致的 DSB增加. 通过对RAD21降解导致的DSBs发生位点进一步分 析, 鉴定到了147个DSB热点基因. 值得注意的是, 超过1/3的 热点基因在数据库中被注释为癌症标志基因或者疾病相关 基因, 如MAPK1、BCR、RUNX1等(图1(a)). 通过分析TCGA (The Cance r Genome At la s )数据库中的病人样本 , 我们发现, 在结直肠癌、非小细胞肺癌、黑色素瘤、胶质细 胞瘤和子宫内膜癌中, 77.6%~98.3%的DSB热点基因与黏连 蛋白蛋白亚基基因具有显著的共同突变现象. 这些数据不仅 说明RAD21降解所导致的黏连蛋白功能丧失可以模拟癌症 中黏连蛋白亚基的功能缺失型突变, 还提示黏连蛋白亚基的 失活突变可能正是癌症及其相关疾病发生和发展的诱因 之一. 那么, 黏连蛋白功能缺失导致DSB发生的机制是什么? 前期研究发现, 大多数在癌细胞中发现的黏连蛋白功能缺失 型突变不会影响细胞分裂过程. 另外, 黏连蛋白功能缺失 只能扰动少量基因的表达. 我们发现, RAD21降解与否并不 影响DSB在染色质相互作用变化最为剧烈的染色质环锚定 位点(loop anchor)处的分布. 而且, 在147个DSB热点基因中, 仅有7个基因的转录水平在RAD21降解前后出现了差异表达. 这些数据共同提示染色质相互作用和转录的改变并不是导 致DSB增加的直接原因, 这与之前的研究结果一致. 有意思 的是, PEM-seq鉴定的DSB位点在正负链上非对称分布, 且该 分布偏向性与冈崎片段的分布高度相似, 提示RAD21的降解","PeriodicalId":34927,"journal":{"name":"Chinese Science Bulletin-Chinese","volume":"76 11","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-11-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135062571","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"综合性期刊","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
