Berry breaking is a critical practice for grape growers as it directly impacts fruit quality, ensuring optimal flavor development and desirable characteristics in the final product. An experiment was conducted over the 2021 and 2022 seasons at a commercial farm with sandy soil, focusing on 12-year-old vines. The experiment involved grafting 'Freedom,' 'Ruggeri 140Ru,' 'Paulson 1103P,' and 'Superior Seedless' rootstocks, and evaluating the results. Harvest dates were determined by testing bundle samples using SSC (16%) and storing them at room temperature for four days. The Paulson 1103P rootstock showed a lower rate of berry breaking compared to other rootstocks. The activity of xylanase (XLN; EC:3.2.1.8), polygalacturonase (PG; EC:3.2.1.15), and cellulase (CEL; EC: 3.2.1.4), was also comparatively reduced. Throughout the four days, the Paulson 1103P rootstock demonstrated superior physical and chemical quality traits. The levels of cell wall hydrolysis enzymes, including PG, XLN, and CEL, were lower in vines grafted onto the Paulson 1103P rootstocks. The influence of the Paulson 1103P rootstock on physical and chemical quality traits persisted throughout the first four days of shelf life. The results suggest that 'Superior Seedless' vines grafted onto the Paulson 1103P rootstock exhibited a more pronounced decrease in berry breaking rate until the end of the four-day trial storage period under ambient air conditions.","PeriodicalId":326835,"journal":{"name":"Journal of Agricultural Chemistry and Biotechnology","volume":"21 19","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-03-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140396664","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

A local fungi isolate was isolated from soil samples and then tested for their silver resistance. The most silver resistant isolate was chosen for the biosynthesis of AgNPs, which was identified genetically as Penicillium oxalicum DS-2 dependent on the sequence of 5.8S rRNA gene. For the detection and characterization of AgNPs, UV-Vis spectroscopy and transmission electron microscopy (TEM) were used. The appearance of a brown color after adding silver nitrate to the fungal filtrate and UV-visible absorbance peak around 410 nm emphasized the biosynthesis of AgNPs. The image of TEM showed that the biosynthesized AgNPs were spherical shape with some elongated particles and the size of particles was in the range 13–35 nm. The biosynthesized AgNPs were coated and stabilized using chitosan in form of chitosan-AgNPs (Cs-AgNPs). The antibacterial activity of the AgNPs and Cs-AgNPs were evaluated on three pathogenic bacterial strains including Pseudomonas aeruginosa , Escherichia coli (Gram negative) and Staphylococcus aureus (Gram-positive). Both AgNPs and Cs-AgNPs achieved strong antibacterial activities compared to streptomycin as standard antibiotic. Cs-AgNPs had higher antibacterial activity than AgNPs. Scanning electron microscopy (SEM) analysis showed that Cs-AgNPs densely accumulated and adhered to the surface of treated bacterial cells, with consequent penetration of silver nanoparticles into the cell walls and cell death. The results of SEM confirmed the superiority of Cs-AgNPs as a promising antibacterial.","PeriodicalId":326835,"journal":{"name":"Journal of Agricultural Chemistry and Biotechnology","volume":"30 2","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140498601","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Cyanobacteria were isolated from samples of soil site polluted with insecticides in Kafr El- Sheikh, Governorate. Cyanobacteria were purified by different purification methods. Twelve cyanobacterial isolates were identified according to standard methods based on cultural and morphological characters of cyanobacteria as following: color of culture, morphology of thallus, vegetative, reproductive cells and heterocyst. Heterocyst from isolates of cyanobacteria were observed in Anabaena sp., Nostoc sp., Oscillatoria sp. and Chroococcus sp. Nostoc sp. was of the highest frequency (66.6 %) among the isolated cyanobacteria. While, Oscillatoria sp. and Chroococcus sp. were less frequency (8.3%). Dry weight and nitrogen fixation of cyanobacteria were determined for efficiency evaluation of these isolates. The superior cyanobacterial isolates, which isolated from soil polluted with insecticides were identified by 16S rRNA as Nostoc muscorum and Anabaena oryzae .","PeriodicalId":326835,"journal":{"name":"Journal of Agricultural Chemistry and Biotechnology","volume":"66 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-06-13","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"125232054","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
