The experiments were performed in the laboratory of Food Research Division, Nepal Agricultural Research Council. The three different medium type rice varieties of Nepal (Lumle-2, Chhomrong and Machhapuchre-3) were exposed to five different degrees of milling (0%, 6%, 8%, 10% and 12%). The degree of milling (DM) level significantly (P≤0.05) affected the milling recovery; head rice yield, nutrient content as well as cooking properties of the rice. Increase in DM resulted in further reduction of protein content, fat content, minerals, milled rice and head rice yield after bran layer was further removed. A positive correlation between DM used in present model, amylose content, kernel elongation and gruel solid loss was observed, however, with an increase in DM; amylose content, kernel elongation and gruel solid loss were found to be increased. Adopting 6 to 8% DM for commercial milling of rice might help to prevent quantitative, qualitative and nutritional loss along with retention of good cooking characteristics.","PeriodicalId":31348,"journal":{"name":"Journal of Nepal Agricultural Research Council","volume":"4 1","pages":"7-17"},"PeriodicalIF":0.0,"publicationDate":"2018-05-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.3126/jnarc.v4i1.19685","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"46721651","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Although the molecular diagnostic protocol used in regulatory programs has been evaluated but till date methods implying direct comparison has rarely used. In this study, a known area soil samples from potato fields where light blight appear every year (both A1 and A2 mating type) was assayed by soil bait method, PCR assay detection and quantification of the inoculums. Suspected disease symptoms appeared on bait tubers were further confirmed by rapid PCR, inoculums were quantified through Real Time PCR, which confirms presence of P. infestans. These diagnostic methods can be highly correlated with one another. Potato tuber baiting increased the sensitivity of the assay compared with direct extraction of DNA from tuber and soil samples. Our study determines diagnostic sensitivity and specificity of the assays to determine the performance of each method. Overall, molecular techniques based on different types of PCR amplification and Real-time PCR can lead to high throughput, faster and more accurate detection method which can be used in quarantine programmes in potato industry and diagnostic laboratory.","PeriodicalId":31348,"journal":{"name":"Journal of Nepal Agricultural Research Council","volume":"4 1","pages":"25-32"},"PeriodicalIF":0.0,"publicationDate":"2018-05-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.3126/jnarc.v4i1.19687","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"46045769","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The parameters used were cardamom production area, type of manure used, drying facilities, technical skills of farmers, market channels and variable cost etc. We purposively selected 30 cardamom producers and stakeholders for interview pre-designed questionnaires. The result showed that average area, production and productivity of large cardamom per household were 0.86 ha, 200 kg and 232 kg.ha-1, respectively, with the average farming experience of 22 years. It was revealed that 13% farmers used farmyard organic manure, the use of 1.5 kg/plant farmyard manure might produce 28.5% higher yield cardamom compared to without using any manure or fertilizers. It was also revealed among the responded only 7% had received improved drying machine from District Agriculture Development Office (DADO) at 50% subsidy, while only 23% of farmers received training and technical services from DADO. The study showed that per hectare average total cost of large cardamom production, selling price and gross revenue were NRs. 2,36,705 ($2255), NRs. 5,50,305 ($5240) and NRs. 3,13,600 ($2985), respectively, with benefit/cost (B/C) ratio of 2 after the completion of gestation period of 4 years. Our survey showed that predominant marketing approach was by direct sell to the traders located at district headquarter. The productivity of large cardamom was influenced by various factors, such as nearly 75.2% of the variation in productivity was explained by the number of active family members, farming period, area, intercultural operations, variable cost and depreciated fixed cost.","PeriodicalId":31348,"journal":{"name":"Journal of Nepal Agricultural Research Council","volume":"4 1","pages":"79-85"},"PeriodicalIF":0.0,"publicationDate":"2018-05-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.3126/jnarc.v4i1.19693","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"44162236","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, availability of disease free planting material (rhizome) is the major problem faced by Nepalese farmers. Tissue culture is the only option to produce disease free rhizome of ginger. Suitable culture media combination is most important for the production of planting material in ginger through tissue culture. Therefore, effect of different level of sucrose and growth regulators on micro-propagation of ginger was studied using local collection ‘Kaski Local’. Early stage bud was used as explant. MS basal media with different level of sucrose and growth regulators was used as tissue culture media. 30 g/L sucrose, 30 g/L sucrose+5mg/L BA, 30 g/L sucrose+5 mg/L BA+0.5 mg/L NAA, 60 g/L sucrose+5mg/L BA, 60 g/L sucrose+5 mg/L BA+0.5mg/L NAA, 90 g/L sucrose+5 mg/L BA was used in this study. The explants were surface sterilized, cultured and incubated at 25±2°C, 90-95% relative humidity and 14:10 hours light:dark photoperiod for 8 weeks. Increased level of the sucrose increased the rhizome weight, however, addition of NAA produced more positive effect for this. MS basal media with 60 g/L sucrose+5 mg/L BA+0.5 mg/L NAA produced higher rhizome weight. The study was performed in a glasshouse system where separate tubers from the root zone were inoculated at different intervals during plant growth along with a novel hydroponic system to inoculate individual tubers at specific times of development growth allowing non-destructive observations of common scab symptoms developing. The window of tuber susceptibility to common scab disease infection was shown to vary with the season or conditions under which the plants were grown. Different internodes on tubers were found susceptible to infection at different times during tuber development. Basal internodes, which are the first sections of the tuber to expand, were susceptible to infection in the beginning of tuber development, whereas apical internodes only became susceptible later in tuber growth when the basal internodes were no longer susceptible.","PeriodicalId":31348,"journal":{"name":"Journal of Nepal Agricultural Research Council","volume":"3 1","pages":"19-26"},"PeriodicalIF":0.0,"publicationDate":"2017-05-08","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.3126/jnarc.v3i1.17271","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"41889562","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Thus, the aim of the present study was to examine the use of resistant root stock of wild brinjal ( Solanum sisymbriifolium ) to reduce the loss caused by the nematodes on tomato. For the management of root-knot nematodes, grafted plant with resistant root stock of the wild brinjal was tested under farmers’ field conditions at Hemza of Kaski district. Grafted and non-grafted plants were produced in root-knot nematode-free soil. Around three week-old grafted and non-grafted tomato plants were transplanted in four different plastic tunnels where root-knot nematodes had been reported previously. The plants were planted in diagonal position to each other as a pair plot in 80 × 60 cm 2 spacing in an average of 20 × 7 m 2 plastic tunnels. Galling Index (GI) was recorded three times in five randomly selected plants in each plot at 60 days intervals. The first observation was recorded two months after transplanting. Total fruit yield was recorded from same plants. In the grafted plants, the root system was totally free from gall whereas in an average of 7.5 GI in 0-10 scale was recorded in the non-grafted plants. Fruits were harvested from time to time and cumulated after final harvest to calculate the total fruit yield. It was estimated that on an average tomato fruit yield was significantly (P>0.05) increased by 37 percent in the grafted plants compared with the non-grafted plants. Grafting technology could be used effectively for cultivation of commonly grown varieties, which are susceptible to root-knot nematodes in disease prone areas. This can be used as an alternative technology for reducing the use of hazardous pesticides for enhancing commercial organic tomato production. Enhancing adoption of improved potato varieties could impact on farmer’s income, household food and nutritional security. As such, using a multistage sampling procedure, a study was conducted to assess the determinants of improved potato varieties adoption in Nepal covering 180 samples in four districts, two in hills and two in Tarai region. The study revealed that; Kavre and Bardiya districts in the hills and Tarai, respectively, were dominated by improved potato varieties adoption. On the other hand, Dhankuta and Jhapa in the hills and Tarai, respectively, were dominated by local potato varieties adoption. The informal seed sources followed by agro-vet and market were the major sources for improved varietal adoption. Farmers’ accesses to training and formal seed sources were important factor determining improved potato varietal adoption. However, households with larger farm size were less likely to allocate more area for improved potato varieties as many of farmers were reluctant to take potato cultivation as agri-business and still follow subsistence farming. Potato R&D programs, therefore, need to strengthen formal seed system to enhance access to quality potato seeds and build producer’s capacity through regular training and exposer visits in order to improve adoption of improved potato varieties in Nepal. Despite of its importance as cash crop in many parts of Nepal, much study on various aspects for enhancing production and productivity has yet to be done. Therefore, to evaluate the production performance different genotypes of garden pea in eastern hills agro-ecological conditions present experiments were carried out consecutively for two years (2015 and 2016) at Agricultural Research Station, Pakhribas. The experiment comprised of 11 different genotypes of garden pea including a check variety Arkel. The production performance was evaluated in a completely randomized block design with three replications. The seeds were sown at 50 × 10 cm spacing during first week of October for two years. The result showed that DGP-05 genotype had earliest 104 days after sowing. The DGP-08 genotype showed 13 which were the maximum numbers of pods per plant (13), while DGP-01 showed 8 numbers of seeds as the maximum per pod. The DGP-03 genotype had the longest pod of 9.78 cm among others. The highest fresh pod yield of 18.14 t/ha was achieved from genotype DGP-09 followed by Arkel with (16.32 t/ha).
