The subject of this study is the bonus system adopted in New York City to create privately owned spaces for public use, known as POPS (Privately Owned Public Spaces). Based on Jan Gehl’s theoretical principles, a quantitative and qualitative survey on Park Avenue was carried out. The analysis of the collected data showed that buildings’ corporate plazas not only contribute to public use of free spaces but also ennoble the urban landscape of the area. {"title":"Antropologia do lugar na Villa Grimaldi: espaço público e pluralidade de simbolismos na paisagem urbana chilena","authors":"M. Diligenti, Maria Alice Medeiros Dias","doi":"10.11606/ISSN.2359-5361.V0I39P183-198","DOIUrl":"https://doi.org/10.11606/ISSN.2359-5361.V0I39P183-198","url":null,"abstract":"Villa Grimaldi, located in the metropolitan area of Santiago de Chile, comprises an area of approximately 10,200 m², which, at the beginning of the 20th century, served as a meeting place for artists and intellectuals, where discussions on the various areas of Chilean and world culture were held. After the military coup of 1973, this space housed the largest detention center of torture under the Chilean military regime. During the democratic “Concertacion” period, after years of neglect, the area was turned into Corporacion Parque por la Paz Villa Grimaldi. In 2004, the area was declared a National Monument, looking back at the period in which the village was used by the military and paying tribute to those who resisted the Chilean dictatorship. In view of the place’s paradoxical uses, this article seeks to establish the relations between the configuration of this collective space, its real and symbolic meanings and its integration in the urban fabric during each or the historical periods investigated in the research. The methodology consisted of a qualitative research of both the literature and the documents collected in the city of Santiago in the years 2014 and 2015. Este trabalho apresenta as digressoes teoricas que foram permitidas ao longo do desenvolvimento de proposta para o Parque Municipal da Brasilândia. O distrito possui uma porcao de area verde alvo de campanhas para transformacao em parque. A proposta aqui discutida tem como diretriz o enraizamento do parque no bairro como estrategia para assegurar sua insercao como espaco fisico e simbolico. Para isso sao sugeridas novas relacoes entre o espaco construido e a area verde, utilizando-se das oportunidades encontradas na estrutura da cidade informal para entrelacar espaco construido e zonas de natureza, tempo de lazer e vida cotidiana. New regulations and current laws still do not guarantee the best management system of these areas, which are inadequately offered to public use. The objective of this research is to evaluate the management system of Conservation Units of the category “Natural City Parks” in the north region of Rio Grande do Sul. Four factors were analyzed during the survey and diagnosis of the current management of these parks: environmental, social, economic/financial and institutional. Extreme events, such as temperature rising, droughts, and floods, also threaten cultural heritage. In historic city centers, floods and heavy rain have caused pathologies in properties and streets. The traditional urban drainage system has shown to be inefficient, in addition to the difficulties of expanding this system due to pre-existing matters. This study seeks to use public open spaces in historic city centers to improve urban drainage, through the concept of green infrastructure. At a first glance, it draws a relation between climate changes, and the disturbances to the cultural landscape, also how a green infrastructure can contribute to a resilience of the historic city centers. Next, the public open spaces system available for this application is analyzed, taking the the city of Laguna, in Santa Catarina, as a case study.As a result, public open spaces should be used as an infrastructural feature instead of being there as mere scenery. Thus, they shall become vital to the urban centers, valuing landscape, and increasing the effect of the drainage, without impacting or disregarding existing geomorphology and urban morphology.Climate change affects city life. {"title":"[Establishment and validation of human cancer cell lines with stable Cas9 expression].","authors":"X C Bian, Z L Yang, H L Feng, X M Zhao, B Gu, J Li, H Sun, Y Q Liu","doi":"10.3760/cma.j.issn.0529-5807.2017.01.010","DOIUrl":"10.3760/cma.j.issn.0529-5807.2017.01.010","url":null,"abstract":"<p><p><b>Objective:</b> To establish human cancer cell strains with stable Cas9 expression, and to validate the gene editing activity of Cas9 for simple gene editing in future study. <b>Methods:</b> Fifteen cancer cell lines of different tissue origins were infected with pLv-EF1α-Cas9-Flag-Neo or pLv-EF1α-Cas9-Flag-Puro by lentivirus and clone selection was employed to screen Cas9 stably expressed cancer cell lines. Afterward designed guide RNA vectors targeting TSC22 gene were transiently transfected into 3 of cell lines, and subsequently the gene editing activity of Cas9 was evaluated by genomic PCR, sequencing and Western blot. <b>Results:</b> Sixty-nine human cancer cell strains with stable Cas9 expression from different cancers were established, and by transient transfection with designed guide RNA, long fragment deletion was detected in TSC22 gene. <b>Conclusions:</b> Sixty-nine human cancer cell strains are successfully established with stable expression of Cas9 protein and gene editing activity. We present the case study of Sao Goncalo do Rio Abaixo, a town that is going through a deep transformation process due to the iron mining activities in the Iron Quadrangle in the state of Minas Gerais. The methodological process presupposes the classification of RapidEye satellite images and mapping of use and land cover; application of landscape metrics (area, perimeter, distance to nearest neighbor, core area, shape index and NRVI); and the integration of metrics using multi-criteria evaluation. Areas that are appropriate for the application of public policies, which promote the management of the vegetation cover, have been identified. And the open space system inside the informal city represents great investigative potential in regard to its physical characteristics, to the articulation established between public and private spaces and, mostly, to the public and ordinary practices developed inside it. With this aim, the article selects three favelas in the city of Sao Paulo as case studies: the favela de Paraisopolis, the second biggest community in the municipality, which is located in the Morumbi neighbourhood with around 100.000 inhabitants in an area of 100 acres; the favela da Linha and the favela do Nove, which together occupy less than 2 acres in the General Stores Company of Sao Paulo (Ceagesp) neighborhood, concentrating 630 families. It reports the experiences resulting from an agreement between the University and the Municipal Government of Campinas to devise a linear park project. It seeks to reflect on the potential of such agreements as a means to develop both research and teaching practices based on projects involving different city planning agencies on a city scale.
