During adolescence, up to 90% of people suffer, and in adulthood — about 20% with varying degrees of severity. Currently, there are many approaches to treating this disease through various cosmetic treatments such as phototherapy, ultrasonic skin cleansing, Mesotherapy, chemical peels, and medication. Therefore, the development of methods and means of differential diagnosis of acne is one of the urgent tasks in the field of biomedical engineering, dermatology, and clinical medicine, since this allows timely identification of the localization of the disease, its causes, and prescribing appropriate treatment. However, the solution to the problem of monitoring the dynamics of external manifestations of the disease is possible only with the use of combined mathematical methods for image analysis.\u0000\u0000Objectives: To develop a comprehensive method for analyzing dermatoscopic images for monitoring the external manifestations of acne disease during treatment and isolating the affected areas of the facial skin.\u0000\u0000Materials and Methods: Dermatological preclinical researches of the skin were conducted in the laboratory of 3D-biomedical technologies of the Department of Biomedical Engineering of the Kharkiv National University of Radio Electronics, using a digital videodermatoscope BIO Bm6+ in daylight and a portable skin analyzer Skin Scope F-102 in the ultraviolet range. Clinical researches were conducted based on the Department of Pediatric Propaedeutics #2 of the Kharkiv National Medical University. The development of a software tool for image analysis was conducted out in Python programming using the libraries OpenCV, Scikit-image, Numpy, PIL, Mathplotlib. Determination of the affected skin areas and calculation of the parameters of inflammation were carried out using multi-Otsu methods and morphological segmentation of digital dermatoscopic images.\u0000\u0000Results: During the research, automated software was developed that allows to analyze in dynamics the nature of inflammatory processes and the area of facial skin lesions, as well as to carry out a differential diagnosis of acne disease. The proposed method for the analysis of dermatoscopic images makes it possible to perform color segmentation and obtain a map of the gradations of skin inflammations to control the dynamics during the prescribed treatment.\u0000\u0000Conclusions: The comprehensive method of analysis of dermatoscopic images of the skin of the face makes it possible to effectively control the condition of the skin of the face from acne during treatment, while analyzing the degree of inflammatory processes and the area of lesions, w

Since that the tissues and bones have different absorption densities from such projections it is possible to get a reasonably accurate idea of the presence of pathological formations or lesions.\u0000\u0000Despite all the advantages of tomography, it has disadvantages. One of the most serious is the exceedance of radiation for humans. Therefore, there are several options for reducing the burden on the patient, one of which is the use of tomosynthesis instead of CT scanning. Tomosynthesis occupies an average position in informativeness between computer tomography and radiology not much higher than the dose of the latter. As a result, work is underway worldwide to improve the tomosynthesis method to increase the number of diseases that can be diagnosed by this method and to improve the quality of the images obtained.\u0000\u0000Objectives: Defines the efficiency of the image recovery method when using the Radon transform in general form compared to classical methods.\u0000\u0000Materials and methods: Mathematical physics methods for reconstructing medical images, namely the Fourier transform in the polar coordinate system and the Radon transform.\u0000\u0000Results: As a result of the work done, an expression for the reverse Radon transform was obtained and the work of the tomograph with point focus of X-ray rays was modeled. Cell-cell and cell-extracellular matrix interactions form a communication network of biochemical and mechanical signals, bring spheroids (SP) closer to native tissues and significantly distinguish them from monolayer cultures. It is important for cell technologies to develop methods for cryopreservation of 3D cultures, that allows creating the stocks of valuable cell samples, save time and materials, and prevent the loss of cultures due to technical failures, contamination, phenotype drift and aging.\u0000\u0000Objectives: Development of approaches to cellular spheroids cryopreservation. Determination of the permeability parameters of L929 cells spheroids at different cultivation periods for the theoretical assessment of optimal freezing regimens.\u0000\u0000Materials and methods: We have used L929 cells, which form SPs of different diameters and can be maintained for a long time in 3D conditions. To determine the integral filtration Lp and permeability for DMSO kp coefficients for SP at different periods of cultivation, the volumetric method was used. The study of the changes in the spheroids volume in time was carried out with a confocal microscope LSM 510 META. The numerical values of the integral SF permeability coefficients were determined by approximating the experimental data on the change in the relative volume of the SP versus the exposure time in the test solution with theoretical curves calculated on the basis of a physical and mathematical model for passive mass transfer between the spheroid and the environment, provided that they coincide as much as possible. Prediction of the osmotic behavior of spheroids under cooling conditions was carried out based on the differential equation describing the kinetics of changes in the relative cell volume during extracellular crystallization of a cryoprotective solution, substituting determined values of integral permeability coefficients Lp and kp and activation energies EAL and EAk into the model equations. The kinetics of changes in the extracellular solution concentration during freezing was set analytically by approximating the phase melting diagram of the DMSO solution.\u0000\u0000Results: The filtration and permeability for DMSO molecules coefficients in SP were determined and their significant decrease with a cultivation duration was shown. The activation energy values for the penetration of water and DMSO molecules into the SP were calculated and their dependence on the cultivation time was determined. Proceeding from the determined parameters of permeability, the dynamic of changes in the volume of SPs for different periods of cultivation at different rates of cooling was calculated.\u0000\u0000Conclusions: The optimal cooling modes of SP from L929 cells were in theory determined: for 7 days of cultivation — 1,5-2 °C/min with cooling to -80°C and subsequent immersion in nitrogen; for 14 and 21 days of cultiva

The use of antimicrobial peptides is one of the possible ways to overcome the threatening rapid growth of resistance of microorganisms to traditional antibiotics. Of the particular role in this context is gramicidin S (GS), which is used for topical medical applications for over 70 years. An acute hemolytic side effect of GS on human cells prevents its systemic use. Understanding the molecular mechanisms of interaction of GS with biological membranes will enhance its bactericidal effect on the one hand, and reduce the negative side effects on human cells and thus expand the range of antibacterial peptides to combat infectious diseases caused by resistant microorganisms.\u0000\u0000Objectives. Study of the effect of different doses of the antimicrobial oligopeptide antibiotic GS on the morphological and electrophysical characteristics of human erythrocytes during in vitro incubation.\u0000\u0000Materials and methods. Morphological changes of erythrocytes of healthy donors after preliminary incubation with GS at concentrations 5–40 μg/ml were studied by flow cytometry using resistance pulse spectroscopy. Single-cell volume, erythrocyte volume distribution in the population was measured, and the electrical breakdown potential of the human erythrocyte plasma membrane was determined.\u0000\u0000Results. Incubation of human erythrocytes with sub-lytic concentrations of GS is accompanied by a redistribution of erythrocytes in this population by volume with an increased number of smaller erythrocytes with less resistance of the membrane to electrical breakdown. However, increasing the concentration of GS to 40 μg/ml leads to an increase in the proportion of cells of increased volume with increased resistance to electrical breakdown of the membrane. Possible mechanisms of morphological changes of erythrocytes under the action of GS are discussed.\u0000\u0000Conclusions. Incubation of erythrocytes with GS at concentrations 5–40 μg/ml is accompanied by a redistribution of cells by volume and changes in the resistance of their plasma membrane to electrical breakdown due to destabilizing membrane-tropic action of the peptide, microsimulation, or cytoskeleton rearrangement.

An apical constriction zone is recommended as a border for root canal treatment and filling. Intraoral radiograph allows you to obtain information about the direction of bending of the root canals, as well as to determine the working length. However, the radiograph is a two-dimensional total image and does not reproduce the entire anatomy of the apical part of the root therefore there are often layers and distortions of the image. When interpreting radiographs, there is a probability of error associated with the subjectivity of the evaluation result of the specialist. Thus, it is impractical to be guided exclusively by this method of determining the working length. The method of apexlocation is based on the difference of electrical resistance of tissues. The hard tissues of the tooth have a higher resistance than the mucous membrane of the mouth and periodontal tissue. Devices for electrometric determination of the working length of the root canal determine the impedance using alternating currents of different frequencies and apply the method of ratio. This measurement is stable and accurate even when working in wet channels and provides smooth visualization of all process of penetration of a top of the channel tool and high accuracy of definition of a place of physiological top of a root (over 80%). Modern algorithms for electrometric determination of the working length of the root canal do not combine the data obtained from the radiograph. In this regard, it is important to develop new methods and means of displaying electrometric data on the radiograph to more accurately determine the location of the physiological apex of the root.\u0000\u0000Objectives: Development of a method of segmentation of the dental radiograph to determine the area of apical narrowing.\u0000\u0000Materials and methods: principles of endodontic tooth preparation; methods for determining the working length of the root canal (radiological, electrometric); threshold segmentation method; method of segmentation of bone structures on tomographic images.\u0000\u0000Results: As a result of the performed work, the structures of the root canals of the tooth were segmented and their length was determined. Comparison of electronic determination of working length with radiological led to the fact that in some cases, X-ray and electronic length do not match. With lateral curvature of the canal, the X-ray may show a shorter working length than apexlocation devices, so the electronic working length is usually more accurate than the length determined by X-ray.\u0000\u0000Conclusions: The advantage of measuring the length of the root canal with an apex locator is much greater accuracy (about 0.5 mm) compared to the method of radiography, but the combination of these two methods is more reliable, which requires f

Due to the lack of an adequate substitute for natural bone, combined approaches are used. For older patients, the problem is exacerbated by a decrease in bone mineral density. When choosing a scheme for the surgical treatment of long bone fractures, preference is given to simple and maximally sparing approaches. In this regard, the main task of osteosynthesis is to provide optimal mechanical channels not only for the fracture healing process, but also to restore full functional capabilities in the future. The paper considers the urgent task of optimizing and increasing the efficiency of planning rehabilitation measures, including taking into account the individual characteristics of a particular patient, and the results are of fundamental and applied importance.\u0000\u0000Objectives: development of physical and mathematical models for modeling the stress-strain state of the elements of the musculoskeletal system to optimize the planning of bone surgeries when installing implants.\u0000\u0000Materials and methods. For the analysis, both specific clinical results and modern methods of computer modeling and processing of results were used. The advantage of physical and mathematical models based on the used finite element method is the possibility of optimizing the design of prostheses and reducing the problems caused by osteopenia.\u0000\u0000Results: To illustrate the proposed approach, a specific example of the treatment of a comminuted fracture of the humerus in an elderly patient is considered. To describe the physicomechanical properties of bone tissue, sets of standard data on the main characteristics of tissues and materials of implants such as elastic modulus and Poisson's ratio were used. As the bone grows together, simultaneously with a decrease in stresses, the difference between the stress on the entire structure and the stress on the bone decreases. This indicates that the bone begins to take on an increasingly significant relative part of the load, which should have a positive effect on its bone mineral density.\u0000\u0000Conclusions: The advantages of modeling using the finite element method and by non-invasive modeling of the work of the patient's musculoskeletal system with various variants of prostheses (implants) and the choice of the most optimal one are shown. A variety of techniques have been developed to characterize the drug-nucleic acid binding, among which the fluorescence dye displacement assay is one of the most informative approaches. Recently, it was demonstrated that cyanine dyes can be successfully employed for the high throughput screening of the interactions between nucleic acids and drugs. To the best of our knowledge, so far, the potential application of cyanine dyes for the drug-displacement studies remains insufficiently evaluated.\u0000\u0000Objectives: The aim of the present study was to investigate the ability of a novel cyanine dye to serve as a competitor for the potential antitumor compounds, lanthanide complexes bearing europium (III) tris-β-diketonate (EC) for the DNA and RNA binding sites.\u0000\u0000Materials and methods: Calf thymus DNA, yeast RNA, trimethine cyanine dye and lanthanide complexes bearing europium (III) tris-β-diketonate were used for sample preparation. Two mechanisms responsible for an appearence of the red luminescence of Mn are proposed. They are the intra-center luminescence and recombination luminescence mechanisms. In this case the red Mn luminiscence prevails and the blue luminescence characterizing the host material has not been observed.\u0000\u0000Conclusion: AlN nanopowder doped with Mn atoms is a prospective material for use as luminescent marker for imaging of biological materials. Properties of this material are in a good agreement with the main requirements obligated to biological materials: i) AlN NP has low toxicity; ii) AlN:Mn NP possesses intensive red luminescence at 600 nm, which can be excited either with the ultraviolet light around 260 nm or with visible light around 480 nm; iii) it is relatively cheep material and it can be synthesized using simple synthesis methods.
