Unfortunately, the exploitation of palm oil constitutes a source of environmental pollution due to the production of enormous quantities of waste during its extraction process, including solid sludge generating greenhouse gases which contribute to global warming climatic. All this leads to the search for alternatives which consists of isolating and characterizing indigenous bacteria with biodegradation capacities in sludge from palm oil extraction. The pH and bacterial counts were determined by the potentiometric method and the decimal dilution technique, respectively. The isolated bacteria were identified by their cultural, cellular and biochemical characteristics. In addition, the identification of Gram- bacteria was further explored by the API 20 E gallery. The palm oil biodegradability test was carried out on M2 medium supplemented with 2% palm oil. The solid sludge biodegradability test was carried out on liquid MSM medium supplemented with 4% sludge stock solution. The results showed that the sludge sample had a slightly alkaline pH of 7.3. A bacterial load of around 10<sup>9</sup> CFU/g of soil was counted. Thirty-one bacterial strains were isolated and purified, including 12 <i>Bacillus sp</i>, 10 <i>Pseudomonas sp</i>, 8 <i>Proteus mirabilis</i> and 1 <i>Klebsiella pneumoniae</i>. All isolates tested for their ability to degrade palm oil or solid sludge grew in culture media with palm oil or solid sludge as the sole source of carbon and energy but with a difference in load. Thus, isolates BI2, BI5, BI31, BI10 and BI 9 showed the highest degradation capacities. Industrial effluents and oil spills are continuously contaminating the soil. Further, seasonality influences the distribution of pollutants in soil. Consequently, soil microbial biota and ecological processes are affected. This study assessed the effect of seasonality on soil fungal and bacterial communities in oil contaminated soils in 12 selected sites in Greater Port Harcourt Area. Standard analytical procedures were used to obtain bio-physicochemical data from the soil samples and t test was used to analyse data. The levels of total petroleum hydrocarbons (TPH) were above 5000 ppm (DPR recommended limit). There was significant difference (p ≤ 0.000) between the means of TPH in wet and dry seasons. Seasonality influenced % HUF and % HUB in the soils of urban, industrial and agricultural sites. Generally, the seasonality trends showed that there was a declining population of THB, HUB, TF, and HUF from the wet season to the dry season. However, the results show that there was a stable trend in % HUB as compared to oscillations observed in % HUF in oil tainted soils across a seasonal divide. The applications of these biomolecules cut across different industries such as detergent, brewing and food industries. Aspergillus flavus has been well documented as one the microbes capable of degrading starch-based agricultural waste materials. This study aimed at optimizing and characterizing amylase produced using agro-waste (potato peels) as a sole carbon source. A. flavus was obtained from the Department of Microbiology laboratory, Adekunle Ajasin University, Akungba-Akoko, Ondo State, Nigeria. The fungus was preserved on PDA and stored at 4°C throughout the study period. This was followed by characterization and purification. Preliminary screening result showed that A. flavus had a clear zone of 13.33 mm (diameter of hydrolytic zone) and amylase activity of 545 U/ml. Stability assay revealed that the enzyme was stable at 50°C and 60°C by retaining 91% and 84% residual activities respectively after 4 h of pre-incubation period, while residual activities of 80% and 90% were observed at pH 6.0 and 8.0 respectively. Protein band analysis showed that the amylase from A . flavus had molecular weight of 35 kDa. These results indicated that the amylase produced from A. flavus had good catalytic properties and compared well with those from other known fungi. Although there are many reports about soil microbiology, overall it is still limited.
