Leaf, stem, cotyledon and hypocotyledon explants cultured on different concentration of auxins and cytokinins, were supplemented on MS medium to test their ability to induce rhizogenesis. Murashige and Skoog (MS) medium fortified with 2,4-dichlorophenoxyacetic (2,4-D), indoleacetic acid (IAA), Indole-Butyric Acid (IBA) and Naphthalene Acetic Acid (NAA). Each at the concentrations of 0, 0.5, 1.0, 1.5, 2.0 and 2.5 mg LG were evaluated for their effects on adventitious root induction in the present study. High concentration of IAA (2.0 mg LG) and low concentration of IBA (1.5 mg LG) is best for induction of rhizogenesis. High concentration of IBA favored formation of malformed and thick roots. The NAA in combination with IBA is most potential in inducing high percentage of rooting (50%) and IAA in combination with IBA induced maximum rooting 62% with highest number of roots per shoot (3.80±0.2) and root length (3.8±0.1 cm) were obtained from leaf, cotyledon and stem than hypocotyls. Results revealed that, the best rooting ability was observed from in stem explants cultured on MS medium supplemented with 2.0 mg LG IAA and 1.5 mg LG IBA, multiple roots fibrous were produced from cotyledon explants on MS fortified with 2.0 mg LG 2,4-d and multiple roots cluster produced from leaf explants 1.5 mg LG IBA and 2.0 mg LG IAA. It was shown that, the maximum growth yield opacity was found to be 70 IOU mLG at 48 h cultivation in single impeller fixed at the bottom position of fermentor vessel. Whereas the optimal and moderate culture opacity 60 and 50 IOU mLG was observed during the two impellers mounted at two different positions on the shaft. Similarly the least growth yield was obtained during three impeller located at top, middle and bottom region of the shaft and the final opacity was found to be 30 IOU mLG at the end of 48 h cultivation with an increasing of culture pH 8.02. However, in two impeller combination experimental study also showed moderate growth yield at 48 h could give to homogenized mixing and good aeration. Thus, the two different location of the impeller combination influences two vortex flows, enhances optimal growth rate, viz; the lower impeller act as gas disperser and creates radial velocity with higher rates of air dispersion, whereas the upper impeller pumps the broth towards down thus the combined agitation of the broth leads to good mixing and even transfer of Dissolved Oxygen (DO2) to the growing cells and found moderate bacterial growth. In the present study, we aimed to detect the genetic variations in species of genus Schistura from Uttarakhand. Very little is known for the pattern of distribution and genetic diversity of the species of genus Schistura from the Garhwal and Kumaun region. Out of six primer only four primers were used to generate the fragments patterns from the samples collected. Polymorphism within and between the populations were assayed and total 60 bands were amplified ranging from 1-150 kb. Maximum number of bands was observed in Schistura montanus from the Srinagar, Garhwal and Uttarkashi regions. Total 60 bands were amplified in the all four primer with high percentage of polymorphic loci 50% Schistura montanus from Garhwal region and 28.8% in the S. montanus from the Kumaun region were observed. The level of heterozygosity were found 0.006-0.18 in the all three species. The percentage of polymorphism was 50% within the populations and high heterozygosity the sample of S. montanus when compared with other species. Callus obtained by culturing young leaf disks on Murashige and Skoog's (MS) medium with 1.0 mg L-1 2, 4.D (dichlorophenoxyacetic acid) was subcultured on MS medium contg. 2.0 mg L-1 6, Benzyl Amino Purine (BAP), produced 30-40 multiple shoots. Rooting was achieved on half or full strength MS medium with 0.5-0.1 mg L-1 Naphthalene Acetic Acid (NAA) in 14 days. Regenerated plantlets were acclimatized and transferred to soil were showed normal morphol. characteristics. Survival of transplants was 90-95% under green house condition.
