The most potent marine bacterium was identified as Bacillus licheniformis SMRH1 , accession number ON777836 by using 16s rRNA as a molecular identification technique. According to the chemical analysis, the obtained polysaccharide content is estimated by using Fourier Transform Infrared spectra FTIR spectrum of the extracted exopolysaccharide from Bacillus licheniformis which showed that it contains νOH at 3420cm -1 , νC-H aliphatic at 2952cm -1 , νC=O in range 1660-1630cm -1 and Nuclear magnetic resonance spectra NMR showed signals δ’ S ppm at 1.22 (s, -CH 3 ), 2.07 (s, -CH 2 chain ) and 2.35(m, -CH 2 ) and Scanning electron microscope SEM which confirmed the amorphus shaped uneven texture polysaccharide at higher magnification of ×5000. The exopolysaccharide that was generated has antibacterial activity by forming a free hollow zone around it against some gram positive and gram-negative bacteria according to disc diffusion method. The innovatively produced exopolysaccharide also has anticoagulant activity. A recent study proved that the generated exopolysaccharide induced β-hemolysis. The high yield of exopolysaccharide was “0.7 g/l” by using different culture media such as nutrient broth, peptone water, and king ̓s B broth.","PeriodicalId":233013,"journal":{"name":"Research Journal of Applied Biotechnology","volume":"190 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-07-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"131581962","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In this prospective cohort observational study, extracellular miRNAs were tested using the miRCURY Locked Nucleic Acid (LNA) system, which used culture media from twenty-two singly transferred embryos. Study samples were collected on day three and embryos were transferred on day five. Based on earlier publications, hsa-miR-29c-3p and hsa-miR-30c-5p, among other miRNAs (not published), were chosen and assessed. The negative-implantation group had higher expression of hsa-miR-30c-5p regarded as statistically significant (P.value <0.001) than the other tested miRNAs. Contrary, there was no significant link between pregnancy outcome and expressed hsa-miR-29c-3p. Study findings, in comparison with findings from other reports, reveal the absence of hsa-miR-29c-3p and hsa-miR30c-5p from day 3 implanting embryos in contrast with day 5 where they are abundant, suggesting specificity to the embryo’s development from cleavage to morula, blastocyst, and implantation stages. These findings could open the gate toward potential biomarkers for embryo development and implantation outcome.","PeriodicalId":233013,"journal":{"name":"Research Journal of Applied Biotechnology","volume":"198 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2022-12-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"121628538","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Stem cells secreted factors are safe and efficient alternative to stem cells in stem cells-based therapies. In this study, this study aimed to evaluate the antibacterial activity of factors secreted by human Wharton’s jelly mesenchymal stem cells secretome (hWJ-MSCs) against Staphylococcus aureus, hWJ-MSCs were cultured and collected their secreted factors to be used for In vitro antibacterial screening against Staphylococcus aureus . Antibacterial activity, antibiofilm, and minimum inhibitory concentrations (MIC) of hWJ-MSC-S against Staphylococcus aureus were determined. The present data showed that hWJ-MSC secreted factors significantly inhibit the growth of two clinical isolate of Staphylococcus aureus. hWJ-MSC-secreted factors reduced the growth of Staphylococcus aureus by mor than 87% compared to un-treated control. MIC values were 4.68 µg/ml and 1.17 µg/ml for Staphylococcus aureus 76, and Staphylococcus aureus 105, respectively. The finding from this work could be utilized to develop an effective therapeutic approach to treat Staphylococcus aureus infection .","PeriodicalId":233013,"journal":{"name":"Research Journal of Applied Biotechnology","volume":"23 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-12-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"121375089","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Pylori) begins its long-term infection in the stomach, where the immune system responds with local neutrophil, lymphocyte, and macrophage infiltration. H pylori infection is known to cause stomach ulcers, but scientists are still largely in the dark about this critical area of health. We transfected HeLa cells with specific siRNA antagonist bacterial CagA gene or siRNA antagonist autophagy-related Atg12, and then H. Pylori reproduction was monitored. We also looked at proinflammatory cytokine secretion from treated cells to see if there was a link between bacterial CagA, host autophagy, and inflammatory events like tumor necrosis factor-alpha (TNF-) and interleukin-6 (IL6). When compared to control-transfected cells, our findings revealed previously unnoticed changes in cell morphology, number of survived cells, and lactate dehydrogenase (LDH) production after transfection with siRNA targeting CagA or Atg12. Furthermore, targeting CagA effector and Atg12 successfully disrupts H. pylori reproduction in infected cells, as measured by quantitative real-time PCR of bacterial 16S ribosomal RNA (16S rRNA) (q-RT-PCR). Furthermore, CagA downregulation causes an obvious decrease in Raf-1 gene expression, which is an indicator of Megaton-Activated Protein Kinase (MAPK) signaling in infected cells. Finally, IL-6 and TNFproduction was mediated in siRNAtransfected cells that attacked either CagA or Atg12 effectors. In conclusion, these findings suggest that","PeriodicalId":233013,"journal":{"name":"Research Journal of Applied Biotechnology","volume":"39 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-11-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"134282521","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
