To date, the effectiveness of osteopathy in the treatment of patients with tension headaches has been shown. It remains an open question to develop a generally accepted method of diagnosis and treatment, including the use of complementary therapies, neuroimaging objectification and determination of the state of functional connections of the brain for more accurate diagnosis and prediction of treatment effectiveness. The present review summarizes the current ideas about the etiopathogenesis of tension headache (including the study of the functional connections of the brain) and available data on the clinical efficacy of osteopathic correction.","PeriodicalId":23243,"journal":{"name":"Translational Medicine","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-06-13","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"141204276","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Periodontal stem cells are a promising material for dentistry and maxillofacial surgery. In this work, we sought to find out how Notch signaling pathway affects the osteogenic potential of periodontal ligament stem cells (PDLSC). Design and methods . In order to verify this, we used lentiviral transduction of PDLSC, adding various amounts of the virus bearing the activated intracellular domain of the Notch1 receptor, NICD. Using real-time PCR, we analyzed the dependence of changes in the expression levels of osteo-markers ( RUNX2, COL1A1, OGN, POSTN ) on the activation strength of Notch signaling pathway, based on the expression of the target gene — HEY1 , in the early stages of differentiation, after 48 and 120 hours after induction of osteogenic differentiation. In addition, using the technique of Alizarin red extraction, we were able to quantify the intensity of mineralization of periodontal MSCs at the final stage of osteogenic differentiation. Results . We found that activation of Notch signaling pathway leads to increased expression of osteogenic markers in the early stages of osteogenic differentiation induction, which in turn leads to the development of the final stage of osteogenic differentiation, characterized by the formation of calcification. Moreover, the more intensive the initial level of Notch, the stronger and more effective were the processes of osteogenesis. Conclusion . In this work, we showed that activation of Notch signaling pathway leads to an increase in the osteogenic potential of periodontal PDLSC in a dose-dependent manner and induces in the cells processes associated with the accumulation of calcifications, the intensity of formation of which directly depends on the “strength” of Notch signal. The results obtained in this work convince us that Notch is able to modulate osteogenic differentiation, affecting its effectiveness, apparently due to the strength of the signal that is transmitted to the cells.","PeriodicalId":23243,"journal":{"name":"Translational Medicine","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-05-19","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"141204003","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In the review the main attention is payed to the role of signaling pathways BMP (bone morphogenic protein), Notch, Wnt and to the role of transcription factors BMP2, RUNX2, Msx2 in vascular calcification. Probably, dysfunction of osteogenic signal pathways and transdifferentiation of vascular cells to osteoblast-like cells is a common prosses not only for vascular calcification or mineralization, but is a way of vascular degradation in general. Proosteogenic changes at cellular and molecular level may play role in pathogenesis of a disease without manifestation of vascular mineralization, such as thoracic aortic aneurysm. Methods of open bypass surgery for chronic atherosclerosis of the arteries have long been developed, put into practice and can be considered individually for each patient. However, given the frequent complicated atherosclerotic history in patients with multifocal lesions of various arterial pools, most patients can be recommended more gentle endovascular methods of blood flow correction — balloon angioplasty or stenting. But given certain technical and general parameters (prolonged occlusion, severe calcification, kidney disease, an allergic reaction to a contrast drug, etc.), it is not yet possible to use intravascular correction in absolutely all patients, as a result of which the method of simultaneous or phased hybrid surgery seems to be very relevant and can be used even in patients at extremely high risk. The review describes the possible uses of hybrid technology depending on the level of steno-occlusive lesion and the general prolongation of narrowing of the arterial bed.
