Chou","doi":"10.1142/S1682648520500146","DOIUrl":"https://doi.org/10.1142/S1682648520500146","url":null,"abstract":"The aim of this study is to investigate whether combinational use of efflux pump inhibitors (EPIs) could improve florfenicol (FF) antimicrobial activity. Five EPIs including Carbonyl Cyanide Chlorophenylhydrazone (CCCP), omeprazole, Phenylalanine-arginine [Formula: see text]-naphthylamide (PA[Formula: see text]N), reserpine and verapamil were individually combined with FF and the minimum inhibitory concentration (MIC) against porcine Actinobacillus pleuropneumoniae and Pasteurella multocida were evaluated by the broth microdilution assay. The results indicated that CCCP demonstrated substantial improvement on the antimicrobial activity (FF MIC reduction [Formula: see text] folds) while PAßN showed minimal effect at high concentrations (80–120[Formula: see text][Formula: see text]g/mL). The MICs of FF were further examined with CCCP at 5[Formula: see text][Formula: see text]g/mL against resistant A. pleuropneumoniae and at 2[Formula: see text][Formula: see text]g/mL against resistant P. multocida. With this combination, a total of 75% (6/8) of A. pleuropneumoniae and 100% (8/8) of P. multocida resistant isolates showed significantly reduced ([Formula: see text] folds) FF MICs. In addition, 100% of the 16 resistant bacterial strains carried the floR gene that regulates the phenicol-specific efflux pump (FloR pump). The time kill experiments were used to verify the effect of CCCP demonstrating a bactericidal effect in resistant A. pleuropneumoniae and synergistic effect in resistant P. multocida. In contrast, the FF MICs were not significantly affected in the FF susceptible strains of these two bacteria. These findings suggested that despite small sample sizes, consistent beneficial effects of CCCP were evident such that the combinational use of selective EPI with FF might be an alternative antibacterial strategy against FF resistant A. pleuropneumoniae and P. multocida. Nevertheless, traditional diagnostic methods may not efficiently detect and accurately demonstrate the endoparasite infestation status. In this study, we developed a multiplex PCR assay targeting the rRNA genes to simultaneously detect and differentiate five endoparasites, including Syphacia obvelata, Syphacia muris, Aspiculuris tetraptera, Spironucleus muris, and T. muris, as well as a housekeeping gene in feces. The multiplex PCR could identify an equivalent infection of pinworm, Spironucleus muris and T. muris, with a detection limit of as few as 10 copies. Furthermore, dual infections with up to 100-fold differences and triple infections with 10-fold differences in parasite loads can also be detected. In comparison of traditional methods with the multiplex PCR assay, 76 rodents from 11 research colonies and 3 pet shops and additional 27 fecal samples from laboratory rodents were screened for the infestation status of the five endoparasites. The multiplex PCR had higher sensitivity (97.2–100%) and accuracy (99–100%) than those of the traditional antemortem (sensitivity: 83–100%; accuracy: 94–100%) and postmortem methods (sensitivity: 75–100%; accuracy: 92.1–100%). In addition, an early stage of S. obvelata contamination in a SPF laboratory animal colony was also successfully detected by this multiplex PCR assay. Myocoptes musculinus (COP), Myobia musculi (MOB) and Radfordia affinis (RDA) have been reported in laboratory mouse colonies. To improve the sensitivity and efficiency of fur mite detection, COP-specific PCR and MOB/RDA-specific PCR assays were developed to detect and differentiate these fur mites, with the existence of a rodent housekeeping gene. The COP-specific PCR and the MOB/RDA-specific PCR could specifically detect COP and MOB/RDA at as low as 10 copies, respectively. In comparison of the specific PCRs with traditional methods (pluck test, tape test and pelt exam) for fur mite diagnosis, 31 rodents and 17 cage environment samples were evaluated. In screening for the infestation status of various fur mites on individual animals, the specific PCR assays showed distinctly higher sensitivity and accuracy (100% and 100%) than those of the traditional methods (sensitivity: 25–80%, accuracy: 83.9–96.8%). Interestingly, by using cage wipe environmental samples, the specific PCR assays exhibited 100% sensitivity and accuracy in the fur mite detection and differentiation. The COP-specific and MOB/RDA-specific PCR assays developed in this study could be reliable alternatives for routine pathogen monitoring of laboratory mice and environments of animal facilities without animal sacrifice. In this study, two new parvoviruses were identified in naturally-infected rats from two different research colonies in Taiwan. The genomic nucleotide sequences and the predicted amino acid sequences of proteins for these two viruses were compared to the previously characterized rodent parvoviruses. The two newly identified viruses were most closely related to each other, also closely related to two variants of rat minute virus (RMV; RMV-1 and RMV-2), and distinct from but closely related to Kilham rat virus and H-1 virus. These two viruses were significantly different from variants of rat parvovirus (RPV; RPV-1 and RPV-NTU1). Phylogenetic data also supported that these two new viruses are variants of the RMV species. These two newly identified viruses were designated RMV type National Taiwan University 1 (RMV-NTU1) and RMV type National Taiwan University 2 (RMV-NTU2). However, the in ovo vaccination for disease control was not applied in practice because this vaccine strain is highly pathogenic to chicken embryos (CEs) causing early death, dwarfing and other harmful effects. We compared the differences in virus replication, pathological changes, and tissue tropism between the wild virus and attenuated vaccine strain in CEs inoculated with different viral titer levels, i.e. 0.1, 1 and 10 EID[Formula: see text]/egg. The wild virus caused dwarfing effect at high titer inoculation, whereas the attenuated vaccine strain caused the dwarfing effect only at a lower viral inoculation accompanied by the earlier infection establishment and embryonic death at high and medium titers. There were no significant differences in the pathological changes in CEs infected by both wild and attenuated strains. Detected by immunohistochemistry, the viral antigens of both strains could be found mainly at the epithelium of the chorioallantoic membrane, lung parabronchus, renal tubules and some in the spleen and heart serosa. These findings indicated that the early embryonic death and dwarfing is not related to the change in cell/tissue tropism of the vaccine strain, rather on the early infection establishment and viral load.
