Kovpak","doi":"10.15421/NVLVET6626","DOIUrl":"https://doi.org/10.15421/NVLVET6626","url":null,"abstract":"Bone marrow is the only adult tissue which normally consists of immature undifferentiated and low differentiated cells which called stem cells and they are similar in structure to embryonic stem cells. But literature data analysis doesn't give an unambiguous answer regarding phenotypic and morphological changes of bone marrow cells culture of rats during their in vitro cultivation which necessitated further research. Investigate phenotypic and morphological changes of bone marrow cells culture of rats during their in vitro cultivation from first to fourth passage. We were used in these research bone marrow cells of rats from the first to the fourth passages. Microscopic analysis and evaluation morphological changes of bone marrow cells culture of rats during cultivation were carried out using inverted microscope Axiovert 40. Control of changes phenotype was performed by detecting CD markers (CD10, CD38, CD34, CD45, CD48, CD54, CD56, CD66e, CD96, CD227, CD326, pan–keratin). The evaluation was performed by the semi– quantitative method (H–Score). The research of primary culture of rat bone marrow cells showed that it morphologically heterogeneous, noted the small number of cells polygonal shape, surrounded by the fibroblast cells. During the cultivation cell culture becomes more homogenous at the expense of fibroblast–like cells. As a result of occurred the transition process from heterogeneous culture in zero passage to the most homogeneous culture in 4 passage. Immunophenotyping population of cell culture derived from rat bone marrow, revealed a high level of expression of pan–keratin; moderate level – CD34, CD48, CD66e, CD95; low level – CD38, CD45, CD56, CD227, CD326; lack of expression – CD10, CD54. Change of the expression of surface markers varies in each passage CD48, CD66e, CD95 increased significantly; CD38, SD45, SD326, pan–keratin reduced significantly. The markers CD34, CD 56, CD 227 were expressed on the one level from the first to the fourth passage. Первинні посіви зроблені із серця, печінки, нирок, селезінки та легень на загальновживані (МПБ і МПА) та селективні поживні середовища (агар Ендо, ксилозо-лізиновий дезоксіхолатний агар (XLD Agar), агар PALCAM, сольові бульйони із різною концентрацією NaCl (2%, 5% та 10%)). У результаті проведених бактеріологічних досліджень з організму черепахи червоновухої було виділено чотири ізоляти мікроорганізмів: два ізоляти із роду Staphylococcus, по одному із родів Klebsiella та Yersinia. Нами було встановлено, що штами Klebsiella pneumoniae є високочутливі до іміпенему, меропенему, чутливі до амікацину, нетилміцину. Штами Yersinia enterocolitica, є чутливі до амоксициліну, ріфампіцину, пеніциліну та цефтриаксону. Staphylococcus epidermidis (№ 1, 2, 4, 5) штами були високочутливі до ванкоміцину та ріфампіцину, чутливі до гатіфлоксацину, лінезоліду. Дані штами отримали позначку РА-11/15. Staphylococcus epidermidis (№ 3) мав чутливість до метициліну. Дана різниця, на наш погляд, є принциповою, тому даний штам було виокремлено, та він отримав позначку Poltava-15. Material was obtained during a routine surgery for the removal of testicular tumors in dogs in clinics of veterinary medicine in Kiev. Tumors were divided into sex-cord stromal tumors and germ cell tumors. Among the sex-cord stromal tumors recorded leydig (interstitial) and sertoli cell tumors. Macroscopical and histologic characteristics of canine interstitial (Leydig) cell tumor are described. It is 12.5% of all testicular tumors. The average age of dogs is 9 years. Largest share is breeds dog such as foxterriers and toyterriers. Interstitial (Leydig) cell tumor has a slower growth compared to other tumors the testicle, no metastases. Half the animals had no clinical signs. Mostinterstitial (Leydig) cell tumor go together with the pathology of adnexal structures. This is because Leydig cell tumor lead to increased levels of androgens. This fact may cause pathology of adnexal structures. Neoplasms are recorded both on the right and the left testis. Leydig cell tumor was not related to the cryptorchid testicles. Most tumors regular oval, brown or gray color, different texture and size with no signs of necrosis or ulceration. The average volume of tumors ranged from 2 to 20 cm2. Microscopically, the tumor is composed of cells that resemble Leydig cells and that are arranged into islands or tubular structures. Find polygonal eosinophilic cells with granular or vacuolated, which contains lipids. Nuclei were round mesh of chromatin and distinct nucleolus. Sometimes noted the large cells with 1, 2 or more nucleoli. In some cases, found elongated fusiform cells with small nuclei and granular eosinophilic cytoplasm. Tumor stroma characterized by calcification. Note mitotically active cells. Cysts lined by tumor cells are present in some tumors. Distinction between benign and malignant forms on purely histological criteria, in the absence of metastasis, often is difficult. Borysevich","doi":"10.15421/NVLVET7026","DOIUrl":"https://doi.org/10.15421/NVLVET7026","url":null,"abstract":"Вasic macro– and microscopic changes in the liver of dogs in case of experimental sarcocystosis were determined as a result of our studies. Three clinically healthy puppy aged 2 – 4 months who were subjected to infection by feeding within three days of minced beef hearts, affected by sarcocysts, were selected for the experiment. The animals were subjected to euthanasia at the 7th, 14th and 21st day after infection. Autopsy was performed by partial evisceration in the conventional sequence. To detect microscopic structure of the liver, slices of 7– 10 µm thickness, obtained by the sliding microtome, were stained by hematoxylin and eosin Karazi. For detection of the lipids, frozen sections of thickness 15 – 20 µm, stained by Sudan III, were produced by using freezing microtome. Morphometry was performed due to G. Avtandylov. As a result of the research it was established that liver of all dogs macroscopically had not changed in size, its edges were sometimes sharpened, areas of different sizes and shapes of bluish and gray colors were seen. Macroscopic changes in the liver of dogs in case of experimental sarcocystosis were uncharacteristic. Conducting histology research, venous stasis and total hard granular dystrophy of hepatocytes in liver were determined. {"title":"ЗМІНИ ГЕМАТОЛОГІЧНИХ ПОКАЗНИКІВ У КОТІВ ЗА НОТОЕДРОЗУ ТА ВНАСЛІДОК ЛІКУВАННЯ РІЗНИМИ СПОСОБАМИ","authors":"T. Bakhur, S. Poberezhets","doi":"10.15421/NVLVET6601","DOIUrl":"https://doi.org/10.15421/NVLVET6601","url":null,"abstract":"Подані результати гематологічного дослідження котів, хворих на нотоедроз, та на 8 і 20 добу лікування за різними схемами. Встановлено, що при паразитуванні Notoedres cati у котів не лише порушується структура шкіри в місцях ураження, а й ушкоджується організму в цілому. Під впливом метаболітів кліщів та продуктів запалення відбувається сенсибілізація та порушення роботи печінки, а за рахунок болючості щелеп – розлад надходження поживних речовин. Ці процеси відображаються у достовірних змінах гематологічних показників хворих котів порівняно зі здоровими: еритроцитопенії (на 16,2%), лейкоцитозі (у 2 раза), еозинофілії (у 5,9 раза), появі базофілів (4,20 ± 0,16%) та юних нейрофілів (5,10 ± 0,28%); зниженні концентрації гемоглобіну (на 19,1%), загального білка (на 18,9%), альбумінів (на 27,1%), Кальцію (на 14,3%); підвищенні вмісту загального білірубіну (на 91,2%), холестерину (на 75,5%), креатиніну (на 39,6%); зростанні активності ферментів АлАТ (у 2,9 раза), АсАТ (у 3,2 раза) і ЛФ (на 34,6%).Застосування котам за нотоедрозу препарату дектомакс у комплексі з екстрактом личинок воскової молі 25% перорально та сірково-дегтярного лініменту з додаванням личинок воскової молі зовнішньо, дозволяє зменшити інтоксикацію організму, прискоривши одужання. {"title":"ВПЛИВ МІКРОЕЛЕМЕНТІВ ТА ГУМІЛІДУ НА ВІДТВОРНУ ФУНКЦІЮ ТЕЛИЦЬ УКРАЇНСЬКОЇ М’ЯСНОЇ ПОРОДИ","authors":"V. Gryban, D. Mylostiva, E. Pecheniy","doi":"10.15421/NVLVET6610","DOIUrl":"https://doi.org/10.15421/NVLVET6610","url":null,"abstract":"The article presents research data on the impact of deficient dietary microelements (cobalt and selenium) and biologically active supplement Humilid on the indicators of the reproductive function of heifers after the first calving. We have found out that Humilid and trace elements influence on the physiological state of reproductive organs and stimulate the sexual heat of animals. Regarding the duration of the period from calving to fertilized insemination, the difference between the control and experimental groups is more visible. Thus, the service period in the experimental group I (under the influence of Humilid) was 293.2 days, which is 2.6 days less than in the control group. In the experimental group II (under the influence of cobalt and selenium), it was, respectively, by 3.9 days less than in the control one. This may indicate that the minerals and dietary supplements contribute to more rapid recovery of the ovarian cycle. Protein composition of blood serum was investigated in the study of the biochemical spectrum of blood. Balancing the nutrition of animals with deficient dietary microelements and adding Humilid increase the concentration of protein molecules, which is visible from the indicator of crude protein and its components - albumen and globulin. In comparison with animals in the control group, a total protein level of heifers in the experimental group I was higher by 8.8% (P <0.05), and in the experimental group II - by 8.6% (P <0.05). This may indicate the increased intensity of synthetic and redox processes in heifers’ bodies in preovulatory period which serve best for the insemination. The level of albumen was also higher by 8.0 and 7.3% respectively under the influence of Humilid and trace elements in relation to the control group, indicating the improvement of transport of substances, in our case estradiol. As far as globulin is concerned, the growth of this class of proteins was marked by 9.4 and 9.6%, indicating the improvement of the level of resistance of the organism.
